1. 组织准备和贮存

Recellularization 生物反应器

</ol>3. 解决方案的准备工作

</ol>4. 内皮介质的制备
<ol><li>添加5毫升的 l-谷氨酰胺, 5 毫升抗反, 50 毫升的人 AB 血清和所有成分的 EGM-2 生长因子套件, 除胎牛血清到500毫升的 MCDB131 培养基下的无菌罩。</li>
</ol>5. 隐静脉去细胞
<ol><li>在室温下将人隐静脉从-80 摄氏度解冻。再次冻结在-80 摄氏度和解冻再次在室温下。</li>

</ol>6. 去细胞的核查
<ol><li>用超纯水清洗福尔马林固定活检15分钟, 在组织处理器中进行标准协议, 并嵌入石蜡22。</li>
	<li>5µm 部分使用切片和染色与迈耶的苏木精和0.2% 酒精的伊红 (H & E) 遵循标准协议22。</li>
	<li>在显微镜下查看与正常组织相比, 瓣膜组织中核的损失。</li>
</ol>7. Recellularization
<ol><li>将静脉放在含有50毫升溶液8的50毫升管中杀菌。搅拌在80旋转每分钟 (rpm) 在振动筛为1小时37摄氏度。</li>
	<li>从现在起, 处理层流 (黎巴嫩) 内阁下的静脉以维持不孕。用无菌钳将静脉转移到新的50毫升管中, 含有50毫升的无菌溶液7和500µL 抗反。为12小时以上的搅动, 改变解决方案7至少两次之间。</li>

	<li>使用无菌手套在黎巴嫩武装部队内阁中组装生物反应器。用无菌缝合和镊子将静脉栓在生物反应器的静脉入口和出口处。</li>
	<li>将静脉连接到生物反应器, 并用相同的培养基填充。添加肝素在50毫升/毫升和灌注1小时在 2 mL/分钟37摄氏度。</li>

decellularization and recellularization methodology for human saphenous veins
