Plant ziekte resistentiegenen spelen een essentiële rol bij het voorkomen van infecties door ziekteverwekkers, met inbegrip van schimmel ziekteverwekkers1,12. Rijst blast is gebruikt als een model te begrijpen van de aard van pathogen bevolking structuren en genen4van de weerstand van de plant. Daarom is het noodzakelijk de ziekte weerstand genotype en avirulentie genotypen van de belangrijkste soorten vezelgewassen op grote schaal om zie...

Rijst blast, veroorzaakt door M. grisea, is een van de ernstigste ziekten voor rijst variëteiten wereldwijd1,2. Het proces waarmee M. grisea gastheerplanten infecteert omvat een conidiën productie als oppervlakte gehechtheid, een conidiën kieming en de vorming van appressorium, een formatie van de penetratie peg en besmettelijke schimmeldraad differentiatie, en een ziekte verspreid 3. al deze fasen zijn gebruikelijk in vele andere plant plantpathogene schimmels en, inderdaad, een blokkade van alle ééntraps voorkomt dat de besmetting van waardplanten. Als gevolg van zijn economische belang en de genetische werkwillig, heeft M. grisea ontpopt als een modelorganisme voor het bestuderen van de mechanismen van de plant-schimmel pathogeen interacties1,4. Dus, het bestuderen van de moleculaire basis van deze ontwikkelingsstadia in M. grisea zal helpen ophelderen van de moleculaire mechanismen die ten grondslag liggen aan de schimmel pathogeniteit en de identificatie van kandidaat-doelgenen voor screening en ontwerpen roman fungiciden5.
Recente verslagen over M. grisea infectie hebben gericht op de moleculaire mechanismen van de stadia van de pre-penetratie, met name de conidiation, de appressorium-vorming, de penetratie pinnen en de besmettelijke groei3, 6. het is daarom essentieel voor de ontwikkeling van een gedetailleerd protocol om te testen van M. grisea infectie. Hierin presenteren wij een gedetailleerde methode voor een infectie-test die maakt gebruik van de spray-gemedieerde infectie testen met een conidial vering en de inoculatie van wonden met mycelial stekkers van M. grisea. In dit verslag, het protocol is gericht op de cultuur van stammen, de voorbereiding van de conidiation oplossing voor het spuiten, en de mycelial plug-gemedieerde inoculatie van planten met M. grisea. Deze stappen worden beschreven in detail hieronder, en een schematische weergave van de volledige workflow van de methode en een typische laesie worden weergegeven in de figuren 1 en 2, respectievelijk.

Planten bezitten een krachtig systeem om zich te verdedigen tegen potentiële bedreigingen door pathogene schimmels. Voor agrarisch belangrijke planten, echter het is gebleken dat huidige maatregelen ter bestrijding van deze ziekteverwekkers te conservatief en, dus, niet voldoende effectief, en ze kunnen potentieel vormen milieurisico's. Daarom is het uiterst noodzakelijke host-weerstand factoren bij het controlerende plantenziektes natuurlijk door de identificatie van resistente kiemplasma, de isolatie en karakterisatie van resistentiegenen en de moleculaire veredeling te identificeren van resistente cultivars. In dit verband is er nodig om een nauwkeurige, snelle en grootschalige beëntingsmethode om te fokken en ontwikkelen van resistentiegenen plant. De rijst blast schimmel pathogeen Magnaporthe grisea oorzaken ernstige ziekte symptomen en verliezen opleveren. Onlangs, M. grisea heeft ontpopt als een modelorganisme voor het bestuderen van de mechanismen van plant-schimmel pathogeen interacties. Vandaar, rapporteren we de ontwikkeling van een plant virulentie testmethode die specifiek is voor M. grisea. Deze methode biedt voor zowel spray inoculatie met een conidial suspensie en verwonden inoculatie met mycelium kubussen of druppels van conidial schorsing. De belangrijkste stap van de verwonding beëntingsmethode voor vrijstaande rijst bladeren is om wonden op de bladeren van de plant, die elke storing die wordt veroorzaakt door gastheer penetratieweerstand vermeden. Deze spray/verwonden protocol draagt bij aan de snelle, nauwkeurige en grootschalige screening van de biotypen van M. grisea isolaten. Dit geïntegreerd en systematische plant infectiemethode zal dienen als een uitstekend uitgangspunt voor het verkrijgen van een breed perspectief van onderwerpen in de fytopathologie.
