De intracraniële subarachnoïdale route van infectie beschreven hier heeft duidelijke voordelen ten opzichte van andere routes van infectie. Streptococcus suis (S. suis) is een grote bacteriële ziekteverwekker van varkens wereldwijd, waardoor ernstige ziekten, met inbegrip van hersenvliesontsteking, longontsteking, septikemie, endocarditis en artritis1. Het is ook een opkomende zoönoseverwekker. Tot nu toe, is het gemeld dat negen serotypen infectie bij de mens veroorzaken kunnen, met inbegrip van serotypen 2, 4, 5, 9, 14, 16, 21, 24 en 312,3,4. Bij mensen en varkens is meningitis een van de belangrijkste klinische symptomen van S. suis infecties. In Vietnam en Thailand is S. suis de voornaamste oorzaak van meningitis in volwassenen5. Microbiële biofilms zijn micro-organismen die zich aan elkaar en zijn geconcentreerd op een interface; ze zijn essentieel voor bacteriële virulentie, overleven in uiteenlopende omgevingen en antibioticaresistentie5. Biofilms zijn meestal omringd door een extracellulaire matrix waarin over het algemeen polysacchariden, eiwitten en DNA-6. De laatste kan uitlokken host inflammatoire reacties en cytokine productie7. Biofilm vorming is gemeld te worden betrokken bij de streptokokken meningitis in eerdere studies. Biofilms bijdragen tot Streptococcus agalactiae meningitis in een model van de vis tilapia en biofilm vorming binnen de hersenen weefsels is geopenbaard en rond meningeal oppervlakken in vivo via intra-abdominale inoculatie8. Tijdens de meningitis, Streptococcus pneumoniae is in een biofilm-achtige toestand en bacteriën in een dergelijke biofilm staat waren effectiever in het inducerende meningitis in een muis besmetting model9. Daarnaast, in onze vorige bestuderen, de biofilm staat gekoppeld aan S. suis in muis hersenen draagt bij aan bacteriële virulentie door survival analyse10. Direct bewijs voor biofilm betrokkenheid bij S. suis meningitis moet echter verder onderzocht worden.
Dierlijke modellen van S. suis -infectie hebben ontwikkeld in muizen met behulp van de intraperitoneaal (i.p.)11, intranasale (i.n.)12, intraveneuze (i.v.)13en de intracisternal (IC) routes van infectie14, 15 , 16. de i.p. i.n. en i.v. routes van infectie zijn echter niet geschikt voor de studie van de rollen van S. suis oppervlakte componenten in meningitis direct in de hersenen. Het gaat hierbij om extracellulaire matrix van biofilms. Hoewel de i.c. inoculatie werd gebruikt voor S. suis -infectie, is de precieze injectieplaats niet beschreven in deze documenten. De stereotaxic coördinaten van de injectieplaats voor intracraniële subarachnoïdale inoculatie is daarentegen duidelijk beschreven in een eerdere studie17. Dit toegestaan eenvoudige erkenning van de inoculatie punt en meer simplistische experimenteel protocol. Bovendien, bootst de intracraniële subarachnoïdale route van infectie bacteriële ingang in het centrale zenuwstelsel van de sinussen of het middenoor17, en de relatie tussen het middenoor en meningitis veroorzaakt door S. suis Madsen et al.18heeft aangetoond. Bovendien, door toepassing van de intracraniële subarachnoïdale route van infectie in muizen, hebben wij aangetoond dat S. suis kleine RNA rss04 tot meningitis in onze eerdere studie10 bijdraagt.
In de huidige studie, de intracraniële subarachnoïdale route van infectie werd gebruikt in muizen te onderzoeken van de rol van biofilms in S. suis meningitis. Muizen werden besmet met planktonische cellen of biofilm staat cellen van S. suis door deze route van infectie. Histopathologische analyses en verhoogde mRNA uitdrukking van TLR2 en cytokines van hersenweefsel van muizen ingespoten met cellen van biofilm staat duidelijk aangegeven dat S. suis biofilm tot meningitis bijdraagt.

intracranial subarachnoidal route of infection for investigating roles of streptococcus suis biofilms in meningitis in a mouse infection model

Streptococcus suis is niet alleen een grote bacteriële ziekteverwekker van varkens wereldwijd, maar ook een opkomende zoönoseverwekker. Bij mensen en varkens is meningitis een grote manifestatie van S. suis infecties. Een model geschikt infectie is een essentieel instrument om het begrijpen van de mechanismen van ziekten veroorzaakt door pathogenen. Verschillende routes van infectie in muizen hebben ontwikkeld om te bestuderen van de pathogenese van S. suis -infectie. De routes van het intraperitoneaal intranasale en intraveneuze van infectie zijn echter niet geschikt voor de studie van de rollen van S. suis oppervlakte componenten in meningitis direct in de hersenen, zoals de extracellulaire matrix van biofilms. Hoewel intracisternal inoculatie is gebruikt voor S. suis -infectie, is de precieze injectieplaats niet beschreven. Hier, werd de intracraniële subarachnoïdale route van infectie beschreven in een muismodel te onderzoeken van de rol van biofilms in S. suis meningitis. S. suis planktonische cellen of biofilm staat cellen werden direct geïnjecteerd in de subarachnoïdale ruimte van muizen via de injectieplaats ligt 3,5 mm rostraal van de bregma. Histopathologisch analyse en verhoogde mRNA uitdrukking van TLR2 en cytokinen van het hersenweefsel van muizen ingespoten met cellen van biofilm staat duidelijk aangegeven dat S. suis biofilm definitieve rollen in S. suis meningitis speelt. Deze route van infectie heeft duidelijke voordelen ten opzichte van andere routes van infectie, waardoor de studie van de gastheer-bacterie interactie. Bovendien, het maakt het mogelijk het effect van bacteriële onderdelen op de immuunrespons van de gastheer direct in de hersenen worden beoordeeld, en bootst bacteriële ingang in het centrale zenuwstelsel. Deze route van infectie kan worden uitgebreid voor het onderzoeken van de mechanismen van meningitis veroorzaakt door andere bacteriën. Daarnaast kan het ook worden gebruikt voor het testen van de werkzaamheid van geneesmiddelen tegen bacteriële meningitis.

SEM analyse werd uitgevoerd om te onderzoeken van biofilm vorming onder de proefomstandigheden.
