Ici, nous avons décrit à oSLO, une en vivo volumétrique fluorescent d’imagerie rétinienne technique avec un FOV de plus de 30 °. Par rapport à octobre, un standard actuel méthode en ophtalmologie, d’imagerie oSLO offre un capacité d’imagerie en 3D similaires mais permet de contraste de fluorescence qui OCT n’est pas sensible aux. L’avantage d’oSLO est qu’elle nécessite qu’une seule méthode raster et permet ainsi la combinaison transparente de PTOM, prévoyant deux techniques complément...

Ophtalmologie et vision science bénéficier grandement les techniques modernes d’imagerie optiques, étant donné que la rétine est facilement accessible avec la lumière. Imagerie rétinienne de fluorescence est un outil essentiel dans le diagnostic et la gestion de Chorio-rétinienne de maladies vasculaires comme la rétinopathie diabétique (RD) et de la dégénérescence maculaire liée à l’âge (DMLA), qui sont causes principales de cécité aux Etats-Unis.
Toutefois, il est toujours difficile d’acquérir un grand champ de vision (FOV), en trois dimensions rétinienne (3D), imagerie par imagerie de fluorescence. La photographie du fond de œil n’a pas la capacité de résolution de profondeur et ne rejette pas la lumière diffuse. Ainsi, le mélange des signaux provenant de différente profondeur réduit la qualité de l’image. Balayage ophtalmoscopie laser (SLO) et confocale SLO (cSLO) ne peut réduire l’effet de lumière diffuse à l’aide de gating confocal1. Toutefois, il est difficile pour SLO ou cSLO d’acquérir une image 3D de rétine humaine en raison de la limite de leur profondeur de champ. Optique adaptative SLO (AOSLO) permettent une superbe résolution et un contraste en corrigeant les aberrations de wavefront introduite par le œil humain. Toutefois, AOSLO aurait encore besoin z d’empilage pour imagerie volumétrique2. Systèmes de l’angiographie (OCTA) de OCT et de tomographie par cohérence optique3 surmonter ces restrictions afin de fournir en trois dimensions (3D) des images anatomiques et vasculaire4,5,6, mais la nature sans colorant d’OCT ne peut pas visualiser fuite indicative de dysfonction vasculaire.
Ce protocole décrit une nouvelle plate-forme multimodale pour rétinienne de fluorescence volumétrique 3D imaging, nommément oblique de balayage laser ophtalmologique (oSLO). Dans ce système d’imagerie, une oblique de la numérisation est générée par un curseur de queue de colombe, et un système d’imagerie final est aligné dans un angle pour détecter la fluorescence Croix images sectionnelles. Le système utilise des méthodes de balayage laser, et ces techniques permettent une intégration facile avec OCT comme volumétrique modalité d’imagerie structurelle complémentaire. La résolution actuelle de profondeur est d’environ 25 µm dans la rétine de rat et le champ de vision est de 30°. Essentiellement, l’oSLO permet une version fluorescente de OCT et peut être associé simultanément OCT et OCTA sur un grand champ de vision.
Dans ce protocole, nous allons décrire l’installation de l’oSLO, la méthode d’alignement et de la construction, la méthode de l’imagerie in vivo de la rétine de rat et les résultats représentatifs.

Alors que l’imagerie de fluorescence est largement utilisée en ophtalmologie, une image rétinienne en trois dimensions (3D) fluorescence du grand champ de vision (FOV) est toujours un grand défi avec la rétine state-of-the-art modalités d’imagerie car il leur faudrait z un empilement de Compiler un ensemble de données volumétrique. Ce protocole décrit une oblique roman analyse technique de laser ophtalmologique (oSLO) qui fournit l’imagerie rétinienne fluorescence volumétrique 3D. Le programme d’installation du système d’imagerie génère l’oblique le balayage par un curseur de queue de colombe et aligne le système d’imagerie final à un angle pour détecter les images fluorescentes en coupe transversale. Le système utilise la méthode de balayage au laser et par conséquent, permet une intégration facile des OCT comme volumétrique modalité d’imagerie structurelle complémentaire. L’imagerie in vivo sur la rétine de rat est démontré ici. Solution de fluorescéine est injectée par voie intraveineuse pour produire l’angiographie à la fluorescéine volumétrique (vFA).

Figure 4 a montre une image de OCT transversale de la rétine de rat. Figure 4 b -4 c montrent les mêmes images rétiniennes transversales de vFA OCTA et oSLO acquis en même temps. L’oSLO permet transversale FA analogue à l’OCT B-scan. En comparaison de l’OCTA, l’image coupe transversale d’oSLO vFA identifie clairement les vaisseaux dans la couche de fibres nerveuses (NFL) et de la couche de cellules...

Nous présentons ici un protocole pour obtenir un grand champ de vision (FOV) en trois dimensions (3D) fluorescence et l’image rétinienne OCT en utilisant une nouvelle plate-forme multimodale d’imagerie.
