For å oppnå høyere MSC differensiering effektivitet, er flere aspekter avgjørende. En er kultur tilstand iPSCs før du starter MSC differensiering. Protokollen presentert i manuskriptet er basert på tidligere studier 9,17. iPSCs må være kultivert på MEFs i minst 2 uker. Mellom 2006 og 2007 førte flere gjennombrudd funn fra laboratoriene Dr. Shinya Yamanaka og James A. Thomson til utvikling av menneskeskapte pluripotent stamceller (iPSCs)1,2,3. Omprogrammere somatiske celler med definerte transcriptional faktorer til skjemaet iPSCs, forskerne kunne generere celler med viktige egenskaper nemlig, pluripotency og selvtillit fornyelse, som var tidligere antatt å bare finnes i menneskelige embryonale stamceller (hESCs). iPSCs kan genereres fra individ eller pasienten og måtte ikke være avledet fra embryo, vesentlig utvide repertoaret tilgjengelig sykdommer og bakgrunner for studier. Siden da har pasient-avledede iPSCs blitt brukt til recapitulate fenotypen av ulike menneskelige sykdommer, Alzheimers sykdom4 og amyotrofisk lateral sklerose5 lang QT syndrom6,7, 8.
Disse fremskrittene i iPSC forskning har også åpnet nye muligheter for kreftforskning. Flere grupper har nylig brukt pasienten iPSCs modell kreft utvikling under en utsatt genetisk bakgrunn9,10,11med vellykket program vist hittil i osteosarcoma9, leukemi10,11,12og endetarmskreft13. Selv om iPSC-avledet kreft modeller er fortsatt i sin barndom, har de vist stort potensial i phenocopying sykdomsassosierte malignitet, Klargjørende patologisk mekanismer, og identifisere terapeutiske forbindelser14.
Li-Fraumeni syndrom (AKU) er en autosomal dominant arvelig kreft lidelse forårsaket av TP53 germline mutasjon15. Pasienter med AKU er disponert for ulike typen malignitet inkludert osteosarcoma, gjør AKU iPSCs og deres avledet celler spesielt godt egnet til å studere denne malignitet16. En iPSC-baserte osteosarcoma modell først ble etablert i 2015 bruker AKU pasient-avledede iPSCs9 senere differensiert til stamceller (MSCs) og deretter til osteoblasts, de stammer cellene av osteosarcoma. Disse AKU osteoblasts recapitulate osteosarcoma-assosiert osteogenic differensiering defekter og kreftfremkallende egenskaper, demonstrere modellen potensielle som "bein svulst i en rett" plattform. Interessant, genomet hele transcriptome analyser åpenbare aspekter av en osteosarcoma genet signatur i AKU osteoblasts og som kjennetegner denne AKU gene expression profilen er korrelert med dårlig prognose i osteosarcoma9, som angir den potensialet i AKU iPSCs sykdom modeller å avsløre funksjoner kliniske relevans.
Dette manuskriptet gir en detaljert beskrivelse av hvordan du bruker AKU pasient-avledede iPSCs til modell osteosarcoma. Det detaljer generering av AKU iPSCs, differensiering av iPSCs MSCs og deretter til osteoblasts, og bruk av en i vivo xenograft modellen med AKU osteoblasts. AKU sykdom modellen består av flere fordeler, blant annet muligheten til å generere ubegrenset celler i alle stadier av osteosarcoma utvikling for mekanistisk studier, biomarkør identifikasjon og narkotikarelaterte screening9,14, 16.
Oppsummert tilbyr AKU iPSC-baserte osteosarcoma modellen en attraktiv komplementære system for fremme osteosarcoma forskning. Denne plattformen gir også en proof-of-concept for kreft modellering bruker pasient-avledede iPSCs. Denne strategien beskrevet nedenfor kan lett utvides til modell malignitet forbundet med andre genetiske lidelser med kreft predisposisjoner.

modeling osteosarcoma using li-fraumeni syndrome patient-derived induced pluripotent stem cells

Li-Fraumeni syndrom (AKU) er en autosomal dominant arvelig kreft lidelse. Pasienter med AKU er disponert for ulike typen svulster, herunder osteosarcoma - en av de mest primære ikke-hematologic malignitet i barndom og oppvekst. AKU gir derfor en ideell modell for å studere denne kreft. Utnytter iPSC metoder, AKU-assosiert osteosarcoma kan modelleres vellykket ved å skille AKU pasienten iPSCs til stamceller (MSCs), og deretter til osteoblasts - cellene i opprinnelse for osteosarcomas. Disse AKU osteoblasts recapitulate kreftfremkallende egenskaper osteosarcoma, gir en attraktiv modellsystem for skildre patogenesen av osteosarcoma. Dette manuskriptet viser en protokoll for generering av iPSCs fra AKU pasienten fibroblaster, differensiering av iPSCs til MSCs, differensiering av MSCs osteoblasts, og i vivo tumorigenesis bruker AKU osteoblasts. Denne iPSC sykdom modell kan utvides for å identifisere potensielle biomarkers eller terapeutisk mål for AKU-assosiert osteosarcoma.
