1. 蚊虫幼虫和成人的养殖

</ol>2. 幼虫接触检测 (单点剂量或剂量响应检测)

</ol>3. 成人局部检测 (单点剂量或剂量反应检测)

	<li>清除1毫升的丙酮玻璃注射器, 用丙酮填充, 并固定在调整后的微涂抹器中, 以提供0.25μl 的体积。</li>
	<li>成批工作的 10, 使用吸入器将3-5天的成年雌性蚊子从笼子里取出, 并在冰箱或冰上的4°c 下麻醉5分钟。接下来, 将蚊子转移到培养皿中, 放在冰上长达 1 0分钟。</li>

	<li>将蚊子转移到贴有标签的纸杯上, 在冰上休息, 并重复 9次, 以获得 n = 10只经过处理的蚊子。用橡皮筋固定的网状方块密封杯子中的蚊子, 并在28°c 和75-85 的恒温下转移到塑料桶或生长室 (在12小时的夜间周期内)。</li>
	<li>重复上述实验两次, 以获得 n = 3个技术复制 (即, 总共30个蚊子) 每个治疗或控制组。</li>

</ol>4. 成人送血试验 (单点剂量或剂量反应测定)
<ol><li>收集大约 150个4-5 的成年雌性蚊子, 并转移到一个单独的笼子。如果进行剂量反应分析, 重复使用6个网箱。在喂养试验前1-24 去除糖的来源。</li>
	<li>按上述方式制备化学库存溶液, 说明化学的纯度 (典型的库存溶液在水或盐缓冲液中为 80 mm), 然后根据需要在水缓冲液中制备连续稀释剂。</li>
	<li>在960μl 的脱颤兔血液中加入 40μl, 以获得所需的检测浓度。将膜膜涂在进料装置上, 并用橡胶圈密封。通过移液器输送1毫升血液, 并将喂养装置连接到加热装置。</li>
	<li>用新鲜制作的10% 乳酸溶液轻轻擦拭膜表面, 并将喂养单元放入成年蚊子的笼子中。用黑布或黑色垃圾袋覆盖笼子, 让蚊子喂一个小时。</li>
	<li>对测试解决方案的其余部分和阴性 (仅限血液) 控制重复步骤 4.2-4.4。</li>
