Djurceller interagera fysiskt med matrisen och de andra cellerna som utgör deras miljö1. Detta krävs att migrera, internalisera organ, förvärva extern information eller skilja. I sådana processer, cellen måste generera mekaniska krafter och, som ett flertal studier har visat de senaste åren, förmågan hos en cell att generera styrkor och sond dess miljö påverkar dess biologiska beteende, till exempel styra spridning eller differentiering2,3. Mätningen av cellulära krafter är i sin tur ett större stöd för att studera regleringen av styrkebidrag och förstå dess implikation i cell beteende och vävnad öde4,5.
Senaste åren har bevittnat utvecklingen av många tekniker för att mäta de krafter som en cell kan utöva på dess miljö6. Majoriteten av dessa har varit avgörande i avslöjar dragkraften tvingar att celler utöva eftersom de dra på mobila sonder eller en deformerbar substrat. Men de mekaniska krafterna som är inblandade i utstick i extracellulära miljön lider brist på mätteknik och är hittills inte väl karakteriserade.
För att övervinna denna begränsning, presenterar vi en metod att mäta krafter som utövas ortogonalt till substratet. Den består i plätering levande celler på en tunn elastisk blad som kan deformera i ortogonal riktning, vilket gör det möjligt att mäta substrat deformation av celler och härleda krafterna som är inblandade. Substratet topografi mäts med nanoskala upplösning med atomic force microscopy och utvärderingen av styrkor från deformation bygger på kunskap om geometri av den protrusive cellulära strukturer7,8, 9.
Här, beskriver vi installationen och dess tillämpning på mäta de krafter som genereras av podosomes, protrusive vidhäftning strukturer bildas av makrofager för deras mesenkymala migrering i tredimensionella miljöer10,11, 12,13,14,15,16,17. Vi tror att denna teknik kommer förväg förståelsen av styrkebidrag och dess reglering i många cellulära processer som involverar utstick.

protrusion force microscopy: a method to quantify forces developed by cell protrusions

I många biologiska sammanhang behöver djurs celler fysiskt interagerar med sin omgivning genom att utveckla mekaniska krafter. Bland dessa dragkraft krafter har varit välkarakteriserad, men det finns en brist på tekniker som möjliggör mätning av de utskjutande delen krafter som utövas av celler ortogonalt till deras substrat. Vi utformade ett experiment att mäta de utstick krafter som utövas av vidhäftande celler på deras substrat. Celler pläterade på en kompatibel Formvar ark deformeras detta substrat och resulterande topografin är mappad av atomic force microscopy (AFM) i nanometer skala. Kraft värden extraheras sedan från en analys av deformation profilen baserat på geometri av de protrusive cellulära strukturerna. Därför kan de krafter som utövas av enskilda utskjutande enheter av en levande cell mätas över tiden. Denna teknik kommer att möjliggöra studiet av styrkebidrag och dess reglering i många cellulära processer som involverar utstick. Här, beskriver vi sin ansökan att mäta de protrusive styrkorna som genereras av podosomes bildas av mänskliga makrofager.
