Brusone do arroz, causada pelo M. grisea, é uma das doenças mais graves para as variedades de arroz em todo o mundo1,2. O processo pelo qual M. grisea infecta plantas hospedeiras inclui uma produção de conídios e a fixação de superfície, uma germinação de conídios e a formação de apressório, uma formação do peg de penetração e diferenciação de hifa infecciosa, e uma doença se espalhar 3. todas as etapas são comuns em muitos outros fungos patogênicos de plantas, e, de fato, um bloqueio de qualquer único estágio impede a infecção de plantas hospedeiras. Portanto, estudar a base molecular destes estágios do desenvolvimento de M. grisea ajudará a elucidar os mecanismos moleculares subjacentes à patogenicidade de fungosa e a identificação de genes-alvo candidato para triagem e projetando romance fungicidas,5.
Relatórios recentes relativas à infecção de M. grisea centraram-se sobre os mecanismos moleculares das fases pré-penetração, especialmente o conidiation, a formação de apressório, os pinos de penetração e o crescimento infecciosas3, 6. por conseguinte, é essencial para desenvolver um protocolo detalhado para testar infecção M. grisea . Neste documento, apresentamos um método detalhado para testes de infecção que utiliza ensaios de infecção mediada por pulverização com uma suspensão conidiais e a inoculação de feridas com plugues micelial de M. grisea. Neste relatório, o protocolo centra-se na cultura de cepas, a preparação da solução para a pulverização de conidiation e a inoculação do micélio plug-mediada de plantas com M. grisea. Essas etapas são descritas em detalhes abaixo e uma visão esquemática mostrando o fluxo de trabalho inteiro do método e uma lesão típica são mostrados nas figuras 1 e 2, respectivamente.

As plantas possuem um sistema poderoso para se defender contra potenciais ameaças por fungos patogênicos. Para plantas agronomicamente importantes, no entanto, medidas atuais para combater tais patógenos revelaram-se demasiado conservadoras e, assim, não é suficientemente eficaz e eles podem potencialmente representar riscos ambientais. Portanto, é extremamente necessário identificar fatores de resistência do hospedeiro para auxiliar no controle de doenças de plantas naturalmente através da identificação de germoplasma resistente, o isolamento e caracterização de genes de resistência e a reprodução molecular de cultivares resistentes. A este respeito, há necessidade de estabelecer um método de inoculação precisa, rápida e em larga escala para criar e desenvolver genes de resistência da planta. O arroz bombardear o patógeno fungo Magnaporthe grisea causas sintomas de doença grave e perdas de rendimento. Recentemente, M. grisea tem emergido como um organismo modelo para estudar os mecanismos de interações planta-fungo patogênico. Daí, nós relatamos o desenvolvimento de um método de teste de virulência de planta que é específico para M. grisea. Este método fornece para inoculação de pulverizador com uma suspensão conidiais e ferindo inoculação com cubos de micélio ou gotículas de suspensão conidiais. A etapa chave do método de inoculação ferindo para folhas de arroz destacado é tornar-se feridas nas folhas da planta, que evita qualquer interferência causada pela resistência à penetração do anfitrião. Este pulverizador/ferir protocolo contribui para o rastreio rápido, preciso e em grande escala das raças de isolados de M. grisea . Este integrado e o método de infecção sistemática vegetal irá servir como um excelente ponto de partida para a obtenção de uma perspectiva ampla de questões em Fitopatologia.
