小さなRho GTPaseは、多様な細胞機能を持つスーパーファミリーのタンパク質です。例えば、これらのタンパク質は、細胞の運命、ストレスに対する細胞応答、細胞運動性、および細胞周期の調節に関与している。小さなRho GTPaseタンパク質の活性は、翻訳後修飾によって調節される。これらの変更は、その活動の厳しい規制に関与しています。例として、タンパク質プレニル化とGTP結合は、これらのタンパク質の翻訳後修飾の最も重要なです。私の研究室では最近、これらのスーパーファミリータンパク質の翻訳後修飾を調査するための簡単な方法を変更し、開発しました。そのために、251個の神経膠芽腫細胞株を用いて、シンバスタチンで標的化し、これらのタンパク質の翻訳後修飾を測定しました。この方法は、このタンパク質スーパーファミリーに対する細胞下分画およびGTP結合タンパク質を含む。このために、RhoAの局在化、およびRhoAに結合したGTPを用いて、このタンパク質の翻訳後修飾を測定しました。37度インキュベーターから細胞を取り除く。顕微鏡下の細胞を見て、合流性を確認します。細胞は70〜80%コンフルエントであるべきである。冷たいPBSで細胞を一度洗います。EDTAを5ミリリットル加え、細胞を37度インキュベーターに5分間戻します。5分間のインキュベーションの後、15ミルチューブに細胞を持つEDTAを収集します。チューブを氷箱に入れ、遠心分離機に進みます。遠心分離機を4度で1500gに設定し、5分間回転させます。スピンに続いて、スーパーネイトを取り除き、冷たいPBSを1ミリリットル加えます。細胞をうまくトリチュレートします。細胞混合物を新しいラベル1.5ミリリットルチューブに移します。移管後、遠心分離機に進みます。遠心分離機を4度で1500gに設定し、5分間回転させます。ペレットのサイズを確認します。サンプルを氷の上に置きます。ペレットを邪魔することなく、スーパーネイトを完全に捨てます。バッファ 1 を追加します。上下にピペットを入れ、よく混ぜます。超音波処理に進みます。5サイクル、それぞれ5秒の超音波処理器を設定し、3回繰り返します。超音波処理は、説明の目的のために、氷の上で進む必要がありますが、ビデオでは示されていません。超遠心分離機に進みます。超遠心分離機を使用して、細胞の均質化物を細胞質および膜分画に分離します。遠心分離機を35分間100,000gに設定し、4度に設定します。ペレットのサイズを確認します。スーパーネイトを分離します。ペレットを乱さないよう注意して、完全に超自然を収集します。上華は、細胞の分画です。新しくラベル付けされたチューブにスーパーネイトを配置します。ペレットにバッファー 2 を追加します。これは膜分画です。上下にピペットを入れ、よく混ぜます。タンパク質定量およびウェスタンブロットサンプル調製に進みます。15%ゲルを使用して、サンプルをSDS-PAGEにロードします。分子量マーカーを可視化するか、またはポンソー染色体を用いてタンパク質の転写を確認する。免疫ブロット分析のための一次抗体を追加します。Rac1/2/3、Cdc42、および RhoA を使用しています。培養プレートをインキュベーターから取り出します。顕微鏡下の細胞を見て、合流性を確認します。細胞は70〜80%コンフルエントでなければなりません。培養プレートを氷の上に置きます。培地を吸引し、7.2に氷冷PBS pHで細胞を洗浄します。PBSを吸引する。PBSのすべての残骸を取り除くために氷の上にプレートを傾けます。残留PBSは、このアッセイに悪影響を及ぼす。プロテアーゼおよびホスファターゼ阻害剤を含む氷冷ライシス緩衝液の適切な量で細胞をリセする。収穫細胞は、細胞スクレーパーでライセートします。この技術の培養プレートを傾斜させます。ライセートをラベル付きの氷冷クライオチューブに移し、氷の上に置きます。渦を使って十分に混ぜます。