En viktig del av denne protokollen er maksimere survivability av cellene. Sikre at vev og celler er alltid i iskalde carboxygenated sACSF er nødvendig for å fremme cellen overlevelse. Riktig kortikale funksjon krever en balanse mellom eksitatoriske projeksjon neurons og hemmende GABAergic interneurons, en svært heterogen befolkning med forskjellige morphologies, elektrofysiologiske egenskaper, tilkobling og nevrokjemiske markører. Unormal utvikling og funksjon av interneurons (og bestemte interneuron undergrupper) har vært knyttet til pathobiology av psykiske lidelser som schizofreni, autisme og epilepsi1,2,3. Videre er mange gener involvert i disse hjernen lidelser sterkt beriket i unge interneurons4. Dermed er større forståelse av mekanismer som regulerer interneuron skjebne besluttsomhet og modning nødvendig å forstå normal utvikling og potensielle etiologies av mange brain sykdommer.
Forebrain interneurons er født hovedsakelig fra to forbigående embryonale strukturer, de mediale og caudal ganglionic eminences (MGE og CGE, henholdsvis). Disse postmitotic cellene (interneuron forløpere) deretter gjennomgå en langvarig tangentiell migrasjon fase å spre gjennom telencephalon der de integrere i en rekke kretser. Mange labs har identifisert flere mekanismer i MGE som regulerer første skjebne beslutninger i PV+ eller SST + interneurons, inkludert romlige graderinger av morphogens, fødselsdato av interneuron prekursorer og modus for neurogenic divisjon 5 , 6 , 7 , 8 , 9 , 10. det har blitt foreslått at interneurons først skille ut "kardinal klasser" og deretter gradvis moden til "definitive klasser" når de samhandler med deres miljø11. Nyere bevis indikerer at noen eldre interneuron undertyper kan Hardwired genetisk som disse cellene blir postmitotic i de ganglionic eminences, som indikerer at tidlig definerte iboende genetiske programmer kan spille en større rolle enn tidligere verdsatt12,13. Tallrike studier har transplantert embryonale MGE celler direkte inn i forskjellige områder av hjernen, med konsensus resultater som podet celler modne og slipp GABA å hemme generelt lokale endogene krets14,15, 16,17,18,19. Dette lovende observasjoner har generert betydelig interesse i å bruke menneskelige indusert pluripotent stilk cellen (hIPSC)-avledet interneurons for å behandle en rekke brain sykdommer. Men tenker svært få av disse studiene vurdere om disse podet cellene eldre inn i forventet typer av eldre interneurons, en kritisk komponent når man på translasjonsforskning tilnærminger.
For å løse hvordan miljø påvirker interneuron differensiering og modning, var en strategi konstruert for å transplantere umodne interneuron forløpere til nye hjernen miljøer for å undersøke om podet interneurons vedta funksjoner av verten miljø eller beholde funksjoner fra donor miljø20. MGE transplantasjoner er ikke egnet til å løse dette spørsmålet fordi MGE inneholder en blandet befolkning av interneuron og GABAergic projeksjon celler som spre seg gjennom mange hjernen regioner21. Uten å vite hvor disse MGE cellene ville har migrert, kan en ikke fullt ut vurdere hvordan disse transplantasjoner påvirkes av hjernen miljøet. Ved å høste interneuron forløpere på tidlig postnatal timepoints, problemet er circumvented ved å skaffe umodne celler som har fullført sine og nådde målet hjernen regionen, men har minimal interaksjon med miljøet. Ved å fokusere på bestemte funksjoner i interneurons som er ulikt uttrykt mellom forskjellige hjernen, kan en så bestemme hvordan vertsmiljøet endrer interneuron egenskaper. Neuronal skjebne besluttsomhet og modning krever en intrikat samspill mellom genetiske programmer og Miljøsignaler. Men er disentangling rollene iboende vs ytre mekanismer som regulerer differensiering prosessen en gåte for alle utviklingsmessige neurobiologists.
