Función cortical adecuada requiere un equilibrio entre las neuronas excitatory de la proyección e inhibitorios interneurons de GABAérgico, una población muy heterogénea con morfologías distintas, propiedades electrofisiológicas, conectividad y neuroquímicos marcadores. Función de interneuronas (y subgrupos específicos interneurona) y desarrollo anormal se ha relacionado con la Patobiología de trastornos psiquiátricos como la esquizofrenia, autismo y epilepsia1,2,3. Además, muchos genes implicados en estos trastornos cerebrales se enriquecen fuertemente en interneuronas joven4. Por lo tanto, una mayor comprensión de los mecanismos que regulan la maduración y la determinación del destino de interneurona es necesario para comprender el desarrollo normal y las posibles etiologías de numerosas enfermedades del cerebro.
Interneuronas de forebrain nacen principalmente de dos estructuras embrionarias transitorias, las eminencias ganglionares mediales y caudales (MGE y CGE, respectivamente). Estas células postmitotic (precursores de la interneurona) luego pasar por una fase prolongada migración tangencial para dispersar en el telencéfalo donde se integran en una gran variedad de circuitos. La evidencia reciente indica que algunos subtipos de interneurona maduro pueden ser genéticamente cableado como estas células se convierten en postmitotic en las eminencias ganglionares, indicando que programas genéticos intrínsecos definidos principios pueden desempeñar un papel más grande que antes apreciada12,13. Numerosos estudios han trasplantado células embrionarias MGE directamente en una variedad de regiones del cerebro, con los resultados del consenso que injertaron células maduras y liberan GABA para inhibir generalmente los circuitos endógeno local14,15, 16,17,18,19. Estos prometedores observaciones han generado un interés significativo en el uso de células pluripotentes inducidas humanas (hIPSC)-derivado de interneuronas para tratar una variedad de enfermedades del cerebro. Sin embargo, muy pocos de estos estudios determinar si estas células injertadas maduran en los tipos esperados de interneuronas maduras, un componente esencial cuando uno piensa en enfoques traslacionales.
Para abordar cómo el entorno influye interneurona diferenciación y maduración, una estrategia fue ideada para trasplante precursores inmaduros interneurona en nuevos entornos de cerebro para examinar si interneuronas injertados adoptan características del anfitrión medio ambiente o conservar características de las donantes medio ambiente20. MGE trasplantes no son adecuados para abordar esta cuestión porque el MGE contiene una población mixta de células de proyección de GABAérgico que dispersan a lo largo de numerosas regiones de cerebro21e interneurona. Sin saber donde estas células MGE habría emigrado, uno puede no evaluar completamente cómo estos trasplantes son afectadas por el entorno del cerebro. Maduración y la determinación de destino neuronal requiere una interacción intrincada entre señales ambientales y programas genéticos. Sin embargo, desenmarañar los papeles de intrínseca vs extrínsecos mecanismos que regulan este proceso de diferenciación es un enigma para los neurobiólogos del desarrollo.
