Cette technique est une méthode utile pour la création de perte soudaine, permanente et complète de la fonction vestibulaire. Cela pourrait servir à étudier les pathologies vestibulaires, tels que la névrite vestibulaire, une tumeur acoustique et la maladie de Ménière. De nombreuses études ont utilisé cette technique pour étudier la plasticité neuronale des noyaux vestibulaires ou les processus centraux liés5,17,18<...

L’organe vestibulaire est essentiel pour le contrôle de balance et oculaires. Une fonction vestibulaire normale dépend des signaux afférents symétriques des organes vestibulaire de l’oreille interne deux. Hypofonction vestibulaire ou perte provoque le vertige, nystagmus et déséquilibre postural. Après lésion aiguë, la fonction vestibulaire récupère spontanément en quelques jours, un processus appelé compensation vestibulaire1,2. La compensation vestibulaire des déficits statiques est un processus de récupération lié au déséquilibre de l’activité spontanée de repos entre les noyaux vestibulaires homolatéral et controlatéral. La compensation vestibulaire des déficits dynamiques est obtenue principalement par l’intermédiaire des substitutions sensoriels et comportementaux (à l’aide des entrées visuelles ou somesthésiques)3. Ces processus sont attrayants pour la plasticité neuronale études4,5.
Un certain nombre de méthodes ont été développé pour étudier le système vestibulaire et les mécanismes qui sous-tendent la plasticité neuronale au cours de compensation vestibulaire, comme labyrinthectomie chirurgicale et chimique et neurectomie vestibulaire5,6 ,7,8. Neurectomie vestibulaire est une certaine façon à induire la perte vestibulaire complète, mais c’est une procédure plus difficile et plus invasive et peut provoquer des lésions cérébrales8,9. Cette méthode nécessite une plus grande dextérité chirurgicale et prend plus de temps que labyrinthectomie. Labyrinthectomie chimique notamment gentamycine, arsanilate et tétracaïne, est plus facile et peut produire des résultats fiables10,11,12. Toutefois, la cochlée peut-être également être endommagée et perte vestibulaire peut se développer au fil du temps,11. En outre, les effets des produits chimiques sur le cerveau, ce qui devrait être préservée pour une évaluation précise, ne sont pas claires. Labyrinthectomie chirurgicale a été introduite dans les études animales en 184215 et a été signalée chez le rat en 1936,16. Cette technique a depuis été utilisée dans de nombreux animaux études5,17,18,19. Labyrinthectomie chirurgicale est une méthode fiable et relativement simple de propre. 13 , 14 par ailleurs, les symptômes de lésions vestibulaires sont vus immédiatement après la chirurgie. Nous décrivons ici notre technique chirurgicale pour rat labyrinthectomie.

surgical labyrinthectomy of the rat to study the vestibular system

Afin d’étudier le système vestibulaire ou le processus de compensation vestibulaire, un certain nombre de méthodes ont été développé pour endommager vestibulaire, y compris une labyrinthectomie chirurgicale ou chimique et neurectomie vestibulaire. Labyrinthectomie chirurgicale est une méthode relativement simple, fiable et rapide. Nous décrivons ici la technique chirurgicale pour rat labyrinthectomie. Une postauricular incision est pratiquée sous anesthésie générale pour exposer le conduit auditif externe et la membrane tympanique, après quoi le tympan et les osselets sont supprimés sans l’étrier. L’artère de l’étrier, qui se situe entre l’étrier et la fenêtre ovale, est une structure vulnérable et doit être préservé pour obtenir un champ clair chirurgical. Un trou de fenêtre le vestibule est fait avec une fraise de 2,1 mm foret supérieur de l’étrier. Puis, 100 % d’éthanol est injecté à travers ce trou et aspiré plusieurs fois. Une dissection minutieuse sous un microscope et un contrôle minutieux du saignement sont essentiels pour obtenir des résultats fiables. Symptômes de perte vestibulaire, comme un nystagmus, inclinaison de la tête et un mouvement de roulis, sont vus immédiatement après la chirurgie. Le test de chaise rotarod ou rotation peut être utilisé pour la fonction vestibulaire doit être évaluée objectivement et quantitativement.

Le succès de la chirurgie a été validé par des tests comportements. Tous les animaux ont montré le comportement typique d’une perte unilatérale de la fonction vestibulaire.
