Coronaire hartziekte is de belangrijkste oorzaak van overlijden wereldwijd. De behandeling van keuze is voor vermindering van ischemische schade en de beperking van de grootte van het infarct in patiënten met ST-segment myocardiaal infarct verheven tijdige en effectieve myocardiale reperfusie via primaire percutane coronaire interventie (PCI)1, 2. Reperfusie veroorzaakt echter bijkomende schade, die kan goed zijn voor maximaal 30% van het laatste infarct grootte3. Het is algemeen erkend dat de mitochondriale permeabiliteit overgang porie (mPTP) niet alleen centraal in mitochondriale schade en cel dood tijdens ischemie/reperfusie is (Ik / R), maar is ook een convergerende doel van bescherming signalering4 , 5. als de mPTP-opening over depolarisatie van de innerlijke mitochondriale membraan potentiële (MMP)4 brengen zou, we ontdekt mPTP openen met behulp van de 5, 5 ', 6, 6 '-Tetrachloro-1, 1, 3, 3 '-tetraethyl-imidacarbocyanineiodide (JC-1) assay.
De bepaling van de JC-1 is een cytofluorimetric methode die zowel kwalitatief als kwantitatief, en het verder is gevalideerd door het analyseren van de MMP op het niveau van een enkele mitochondriën6. JC-1 bestaat als een geaggregeerde vorm, de opbrengst van een rood tot oranje gekleurde emissie (590 ± 17,5 nm) in de matrix van mitochondriën met normale MMP; met het verlies van MMP, wordt JC-1 geconverteerd naar de monomere vorm die groene fluorescentie met een uitstoot van 530 ± 15 nm levert. Dus een daling in de verhouding rood/groen fluorescentie intensiteit kan duiden op de verlaging van MMP in omstandigheden zoals ischemie/reperfusie (Ik / R).
Naast JC-1, is MMP ook bestudeerd met membraan-permeabele lipofiele kationen zoals Rhodamine 123 en 3, 3 '-dihexyloxadicarbocyanine jodide [DiOC6(3)]. Echter, vergeleken met deze twee sondes, JC-1 is meer betrouwbaar voor het analyseren van MMP. Rhodamine 123 heeft relatief arme gevoeligheid (vooral in het blussen van mode7,8) en slechte specificiteit. De verschuiving in rhodamine 123 is soms zo klein dat het is moeilijk voor onderzoekers of apparatuur om te observeren/detecteren. Bovendien, in een enkele cel, er zijn verschillende mitochondrion bandplaatsen voor rhodamine 123 en dus dat het verschillende fluorescentie emissies9kan hebben. DiOC6(3) wordt niet aanbevolen voor het opsporen van MMP hetzij als het gevoelig op de depolarisatie van plasmamembraan10 reageert.
Daarom hier gebruiken we de JC-1-test om te beoordelen de MMP van HCMs na wordt blootgesteld aan hypoxie/heroxygenatie met of zonder een beschermende agent.

Tijdige en efficiënte reperfusie van de afgesloten kransslagader is de beste strategie voor het verkleinen van het myocard infarct bij patiënten met een ST-segment verheven myocardiaal infarct. Echter kan reperfusie per se resulteren in verdere cardiomyocyte dood, een fenomeen dat bekend staat als reperfusie letsel. De opening van de mitochondriale permeabiliteit overgang porie (mPTP), met een daling van de Mitochondriale membraanpotentiaal (MMP) of een mitochondriale depolarisatie, is algemeen erkend als de laatste stap van reperfusie letsel en is verantwoordelijk voor mitochondriale en dood van de cardiomyocyte. JC-1 is een lipofiele kationogene kleurstof dat zich in de mitochondriën afhankelijk van de waarde van MMP ophoopt. Hoe hoger de MMP is, hoe meer JC-1 hoopt zich op in de mitochondriën. De stijgende hoeveelheden voor JC-1 in de mitochondriën kunnen worden weergegeven door een verschuiving van de emissie fluorescentie van groen (~ 530 nm) tot rood (~ 590 nm).
