Koronar hjertesygdom er den hyppigste årsag til dødsfald på verdensplan. Behandling af valg er for at reducere iskæmisk skade og begrænse infarkt størrelse hos patienter med ST-segment forhøjet myokardieinfarkt rettidig og effektiv Myokardie reperfusion via primær perkutan koronar intervention (PCI)1, 2. Dog forårsager reperfusion yderligere skader, som kan tegne sig for op til 30 procent af den afsluttende infarkt størrelse3. Det er almindeligt anerkendt, at mitokondriel permeabilitet overgangen pore (mPTP) ikke er kun centrale i mitokondrie skader og celle død under iskæmi/reperfusion (jeg / R), men er også et konvergerende mål af cardioprotective signalering4 , 5. som mPTP åbningen vil medføre depolarisering af den indre mitokondrielle membran potentiale (MMP)4, vi opdaget mPTP åbning ved hjælp af 5, 5, 6, 6 '-Tetrachloro-1, 1′, 3, 3 '-tetraethyl-imidacarbocyanineiodide (JC-1) assay.
JC-1 analysen er en cytofluorimetric metode, der er både kvalitative og kvantitative, og yderligere er valideret ved at analysere MMP på niveau med en enkelt mitokondrier6. JC-1 eksisterer som en aggregeret form, giver en rød til orange farvede emission (590 ± 17,5 nm) i matrixen af mitochondrier med normal MMP; med tabet af MMP, er JC-1 konverteret til monomere form, der giver grønne fluorescens med en emission af 530 ± 15 nm. Derfor, et fald i rød/grøn fluorescens intensitet forholdet kunne angive reduktion af MMP i forhold såsom iskæmi/reperfusion (jeg / R).
Ud over JC-1, er MMP også blevet undersøgt med membran-gennemtrængelige lipofile kationer såsom rodamin 123 og 3, 3 '-dihexyloxadicarbocyanine Iodid [DiOC6(3)]. Men i forhold til disse to sonder, JC-1 er mere pålidelige til at analysere MMP. Rodamin 123 har relativt ringe følsomhed (især i dæmper tilstand7,8) og fattige specificitet. Skift i rodamin 123 er undertiden så lille, at det er svært for forskere eller udstyr til at observere/opdage. Desuden, i en enkelt celle, der er forskellige mitochondriet bindingssteder for rodamin 123 og så at det kan have forskellige fluorescens emissioner9. DiOC6(3) er ikke anbefalet til påvisning af MMP enten som det reagerer følsomt på depolarisering af plasma membran10.

Rettidig og effektiv reperfusion af tilstoppet koronararterie er den bedste strategi for faldende Myokardie infarkt størrelse hos patienter med en ST-segment forhøjet myokardieinfarkt.
