Plante, gènes de résistance jouent un rôle essentiel dans la prévention des infections par des agents pathogènes, y compris les champignons pathogènes1,12. Pyriculariose, causée par M. grisea, est l’une des maladies plus graves pour les variétés de riz dans le monde1,2. Le processus par lequel M. grisea infecte les plantes hôtes comprend une production de conidies et l’attachement de surface, une germination des conidies et la formation de l’appressorium, une formation de l’hyphe de pénétration et de différenciation de l’hyphe infectieux, et la propagation d’une maladie 3. toutes ces étapes sont communs dans de nombreux autres champignons phytopathogènes, et, en effet, un blocus de n’importe quelle étape unique empêche l’infection des plantes hôtes. En raison de son importance économique et de la traçabilité génétique, m.grisea est devenu un organisme modèle pour étudier les mécanismes de la plante-champignon pathogène des interactions1,4. Par conséquent, étudier la base moléculaire de ces stades de développement de m.grisea aidera à élucider les mécanismes moléculaires qui sous-tendent la pathogénicité fongique et l’identification des gènes de cible de candidats pour le dépistage et le roman de conception 5de fongicides.
Les rapports récents concernant M. grisea infection ont mis l’accent sur les mécanismes moléculaires des stades avant la pénétration, surtout la conidiation, la formation de l’appressorium, les hyphes de pénétration et la croissance infectieuses3, 6. par conséquent, il est essentiel d’élaborer un protocole détaillé pour tester l’infection de M. grisea . Ici, nous présentons une méthode détaillée pour un test de l’infection qui utilise des tests d’infection véhiculée par pulvérisation avec une suspension de conidies et l’inoculation des plaies avec des bouchons mycéliennes de m.grisea. Dans ce rapport, le protocole met l’accent sur la culture des souches, la préparation de la solution de la conidiation pour la pulvérisation et l’inoculation mycélienne d’induite par la prise de plantes avec M. grisea. Ces étapes sont décrites en détail ci-dessous et une vue schématique montrant l’intégralité du workflow de la méthode et une lésion typique sont présentés aux Figures 1 et 2, respectivement.

Les plantes possèdent un système puissant pour se défendre contre d’éventuelles menaces de champignons pathogènes. Pour les plantes agricoles importantes, cependant, les mesures actuelles pour lutter contre ces agents pathogènes ont prouvé trop conservateurs et, ainsi, pas suffisamment efficace et ils peuvent potentiellement poser des risques environnementaux. Par conséquent, il est extrêmement nécessaire identifier les facteurs de résistance de l’hôte pour aider au contrôle des maladies des plantes naturellement par le biais de l’identification du matériel génétique résistant, l’isolement et caractérisation des gènes de résistance et la sélection moléculaire des cultivars résistants. À cet égard, il y a besoin d’établir une méthode d’inoculation précise, rapide et à grande échelle pour se reproduire et de développer des gènes de résistance des plantes. Le riz souffle champignon pathogène Magnaporthe grisea causes symptômes sévères de la maladie et les pertes de rendement. Récemment, M. grisea est devenu un organisme modèle pour étudier les mécanismes des interactions plante-pathogène. Par conséquent, nous présentons l’élaboration d’une méthode de test de virulence de plante qui est spécifique pour m.grisea. Cette méthode fournit pour l’inoculation de pulvérisation avec une suspension de conidies et blessant l’inoculation avec cubes de mycélium ou gouttelettes de suspension de conidies. L’étape clé de la méthode d’inoculation blessant pour les feuilles de riz détaché est de faire des plaies sur les feuilles de la plante, qui évite toute interférence causée par la résistance à la pénétration hôte. Ce spray/blessant protocole contribue au dépistage rapid, précis et à grande échelle des pathotypes de m.grisea isolats. Cette intégrée et méthode d’infection systématique des plantes servira de point de départ idéal pour gagner une large perspective de questions en phytopathologie.
