Deze studie beschreven een stapsgewijze proeflezen activiteit assay geanalyseerd door de gekozen commerciële instrument (Zie de Tabel van materialen) met behulp van MALDI-TOF MS. De grote voordelen zijn dat de primer en de sjabloon label gratis en eenvoudig uit te voeren zijn, meer flexibiliteit bij het ontwerpen van experimenten. Een stream-kalk volledige verwerking van 30 proeflezen proeven zou duren 4 h, met inbegrip van 3 h voor het handmatig uitvoeren van de proeflezen reacties en hun opruimen, ter...

De proeflezen functies van de polymerase van DNA tijdens de DNA-replicatie zijn van essentieel belang om ervoor te zorgen de hifi van genetische informatie die moet worden overgedragen aan nakomelingen1,2,3,4, 5,6,7. Zijnde kundig voor het beoordelen van de bijdragen van polymerase proeflezen van exonucleases zou het verduidelijken van de mechanismen om genetische stabiliteit te waarborgen.
Isotoop labeling en gel gebaseerde testen in combinatie met densitometric analyses van autoradiograms of fosfor imaging8,9,10 zijn traditioneel gebruikt voor het detecteren van proeflezen activiteit van DNA polymerase. Hoewel functioneel, zijn deze tests arbeidsintensief, duur en niet vatbaar voor high-throughput formaten. Daarnaast lijden de radio-isotopen veiligheidskwesties met inbegrip van de verwijdering van afval. Anderzijds hebben proeflezen activiteiten zijn geanalyseerd door fluorimetrische technieken. 2-aminopurine (2-AP) kan bijvoorbeeld worden opgenomen in extensie producten tijdens in vitro polymerase proeflezen testen om te produceren een fluorescent signaal11,12. Helaas, deze benaderingen lijden onder een lage specificiteit, aangezien 2-AP met zowel thymine en cytosine koppelen kunt. Meer recente benaderingen omvatten een gevoelige G quadruplex-gebaseerde verlichte schakelaar-op sonde voor een polymerase van 3' - 5' exonuclease assay13 , alsmede een afzonderlijk-label fluorescente sonde voor een polymerase van proeflezen assay overwint enkele van de bovengenoemde nadelen14. Enthousiasme voor deze fluorimetrische methoden is verminderd als gevolg van de noodzaak van de specifieke etikettering van DNA substraten.
In tegenstelling, een MALDI-TOF MS voor DNA-analyse is werkzaam in de PinPoint-assay, waar de primer extensie reacties met labelloze 4 ddNTPs kunnen worden gebruikt voor het identificeren van polymorfismen bij een bepaald locus15,16,17 en grote schaal is goedgekeurd in klinische toepassingen voor mutatie detecties en kanker diagnoses18. Met behulp van deze fundamentele beginselen, hebben we een label-gratis test voor de bepaling in vitro van DNA-polymerase activiteit benutting van de hoge resolutie, hoge specificiteit en high-throughput potentieel van MALDI-TOF MS. gebruiken de E. proeflezen coli polymerase van DNA ik Klenow als een model-enzym, dideoxyribonucleotide trifosfaat (ddNTPs fragment) als substraten een "momentopname nemen kunnen" van proeflezen producten na een single nucleotide uitbreiding via MALDI-TOF MS (Figuur 1).
Ook werd deze methode ook ontwikkeld voor een DNA repair assay waar inleidingen met 3' voorlaatste dI laesies zijn onderworpen aan een pol die ik herstellen assay welke nabootsers endo V gejat reparatie tussenproducten. Hoewel niet volledig begrepen, is de endo V reparatie traject het enige reparatie systeem bekend om de pol in dienst ik proeflezen exonucleaseactiviteit voor laesie excisie19,20. Met behulp van MALDI-TOF MS, laten we zien een duidelijk omschreven reparatie patch waar dI kan worden weggesneden door pol ik wanneer die zich voordoen in de laatste 2 nt van de primer voordat u toevoegt de juiste aangevuld nucleotide.
Voor de studie van proeflezen en DNA-herstel, deze methode is sneller en minder arbeidsintensief dan eerdere methoden en biedt extra informatie naar mechanisme en functie.

De handhaving van het genoom en de trouwe replicatie is essentieel voor het behoud van de genetische informatie. Om te beoordelen HiFi replicatie, hebben we een eenvoudige niet-gelabeld en niet-radio-isotopische methode met behulp van een laser matrix-bijgewoonde desorptie ionisatie met time-of-flight (MALDI-TOF) massa spectrometrie (MS) analyse voor een proeflezen studie. Hier, een polymerase van DNA [bijvoorbeeld het Klenow fragment (KF) van de polymerase van DNA van Escherichia coli ik (pol ik) in deze studie] in het bijzijn van alle vier dideoxyribonucleotide trifosfaat is gebruikt voor het verwerken van een niet-overeenkomende primer-template-duplex. De niet-overeenkomende primer is vervolgens proeflezen en verruimde en onderworpen aan de MALDI-TOF MS. De producten worden gekenmerkt door de massale verandering van de primer tot één nucleotide variaties. Nog belangrijker is, kan een proeflezen ook worden bepaald voor interne enkele problemen, zij het op verschillende efficiëntie.
