Koronarer Herzkrankheit ist die häufigste Todesursache weltweit. Die Therapie der Wahl ist für ischämische Schädigung Reduzierung und Begrenzung Infarkt Größe bei Patienten mit ST-Segment Myokardinfarkt erhöhten rechtzeitige und wirksame myokardialen Reperfusion über primäre perkutane Koronarintervention (PCI)1, 2. Reperfusion verursacht zusätzlichen Schaden, die bis zu 30 Prozent des letzten Infarkt Größe3ausmachen kann. Es ist allgemein anerkannt, dass die mitochondrialen Permeabilität Übergang Pore (mPTP) nicht nur zentral in mitochondrialen Schäden und Zelle Tod während Ischämie/Reperfusion (ich / R), sondern ist auch ein konvergierende Ziel kardioprotektive Signalisierung4 , 5. mPTP Eröffnung Depolarisation der inneren mitochondrialen Membran potenzielle (MMP)4bringen würde, wir haben festgestellt, mPTP Eröffnung mit 5, 5 ', 6, 6 '-Tetrachloro-1, 1, 3, 3′-Tetraethylblei-Imidacarbocyanineiodide (JC-1) Assay.
Die JC-1-Assay ist eine Cytofluorimetric Methode, die sowohl quantitative als auch qualitative, und es wurde weiter durch die Analyse der MMP auf der Ebene eines einzelnen Mitochondrien6validiert. JC-1 existiert als aggregierter Form, eine rote bis orange farbigen Emission (590 ± 17,5 nm) in der Matrix der Mitochondrien mit normalen MMP nachgeben; mit dem Verlust von MMP ist JC-1 in monomerer Form konvertiert, die grünen Fluoreszenz mit einer Emission von 530 nm ± 15 ergibt. Daher deutet eine Abnahme in der rot/grünen Fluoreszenz Intensitätsverhältnis den Rückgang von MMP Bedingungen wie Ischämie/Reperfusion (ich / R).
MMP ist neben JC-1 auch mit Membran durchlässig lipophilen kationen wie Rhodamin 123 und 3, 3′-Dihexyloxadicarbocyanine Jodid [DiOC6(3)] untersucht worden. JC-1 ist jedoch im Vergleich zu diesen zwei Sonden, zuverlässiger für die Analyse von MMP. Rhodamin 123 hat relativ schlechte Sensitivität (insbesondere Quench Modus7,8) und schlechte Spezifität. Die Verschiebung der Rhodamin 123 ist manchmal so klein, dass es schwer für Forscher oder Ausrüstung zu beobachten/zu erkennen. Außerdem in einer einzelnen Zelle, gibt es verschiedene Mitochondrium Bindungsstellen für Rhodamin 123 und so, daß es eventuell unterschiedliche Fluoreszenz Emissionen9. DiOC6(3) wird nicht empfohlen für die Erkennung von MMP entweder so, wie es sensibel auf die Depolarisation der Plasmamembran10 reagiert.
Deshalb verwenden wir hier die JC-1-Assay MMP HCMs bewerten nach Hypoxie/flussbettfilters mit oder ohne einem Schutzmittel ausgesetzt wird.

Zeitnahe und effiziente Reperfusion der verschlossenen Koronararterie ist die beste Strategie zur Verringerung der Größe der myokardialen Infarkt bei Patienten mit ST-Segment Herzinfarkt erhöht. Reperfusion schlechthin kann jedoch im weiteren Cardiomyocyte Tod, ein Phänomen bekannt als Reperfusion Verletzungen führen. Die Eröffnung der mitochondrialen Permeabilität Übergang Pore (mPTP), mit der Abnahme des mitochondrialen Membranpotentials (MMP) oder mitochondriale Depolarisation ist allgemein anerkannt als der letzte Schritt der Reperfusion Verletzungen und ist verantwortlich für Mitochondriale und Cardiomyocyte Tod. JC-1 ist eine lipophile kationischen Farbstoff, der in den Mitochondrien abhängig vom Wert des MMP ansammelt. Je höher die MMP ist, desto mehr JC-1 sammelt sich in den Mitochondrien. Die zunehmende Mengen von JC-1 in den Mitochondrien können durch eine Fluoreszenz Emission Verlagerung von Grün zum Ausdruck kommen (~ 530 nm) bis rot (~ 590 nm). Daher kann die Verringerung der rot/grünen Fluoreszenz Intensitätsverhältnis die Depolarisation der Mitochondrien angegeben. Hier nehmen wir Vorteil von JC-1 MMP Messen oder der Eröffnung des mPTP in menschlichen kardialen Myozyten nach Hypoxie/flussbettfilters, durch Durchflusszytometrie nachgewiesen.

Vor der Durchführung des JC-1-Tests um die Veränderungen der MMP zu bewerten, es empfiehlt Versuche durchgeführt werden, bestätigen die Bedingungen erfolgreich von den Forschern festgelegt.
