Utarbeidelse av primært cellekulturer fra 2 dpf sebrafisk er relativt lett å utføre for noen med erfaring i grunnleggende celle kultur teknikker. Sebrafisk (Danio rerio, D. rerio) er en populær modell virveldyr for mange felt av grunnleggende og biomedisinsk forskning1. Sebrafisk embryo utvikle raskt ex utero, gjennomsiktig og passe under et mikroskop, noe som gir gode forutsetninger for å studere virveldyrenes utvikling i en levende organisme. På grunn av genetisk tractability sebrafisk2, har mange stabil transgene reporter linjer med celle type-spesifikk uttrykk for ulike fluorescerende etablert tillater for observasjon av bestemt celle populasjoner. Sebrafisk samfunnet tilbyr et bredt spekter av såkalte Gal4-driver linjer som bærer en transgene uttrykke den syntetiske Kal4TA4 (eller KalTA3 tilsvarende GalFF) genet med Gal4-DNA-bindende domene gjær del viral transcriptional aktivisering domener kontrollert av celle type-spesifikk enhancers. Disse driveren linjene er krysset effektor linjene som bærer effekter av transgener som består av en definert oppstrøms aktivering sekvens (UAS) smeltet til et reporter gen. Kal4TA4 protein binder UAS-elementet, og dermed aktivere celle type-selektiv uttrykk for reporter genet3,4. Denne tilnærmingen gir svært variert kombinasjon studier av nesten alle tilgjengelige enhancer og reporter elementer i dobbel-transgene dyr.
Men er grundig live bildebehandling med fokus på enkeltceller eller subcellular innhold begrenset i en hel og stadig skiftende embryo. For å håndtere bestemt celle biologiske spørsmål med høyeste oppløsningen, er bruk av cellekulturer ofte å foretrekke. Noen linjer av sebrafisk finnes, men de anses som svært valgte5,6,7 og deres forplantning er ofte tidkrevende. Videre er alle tilgjengelige celle linjene fibroblast stammer, begrense eksperimenter ved hjelp av cellekultur til én type celler. Derfor opprettet vi både en effektiv og enkel å bruke protokoll for å forberede primære celler direkte fra sebrafisk embryoer og voksen sebrafisk hjernen, sammen med tilnærminger til å øke levetiden til kultur og å utvide mangfoldet av dyrket celletyper. I tillegg presenterer vi en prosedyre for å transfect embryonale primære celler med uttrykket konstruksjoner for fluorescerende organelle markører. Dermed kan mobilnettet morphologies og subcellular strukturer analyseres med høy romlig og tidsmessige oppløsning i forskjellige celletyper som beholde sine viktige funksjoner.

culture and transfection of zebrafish primary cells

Sebrafisk embryoer er gjennomsiktig og utvikle raskt utenfor mor, dermed gir for utmerket i vivo avbilding av dynamisk biologiske prosesser i en intakt og utvikle virveldyr. Men er detaljert avbilding av morphologies av forskjellige celletyper og subcellular strukturer begrenset i hele mounts. Derfor etablert vi en effektiv og enkel å bruke protokoll til kultur lever primære celler fra sebrafisk embryoer og voksen vev.
I korthet, er 2 dpf sebrafisk embryoer dechorionated, deyolked, sterilisert og avstand til enkeltceller med collagenase. Etter filtrering trinn, er primær celler belagt på glass bunn retter og dyrket i flere dager. Frisk kulturer, som lang sikt differenciated seg, kan brukes for høy oppløsning AC confocal imaging studier. Kulturen inneholder ulike celletyper, med striated myocytter og neurons blir fremtredende på poly-L-lysine belegg. Spesielt etiketten subcellular strukturer av fluorescerende markør proteiner etablert vi også en electroporation protokoll som tillater transfection av plasmider DNA i ulike celletyper, inkludert nerveceller. Således, i nærvær av operatør definerte stimuli, komplekse celle atferd og intracellulær dynamikken i primære sebrafisk celler kan vurderes med høy romlig og tidsmessige oppløsning. I tillegg bruker voksen sebrafisk hjernen, viser vi at teknikken beskrevet dissosiasjon, i tillegg til de grunnleggende culturing forholdene, også arbeide for voksen sebrafisk vev.

Figur 1G viser en overføres lett bilde av en typisk kultur avledet fra vill type embryo med striated myocytter og klynger av Nevron som cellene blir mest tallrike. For å identifisere visse celletyper lettere, en transgene linje med celle type-spesifikk uttrykk for et lysstoffrør protein kan brukes (figur 1 H).