タンパク質アッセイのために10マイクロリットルのライセートを保管してください。液体窒素中の残りの細胞ライゼを凍結スナップします。スナップ冷凍クライオチューブをマイナス80冷凍庫に移します。これらのサンプルを保存します。なお、サンプルは14日以上保存しないでください。ステップ1~8を1つずつ繰り返して、他の培養プレートから細胞ライセートを調製する。迅速に作業し、10分以上氷の上にサンプルを残すことはありません。すべての培養プレートを同時に扱うことはありません。タンパク質濃度を測定します。サンプル間のタンパク質の濃度を正規化するために、リシスバッファーの体積を計算します。注:1ミリリットル当たり1ミリグラムは通常最高ですが、ミリリットル当たり0.3〜2ミリグラムは検出可能です。60マイクロリットルのライシスバッファーを加え、60マイクロリットルの結合バッファーをマイクロチューブに加えてブランクコントロールを用意します。12マイクロリットルのRho対照タンパク質、48マイクロリットルのリシスバッファー、および60マイクロリットルの結合バッファーを添加して、ポジティブコントロールを準備します。ローアフィニティプレートを袋から取り出し、氷の上に置きます。100マイクロリットルの氷冷蒸留水で粉末を井戸に溶かします。プレートを氷の上に置いておきなさい。25°Cに設定された水浴でスナップ冷凍細胞ライセートを解凍します。各サンプルに氷冷ライシスバッファーの計算された量を加え、タンパク質濃度を正規化します。60マイクロリットルの正規化された氷冷サンプルをマイクロチューブに移し、60マイクロリットルの結合バッファーを加えます。よく混ぜ、氷の上に置いておく。激しいフリックでマイクロプレートから水を完全に取り除き、ラボマットに5〜7回のハードタップを加えます。正規化されたサンプルの50マイクロリットル、ブランクコントロール、および正のコントロールを重複する井戸に加えます。プレートを軌道シェーカーの上に30分間摂氏4度で置きます。注:シェーカーの速度は300 rpmに設定する必要があります。フリックしてプレートからサンプルを取り除き、200マイクロリットルの室温洗浄バッファーで2回洗浄します。各洗浄後に各洗浄後にウェルから洗浄バッファーを激しく取り出し、続いてタッピングし、プレートを室温でベンチに置きます。各ウェルに200マイクロリットルの室温抗原提示バッファーを加え、室温で2分間インキュベートします。ウェルから溶液をフリックアウトし、室温洗浄バッファーの200マイクロリットルで3回洗浄します。調製したての1~250個の抗RhoA一次抗体を50マイクロリットルずつ各ウェルに加えます。プレートを軌道シェーカーの上に300 rpmで45分間置き、摂氏25度に設定します。井戸から溶液をフリックアウトし、室温で200マイクロリットルの洗浄バッファーで3回洗浄します。50マイクロリットルの作製した1~250個の抗RhoA二次抗体を各ウェルに加えます。プレートを軌道シェーカーの上に45分間置き、300 rpmで摂氏25度に設定します。各ウェルから溶液を取り出す試薬Aと試薬B.Flickの等量を混合してHRP検出試薬を調製し、室温洗浄バッファーの200マイクロリットルで3回洗浄します。調製したHRP検出試薬を50マイクロリットルずつ各ウェルに加えます。3~5分以内に発光信号を読み、最大信号を得る。適切なソフトウェア パッケージを使用して結果を分析します。我々の結果は、シンバスタチンが膜分画におけるRhoAの局在を減少させ、示した。増加したのは、細胞の分数に蓄積する。興味深いことに, シンバスタチンは、RhoAにバインドされた GTP を増加.なぜ?シンバスタチンはこの活動を減少させると予想されます。このタンパク質の細胞下局在化を測定し、このタンパク質に結合するGTPを測定して、小さなRho GTPasesの活性を最終的に評価することが非常に重要です。
