IELTS er preget av tunge proteinuria, hypoalbuminemia, ødem og hyperlipidemi, og kan betraktes som den mest ekstreme manifestasjonen av proteinuric nyresykdom. I Red, kan eksperimentelle IELTS indusert ved enkelt injeksjon antracykliner eller puromycin som fører til podocyte skader og ligner menneskelige minimal endre sykdom og fokal Segmentinformasjon glomerulosclerosis (FSGS)1. Etter første beskrivelsen i 1955 av Frenk et al. 2, puromycin nukleosid nefrose (PAN) i rotter har blitt en standard modell å undersøke i Patofysiologien ved IELTS i mange studier3,4,5,6. I mus, kan tilsvarende modellen indusert ved anthracycline doksorubicin7. Men er det en sterk belastning-avhengighet som genetisk etter minst to genetisk loci8. I tillegg finnes forskjeller i proteinuric svar og selvfølgelig IELTS9,10. Bruker 129S1/SvImJ mus og rask intravenøs injeksjon av doksorubicin via retrobulbar sinus, nå proteinuria svar verdier som er tilstrekkelig til å indusere de typiske egenskapene til IELTS, spesielt volum oppbevaring preget av ascites og nesten natrium gratis urin7. Natrium oppbevaring i eksperimentell IELTS har vært antatt å være et resultat av aktivering av epitelial natrium kanalen (ENaC) i den distale tubule av aberrantly filtrert serine proteaser som plasmin forårsaker proteolyse av sin γ-delenhet4 ,11,12. Nylig ble dette konseptet påvist i nephrotic mus som var beskyttet fra proteolytisk ENaC aktivisering og natrium oppbevaring av behandling med serine protease hemmer aprotinin som var like effektivt som ENaC blokkering amilorid13. IELTS er mest ekstreme manifestasjonen av proteinuric nyresykdom og preget av tunge proteinuria, hypoalbuminemia og ødem på grunn av natrium oppbevaring og hyperlipidemi. For å studere i Patofysiologien ved dette syndromet, basert gnager modeller er utviklet på injeksjon av giftige stoffer som doksorubicin forårsaker podocyte skade. Denne modellen er ideell for å studere i Patofysiologien ved proteasuria, dvs., utskillelse av aktive serine proteaser at ENaC aktivisering ved protolyse av sine γ-delenhet. Dette kan betraktes som den primære mekanismen for ENaC aktivisering og natrium oppbevaring i proteinuric nyresykdom.

Etter induksjon av isoflurane narkose, ble doksorubicin raskt inn venstre retrobulbar sinus. Hele volumet av 7.25 µL/g bw ble injisert uten noen motstand og bloduttredelse beviser riktig intravenøs plasseringen av kanyle. Musen utvinnes fra narkose raskt og hadde ingen verdifall på den dagen og deretter. Spesielt, var det ingen tegn til skade på det venstre øyet. Etter doksorubicin injeksjon, mat og væske inntak falt over de første 3 dagene på grunn av en generell toksiske effekte...
