Maladie coronarienne est la principale cause de décès dans le monde. Le traitement de choix pour réduire une lésion ischémique et de limiter la taille de l’infarctus chez les patients avec segment ST élevé l’infarctus du myocarde est une reperfusion myocardique en temps opportun et efficace via une intervention coronarienne percutanée primaire (PCI)1, 2. Toutefois, la reperfusion provoque des dégâts supplémentaires, ce qui peuvent représenter jusqu'à 30 pour cent de la taille finale infarctus3. Il est universellement reconnu que le pore de transition de perméabilité mitochondriale (mPTP) n’est pas seulement central dans les mitochondries des dommages et la mort cellulaire durant l’ischémie/reperfusion (I / R), mais est aussi une cible convergente de cardioprotecteurs signalisation4 , 5. comme l’ouverture du mPTP entraînerait une dépolarisation de la membrane mitochondriale interne potentielle (MMP)4, nous avons détecté des PFMP ouverture utilisant le 5, 5′, 6, 6′-tétrachloro-1, 1′, 3, test de tétraéthyle-3′-imidacarbocyanineiodide (JC-1).
L’analyse de JC-1 est une méthode de cytofluorométrique qui est tant qualitatives que quantitatives, et elle a été validée plus loin en analysant les MMP au niveau d’une mitochondrie unique6. JC-1 existe sous une forme agrégée, ce qui donne un rouge aux émissions couleur orange (590 ± 17,5 nm) dans la matrice des mitochondries avec MMP normale ; avec la perte de MMP, JC-1 est converti en la forme monomère qui donne une fluorescence verte avec une émission de 530 nm ± 15. Par conséquent, une diminution du ratio d’intensité de fluorescence rouge/vert pourrait indiquer la réduction de MMP dans des conditions telles que l’ischémie/reperfusion (I / R).
En plus de JC-1, MMP a également été étudié avec membrane perméable cations lipophiles comme la Rhodamine 123 et 3, l’iodure de 3′-dihexyloxadicarbocyanine [DiOC6(3)]. Toutefois, par rapport à ces deux sondes, JC-1 est plus fiable pour l’analyse des MMP. La rhodamine 123 a une sensibilité relativement médiocre (surtout en trempe mode7,,8) et la spécificité médiocre. Le changement dans la rhodamine 123 est parfois si petit qu’il est difficile pour les chercheurs ou les équipements à observer/détecter. En outre, dans une seule cellule, il existe des sites de liaison différents mitochondrie pour la rhodamine 123 et donc qu’elle peut avoir différentes fluorescence émissions9. DiOC6(3) n’est pas recommandé pour la détection des MMP soit comme il réagit sensiblement à la dépolarisation de la membrane plasmique10.
Donc, ici, nous utilisons le test JC-1 pour évaluer la MMP de ces mesures après avoir été exposés à une hypoxie/réoxygénation avec ou sans agent de protection.

Méthode rapide et efficace de reperfusion de l’artère coronaire obstruée, c’est la meilleure stratégie pour diminuer la taille de l’infarctus du myocarde chez les patients avec un segment ST élevée d’infarctus du myocarde. Toutefois, reperfusion en soi peut entraîner en outre du cardiomyocyte, un phénomène appelé lésion de reperfusion. L’ouverture de la pore de transition de perméabilité mitochondriale (mPTP), avec la diminution du potentiel membranaire mitochondrial (MMP), ou la dépolarisation mitochondriale, est universellement reconnue comme la dernière étape de la lésion de reperfusion et est responsable mitochondriale et du cardiomyocyte. JC-1 est un colorant cationique lipophile qui s’accumule dans les mitochondries selon la valeur de MMP. Plus le PSM est, plus JC-1 s’accumule dans les mitochondries. Les quantités croissantes de JC-1 dans les mitochondries peuvent se traduire par un changement de d’émission de fluorescence du vert (~ 530 nm) au rouge (~ 590 nm). Par conséquent, la réduction du ratio d’intensité de la fluorescence rouge/vert peut indiquer la dépolarisation des mitochondries.
