Et kritisk aspekt i denne protokol maksimering overlevelsesevne af celler. At sikre, at væv og celler er altid i iskolde carboxygenated sACSF er nødvendigt at fremme celle overlevelse. Korrekt kortikal funktion kræver en balance mellem excitatoriske projektion neuroner og hæmmende GABAergic interneurons, en meget heterogene befolkning med særskilte morfologier, elektrofysiologiske egenskaber, tilslutningsmuligheder og neurokemiske markører. Unormal udvikling og funktion af interneurons (og specifikke interneuron undergrupper) har været knyttet til pathobiology for psykiatriske lidelser som skizofreni, autisme og epilepsi1,2,3. Desuden, mange gener involveret i disse hjernen lidelser er stærkt beriget i unge interneurons4. Således en større forståelse af de mekanismer, der regulerer interneuron skæbne beslutsomhed og modning er nødvendig for at forstå normale udvikling og potentielle etiologies af talrige hjernesygdomme.
Forhjernen interneurons er født primært fra to forbigående embryonale strukturer, de mediale og caudale ganglionære eminences (MGE og CGE, henholdsvis). Disse postmitotic celler (interneuron prækursorer) derefter gennemgå en langvarig tangential migration fase at sprede sig i hele telencephalon hvor de integreres i en bred vifte af kredsløb. MGE-afledte interneurons består af tre stort set ikke-overlappende, neurochemically definerede undergrupper: hurtigt spiking parvalbumin (PV+) interneurons, ikke-fast spiking somatostatin (SST+) interneurons, og sent spiking neuronal salpetersyre nitrogenoxid syntase (nNOS+) interneurons, der udgør hippocampus neurogliaform og vedbend celler. Talrige undersøgelser har transplanteret embryonale MGE celler direkte ind i en række forskellige områder af hjernen, med konsensus resultaterne, der podede celler modne og frigive GABA for at generelt hæmme den lokale endogene kredsløb14,15, 16,17,18,19. Disse lovende observationer har genereret betydelig interesse i at bruge menneskelige inducerede pluripotente stamceller (hIPSC)-afledt interneurons for at behandle en række hjernesygdomme. Men meget få af disse undersøgelser vurdere hvis disse podede celler modne i de forventede former for modne interneurons, et afgørende element, når man tænker på translationel tilgange.
For at løse, hvordan miljøet påvirker interneuron differentiering og modning, udtænkt en strategi for at omplante umodne interneuron prækursorer til ny hjerne miljøer for at undersøge, om podede interneurons vedtage funktioner af værten miljøet eller bevare funktioner fra donor miljø20. MGE transplantationer er ikke egnede til at behandle dette spørgsmål, fordi MGE indeholder en blandet population af interneuron og GABAergic projektion celler, der spredes i hele mange hjernen regioner21. Uden at vide hvor disse MGE celler ville have overført, man ikke fuldt ud kan vurdere hvordan disse transplantationscentrene påvirkes af hjernen miljø. Ved høst interneuron prækursorer på tidlige postnatal timepoints, dette problem er omgået ved at opnå umodne celler, der har afsluttet deres migration og nået deres mål hjernen regionen men har minimal interaktion med miljøet. Ved at fokusere på specifikke funktioner i interneurons, der udtrykkes varierende mellem forskellige hjerneregioner, kan man derefter bestemme, hvordan værtsmiljøet ændrer interneuron egenskaber. Neuronal skæbne beslutsomhed og modning kræver et indviklet samspil mellem genetiske programmer og miljømæssige signaler. Udrede roller af iboende vs extrinsic mekanismer, der regulerer denne differentiering proces er dog en gåde for alle udviklingsmæssige neurobiologists.
