Een kritisch aspect van dit protocol is het maximaliseren van het overlevingsvermogen van de cellen. Ervoor te zorgen dat de weefsels en cellen altijd in ijskoude carboxygenated sACSF is noodzakelijk ter bevordering van de overleving van de cel. Dit vereist een efficiënte dissectie en dissociatie strategie om te minimaliseren van de lengte van de tijd dat de cellen in diverse oplossing en buiten de omgeving van de hersenen. Corticale werking vereist een evenwicht tussen excitatory projectie neuronen en remmende GABAergic interneuronen, een uiterst heterogene populatie met verschillende morphologies, elektrofysiologische eigenschappen, connectiviteit en neurochemical markeringen. Abnormale ontwikkeling en functie van interneuronen (en specifieke interneuron subgroepen) is gekoppeld aan de pathobiology van psychiatrische aandoeningen zoals schizofrenie, autisme en epilepsie1,2,3. Bovendien zijn vele genen betrokken bij deze hersenafwijkingen sterk verrijkt met jonge interneuronen4. Dus is een groter inzicht in de mechanismen die interneuron lot vastberadenheid en rijping regelen nodig om te begrijpen van de normale ontwikkeling en potentiële etiologie van talrijke hersenziekten.
Reukkolf interneuronen zijn geboren voornamelijk uit twee voorbijgaande embryonale structuren, de mediale en caudal ganglionaire eminenties (MGE en CGE, respectievelijk). Deze postmitotic cellen (interneuron precursoren) dan het ondergaan van een migratiefase van de langdurige tangentiële te verspreiden in de telencephalon waar ze integreren in een breed scala aan circuits. MGE afkomstige interneuronen bestaan uit drie grotendeels niet-overlappende, neurochemically gedefinieerde subgroepen: snel stekelige parvalbumin (PV+) interneuronen, niet-fast stekelige interneuronen Somatostatine (SST+), en laat stekelige neuronale nitraatstikstof oxide synthase (nNOS+) interneuronen die hippocampal neurogliaform en ivy cellen vormen. Talrijke labs hebben verschillende mechanismen binnen de MGE die eerste lot besluiten in PV+ of SST + interneuronen, met inbegrip van ruimtelijke gradiënten van morphogens, geboortedatum van interneuron precursoren, en de wijze van neurogene divisie regelen vastgesteld 5 , 6 , 7 , 8 , 9 , 10. er is voorgesteld dat interneuronen aanvankelijk in 'kardinaal klassen onderscheiden' en vervolgens geleidelijk in 'definitieve klassen' rijpen als ze met hun omgeving-11 interageren. Recente gegevens blijkt dat sommige volwassen interneuron subtypen genetisch hardwired wellicht deze cellen naarmate postmitotic in de ganglionaire eminenties, die aangeeft dat de vroege gedefinieerde intrinsieke genetische programma's een grotere rol dan voorheen kunnen spelen gewaardeerd12,13. De belangrijke vraag hoe de intrinsieke genetische programma's met milieu aanwijzingen naar station differentiatie in verschillende interneuron subtypen communiceren blijft echter grotendeels onontgonnen.
Talrijke studies hebben getransplanteerde embryonale MGE cellen rechtstreeks in een verscheidenheid van hersengebieden, met de resultaten van de consensus die geënt van cellen volwassen en vrijgeven van GABA te remmen in het algemeen de lokale endogene circuit14,15, 16,17,18,19. Deze waarnemingen veelbelovend hebben gegenereerd significant belang in het gebruik van menselijke geïnduceerde pluripotente stamcel (hIPSC)-afgeleid van interneuronen voor de behandeling van een verscheidenheid van hersenziekten. Nochtans, zeer weinigen van deze studies beoordelen als deze geënte cellen in de verwachte soorten rijpe interneuronen rijpen, een kritische component wanneer men denkt over translationeel benaderingen.
Om aan te pakken hoe milieu invloeden interneuron differentiatie en rijping, werd een strategie ontworpen om de transplantatie van onrijpe interneuron precursoren in nieuwe omgevingen van de hersenen om te onderzoeken of geënte interneuronen kenmerken van de host nemen milieu of functies van de donor milieu20behouden. MGE transplantaties zijn niet geschikt om aan te pakken van deze vraag, omdat de MGE bevat een gemengde bevolking van interneuron en GABAergic projectie cellen dat in talrijke regio's van de hersenen21 dispergeren. Zonder te weten waar deze MGE cellen zou zijn gemigreerd, kan men niet volledig beoordelen hoe deze transplantaties worden beïnvloed door het milieu van de hersenen. Door interneuron precursoren op vroege postnatale timepoints te oogsten, wordt dit probleem omzeild door het verkrijgen van onrijpe cellen die hebben hun migratie voltooid en hun doel bereikt brain regio maar hebben minimale interactie met de omgeving. Door te focussen op specifieke kenmerken van interneuronen die differentieel tussen verschillende hersengebieden worden uitgedrukt, kan men vervolgens bepalen hoe de hostomgeving interneuron eigenschappen verandert. De algemene aanpak in dit protocol dient te gelden tot een onderzoeker dat wil onderzoeken hoe jonge neuronen zich wanneer gedragen uitgedaagd in een nieuwe omgeving.

Vereist een ingewikkelde interactie tussen genetische programma's en Milieusignalen neuronale lot vastberadenheid en rijping. Ontwarren van de rollen van intrinsieke vs. extrinsieke mechanismen die deze differentiatie proces regelen is echter een raadsel voor alle ontwikkelingsstoornissen neurobiologists. Dit probleem is vergroot voor GABAergic interneuronen, een ongelooflijk heterogene celpopulatie dat is geboren uit voorbijgaande embryonale structuren en ondergaan een langdurige trekkende fase te verspreiden in de telencephalon. Om te ontdekken hoe verschillende hersenen omgevingen beïnvloeden interneuron lot en rijping, ontwikkelden we een protocol voor het oogsten van fluorescently geëtiketteerde onvolwassen interneuron precursoren van specifieke hersengebieden bij pasgeboren muizen (P0-P2). Op deze leeftijd, interneuron migratie is bijna voltooid en deze cellen zijn die woonachtig zijn in hun laatste rustplaats omgevingen met relatief weinig synaptic integratie. Na inzameling van eencellige oplossingen via stroom cytometry, deze interneuron precursoren in P0-P2 wildtype postnatale pups worden getransplanteerd. Door het uitvoeren van beide homotoop (bijvoorbeeld cortex-naar-cortex) of heterotopic (bijvoorbeeld cortex-naar-hippocampus) transplantaties, men kan beoordelen hoe het uitdagende onvolwassen interneuronen in nieuwe omgevingen van de hersenen van invloed is op het hun lot, rijping en integratie van het circuit. Hersenen kunnen worden geoogst in volwassen muizen en vehiculumcontrolegroep met een breed scala aan posthoc analyse op geënte cellen, met inbegrip van immunohistochemical, elektrofysiologische en transcriptionele profilering. Deze algemene aanpak biedt onderzoekers met een strategie voor de gehaltebepaling van hoe verschillende omgevingen: hersenen kunnen invloed hebben op tal van aspecten van neuron ontwikkeling en identificeren als neuronale specificiteit voornamelijk door hardwired genetische programma's gedreven worden of de signalen van het milieu.
