Hier, we laten zien dat Doxorubicine injectie via retrobulbaire sinus injectie experimentele nefrotisch syndroom bij 129S1/SvImJ muizen met Proteïnurie, retentie van natrium, hypoalbumenia en hyperlipidemie induceert. Er zijn echter twee belangrijke onderwerpen die rekening worden gehouden moet bij het gebruik van dit model. In de eerste plaats de model-inductie is strikt dosis afhankelijk en afwijkingen van Doxorubicine dosis zo klein als 0,3 µg/g invloed op de reactie van de muizen15. Wanneer ...

Nefrotisch syndroom wordt gekenmerkt door zware Proteïnurie, hypoalbuminemia, oedeem en hyperlipidemie, en kan worden beschouwd als de meest extreme uiting van proteinuric nierziekte. Bij knaagdieren, kan experimentele nefrotisch syndroom worden opgewekt door enkele injectie van anthracyclines of puromycin die leidt tot schade van de podocyte en lijkt op de menselijke minimale verandering ziekte en focale segmentale glomerulosclerosis (FSGS)1. Na de eerste beschrijving in 1955 door Frenk et al. 2, puromycin nucleoside nefrose (PAN) in ratten is een standaard model te onderzoeken de pathofysiologie van nefrotisch syndroom in talrijke studies3,,4,5,6geworden. Bij muizen, kan het desbetreffende model worden opgewekt door de anthracycline Doxorubicine7. Er is echter een sterke stam-afhankelijkheid die is genetisch bepaald door ten minste twee genetische loci8. Daarnaast zijn er verschillen in de proteinuric reactie en het verloop van het nefrotisch syndroom9,10. Met 129S1/SvImJ muizen en snelle intraveneuze injectie van Doxorubicine via de retrobulbaire sinus bereikt, Proteïnurie reacties waarden die toereikend zijn voor het opwekken van de typische kenmerken van nefrotisch syndroom, vooral volume retentie gekenmerkt door ascites en bijna natriumvrij urine7. Natrium retentie in experimentele nefrotisch syndroom heeft zijn ervan uitgegaan dat het resultaat van de activering van het kanaal (ENaC) van de epitheliale natrium in de distale zaadvormende door aberrantly gefilterde serine proteasen zoals Plasmine veroorzaakt proteolyse van de γ-subunit4 ,11,12. Dit concept werd onlangs, bewezen in nefrotisch muizen die werden beschermd tegen Proteolytische ENaC activatie en retentie van natrium door behandeling met de serine protease inhibitor aprotinin dat even effectief als de ENaC blocker amiloride13 was. Om te zorgen voor continue levering aan de distale zaadvormende, werd aprotinin toegediend via subcutaan geïmplanteerde aanhoudende release pellets. Toekomstige studies zijn vereist ter aanduiding van de serine proteasen die verantwoordelijk voor Proteolytische ENaC activering in nefrotisch syndroom, die wordt beschouwd zijn als de menselijke situatie parallel. Voor dit doel is Doxorubicine-geïnduceerde nefrotisch syndroom een waardevol model dat kan worden gebruikt in wild-type muizen of uitgebreid tot genetisch gemanipuleerde muizen. De voordelen zijn lage kosten van de drug, lagere complexiteit van management en goede reproduceerbaarheid14.

Nefrotisch syndroom is de meest extreme uiting van proteinuric nierziekte en gekenmerkt door zware Proteïnurie, hypoalbuminemia en oedeem te wijten aan natrium retentie en hyperlipidemie. Studeren de pathofysiologie van dit syndroom, knaagdier modellen zijn ontwikkeld gebaseerd op de injectie van giftige stoffen zoals Doxorubicine podocyte schade. Bij muizen zijn alleen enkele stammen gevoelig voor dit model. In wildtype 129S1/SvImJ muizen induceert het beheer van Doxorubicine door snelle intraveneuze injectie aan de sinus van retrobulbaire experimentele nefrotisch syndroom dat beschikt over alle symptomen van de ziekte van de mens, met inbegrip van de retentie van natrium en oedeem. Na het begin van Proteïnurie verhoogde muizen vertonen urine serine protease activiteit die tot de activering van het kanaal van de epitheliale natrium (ENaC) en de retentie van natrium leidt. Farmacologische remming van urine serine proteasen door de behandeling met aanhoudende release aprotinin intrekt ENaC activatie en retentie van natrium voorkomt. Dit model is ideaal om te studeren de pathofysiologie van proteasuria, dwz., de uitscheiding van actieve serine proteasen waardoor ENaC activering door de proteolyse van de γ-subunit. Dit kan worden beschouwd als het primaire mechanisme van ENaC activatie en retentie van natrium in proteinuric nierziekte.

Na de inductie van Isofluraan versuffing, was Doxorubicine snel geïnjecteerd via de linker retrobulbaire sinus. Het gehele volume van 7,25 µL/g bw werd geïnjecteerd zonder enige weerstand en extravasation blijkt intraveneuze locatie van de canule juist. De muis verhaald versuffing snel en had geen bijzondere waardevermindering op die dag en daarna. Met name, was er geen teken van schade in het linkeroog.
