Hier presenteren we twee verschillende protocollen cultuur primaire cellen uit 2 dpf zebrafish embryo's of volwassen zebrafish hersenen.
De bereiding van primaire celculturen van 2 dpf zebrafish is relatief eenvoudig voor iemand met ervaring in fundamentele cel cultuur technieken uit te voeren. Echter, met het oog op een goede en reproduceerbare resultaten, een voldoende aantal embryo's als grondstof is belangrijk (100 is het minimum). De zebrafish (Danio rerio, D. rerio) is een populair model gewervelde voor tal van gebieden van fundamenteel en biomedisch onderzoek1. Zebravis embryo's ontwikkelen snel ex utero, transparant, en fit onder een Microscoop, waardoor uitstekende voorwaarden voor het bestuderen van gewervelde ontwikkeling in een levend organisme. Als gevolg van de genetische werkwillig van zebravis2, zijn veel stabiele transgene verslaggever regels met cell type-specifieke uitdrukking van verschillende fluorescente markeringen vastgesteld rekening houdend met de waarneming van specifieke cel populaties. De zebravis Gemeenschap biedt een breed scala van zogenaamde Gal4-driver regels waaraan een uiting van de synthetische Kal4TA4 (of de GalFF van het KalTA3-equivalent) transgenic gen met het Gal4-DNA-bindende domein van gist gesmolten virale transcriptionele activering domeinen onder de controle van cel type-specifieke versterkers. Deze bestuurder lijnen zijn overgestoken naar effector lijnen waaraan transgenen dat bestaat uit een gedefinieerde upstream activerend reeks (UAS) gesmolten tot een verslaggever-gen. De Kal4TA4 proteïne bindt aan de UAS-element, dus het activeren van de cel type-selectieve uitdrukking van de verslaggever gen3,4. Deze aanpak zorgt voor zeer uiteenlopende combinatorische studies van bijna alle beschikbare enhancer en verslaggever elementen in dubbel-transgene dieren.
Diepgaande live beeldbewerking met focus op afzonderlijke cellen of hun subcellular inhoud wordt echter beperkt in een geheel en voortdurend veranderende embryo. Om aan te pakken bepaalde cel biologische vragen met de hoogste resolutie, is het gebruik van celculturen handiger. Sommige cellijnen van zebravis bestaan, maar ze worden beschouwd als zwaar geselecteerde5,6,7 en hun voortplanting is vaak tijdrovend. Bovendien zijn alle beschikbare cellijnen fibroblast afgeleid, experimenten met behulp van celkweek voor één soort cellen te beperken. Dus vastgesteld we beide een efficiënte en easy-to-use protocol ter voorbereiding van primaire cellen rechtstreeks vanuit de zebravis embryo's en volwassen zebrafish hersenen, samen met benaderingen te verhogen de levensduur van de cultuur en aan het verbreden van de diversiteit van de gecultiveerde celtypes. Daarnaast presenteren wij een procedure om te transfect van embryonale primaire cellen met expressie constructies voor fluorescerende organel markeringen. Dus, cellulaire morphologies en subcellular structuren kunnen worden geanalyseerd met hoge ruimtelijke en temporele resolutie in verschillende celtypen, die hun belangrijkste functies te behouden.

culture and transfection of zebrafish primary cells

Zebravis embryo's transparant zijn en snel te ontwikkelen buiten de moeder, waardoor voor uitstekende in vivo imaging van dynamische biologische processen in een gewerveld dier intact en de ontwikkelingslanden. De gedetailleerde beeldvorming van de morphologies van verschillende celtypes en subcellular structuren wordt echter beperkt in hele mounts. Daarom opgericht wij een efficiënte en easy-to-use protocol om cultuur levende primaire cellen van zebravis embryo's en volwassen weefsel.
2 dpf zebrafish embryo's zijn in het kort, dechorionated, deyolked, gesteriliseerd, en losgekoppeld op enkele cellen met collagenase. Na een filtratie stap, zijn primaire cellen vergulde op glazen bodem gerechten en geteeld voor meerdere dagen. Verse culturen, zo veel als lange termijn differenciated ones, kunnen worden gebruikt voor hoge resolutie confocal imaging studies. De cultuur bevat verschillende soorten cellen, met gegroefde myocytes en neuronen worden prominente op poly-L-lysine coating. Wij vastgesteld tot een specifiek label subcellular structuur door fluorescerende marker eiwitten een electroporation protocol waarmee de transfectie van plasmide DNA in verschillende soorten cellen, met inbegrip van neuronen. Dus, in aanwezigheid van de exploitant gedefinieerd prikkels, het gedrag van de complexe cel en intracellulaire dynamiek van primaire zebrafish cellen kan worden beoordeeld met hoge ruimtelijke en temporele resolutie. Bovendien, met behulp van volwassen zebrafish hersenen, we laten zien dat de techniek beschreven dissociatie, evenals de basisvoorwaarden kweken, ook voor volwassen zebrafish weefsel werken.

Figuur 1G toont een doorvallend licht beeld van een typische cultuur afgeleid van wild type embryo's met gegroefde myocytes en clusters van neuron-achtige paneeltechnologie overvloedigste. Voor de identificatie van bepaalde celtypen gemakkelijker zijn een transgene lijn met cel type-specifieke expressie van een fluorescente proteïne kan worden gebruikt (Figuur 1 H).
