Malaria er en ødelæggende sygdom forårsaget af protozo parasitter i slægten Plasmodium. P. falciparum, den mest dødbringende menneskelige malaria parasitten, der forårsager ca 445,000 dødsfald om året, for det meste i børn under en alder af fem1. Plasmodium parasitter har en indviklet livscyklus, der involverer en myg vektor og et hvirveldyr vært. Mennesker bliver først smittet, når en inficeret myg tager en blodmel. Derefter, parasitten invaderer leveren hvor det vokser, udvikler og deler sig for ca. en uge. Her præsenterer vi en protokol for generation af betingede knockdown mutanter i P. falciparum bruger CRISPR/Cas9 genom-redigering. Protokollen viser brugen af glmS ribozym til betinget knockdown protein niveauer af PfHsp70x (PF3D7_0831700), en anstandsdame eksporteret af P. falciparum i vært RBC13,14. GlmS ribozym aktiveres ved behandling med glucosamin (som konverteres til glucosamin-6-fosfat i celler) for at kløve dens tilknyttede mRNA, fører til en reduktion i protein14. Denne protokol kan tilpasses nemt for at udnytte andre betinget knockdown værktøjer, såsom destabilisering domæner eller RNA aptamers4,5,15. Vores protokoloplysninger generation af en reparation plasmid bestående af hemagglutinin (HA) tag og glmS ribozymer kodende sekvens flankeret af sekvenser der er homologe til den PfHsp70x åben behandling ramme (ORF) og 3'-UTR. crispr/cas9 gene editing to make conditional mutants of human malaria parasite p. falciparum

Malaria er en væsentlig årsag til sygelighed og dødelighed på verdensplan. Denne sygdom, der primært påvirker dem, der lever i tropiske og subtropiske regioner, er forårsaget af infektion med Plasmodium parasitter. Udviklingen af mere effektive lægemidler til at bekæmpe malaria kan fremskyndes ved at forbedre vores forståelse af biologi af denne komplekse parasit.
