1. 细胞和材料的制备

在这项研究中, 我们开发了 o 型容器, 并在其中进行了 hek293 悬浮培养, 以形成和扩大均匀的集料。在传统的培养皿中, 轨道晃动培养产生了两种不同直径的集料, 而我们观察到 o 形容器中的均匀集料 (图 1)。根据对染色珠子在轨道晃动条件下的分布的观察, 珠子聚集在传统培养盘的中底。这种聚集大概导致了细胞密度的差异, 导致了不同大小的聚集体。或者, 珠子分布在 o 形的盘子中, 没有中间底部区域 (补充图 1)。这种分布可能会导致 o 型容器中的聚集体尺寸均匀。另一种广泛使用的生产均匀集料的方法是微孔培养, 但这种方法存在一些问题, 例如在提供培养基时没有从微孔13中退出的聚集物。在 o 型容器的情况下, 可以在简单的悬浮培养中产生均匀的集料。
轨道晃动培养是一种常用的哺乳动物细胞培养系统。哺乳动物细胞的批量生产有多种培养方法, 如搅拌罐生物反应器14、波动袋15、旋转瓶.轨道晃动培养不包括用于搅拌介质的内叶轮, 与搅拌罐生物反应器不同。此功能类似于波浪形袋和旋转瓶。这些无叶轮培养系统可以避免叶轮周围剪切力对细胞的破坏, 实现悬浮培养中的低剪切应力。特别是轨道晃动培养系统具有高可扩展性和低剪切应力的优点, 对哺乳动物细胞等敏感细胞的大规模生产是有效的。
o 型容器可以改善集料形成过程中剩余的轨道晃动培养问题。在轨道晃动培养系统中, 漂浮的细胞迁移到容器的中心和底部, 这被称为 "爱因斯坦的茶叶悖论"11。这种迁移导致常规轨道晃动容器中的聚集不均匀和集料生产不均匀。在这项研究中, o 形血管防止细胞浓度进入血管的中心底部, 这被推测为轨道晃动 o 形血管均匀聚集的原因。
组织学分析显示, 常规菜中的聚集体含有脱核细胞 (图 2a)。相反, 脱核细胞并没有出现在 o 型血管的集料中 (图 2b和2b)。这些脱核细胞可能是由葡萄糖、谷氨酰胺和氧气等底物缺乏引起的.根据尺寸测量, o 形容器中的集料直径均相低于400μm。相反, 在传统的盘子中, 一些集料的直径大于 400μm, 而这些集料包括脱核细胞。这一结果表明, 在 o 型血管中建立均匀规模的集料是控制集料质量的有效方法。此外, 还推测通过透气聚乙烯薄膜进行氧合, 防止了 o 形袋中脱核细胞的出现。

悬浮细胞培养在再生医学1和重组蛋白生产2的细胞生产中发挥着重要作用, 因为它很容易扩大规模, 实现高细胞密度, 有时高达 107个细胞/ml5。中国仓鼠卵巢 (cho) 细胞3和人类胚胎肾细胞 293 (hek293)4已在悬浮培养中生长, 人类多能干细胞 (hpsc) 最近在悬浮培养系统中生长, 以促进再生治疗1,6,7。预计今后暂停文化的使用将增加。
在悬浮培养中, 一些细胞系不能作为单个细胞生长, 因此必须形成聚集体。例如, 如果不形成聚集体, hpsc 就无法在悬浮条件下生存。然而, 对综合增长的这一要求给统一的悬浮文化带来了困难。一个困难是在文化早期形成了均匀的集料, 这决定了悬浮培养的效率.另一个困难是营养物质的大量转移转化为集料。特别是氧气供应限制了集料的最大大小, 氧气供应不足导致集料中心9 处坏死。因此, 细胞聚集物的悬浮培养比传统的悬浮培养物更难获得。然而, 悬浮培养对于生物医学和生物技术的应用至关重要。
轨道晃动容器是最简单的悬浮培养系统之一, 可实现培养基混合物, 而不需要叶轮搅拌。叶轮的剪切应力和动态介质流动是悬浮培养的主要问题, 因为它导致细胞损伤和分化。为了实现较低剪切应力的搅拌, 研究人员和行业开发了各种轨道晃动容器系统2,10.
然而, 传统的养殖船并不是为轨道晃动培养而设计的。在轨道晃动的容器中, 细胞被一种称为 "爱因斯坦的茶叶悖论"的介质流吸引到血管的中心底部, 这种流动导致细胞的不均匀聚集。由离心力和培养基与容器之间的摩擦引起的圆形流动将细胞扫入中心11,12。此外, 大众文化的传统文化袋是方形的, 不适合轨道晃动。

根据我们的测量, 在传统的盘子中生长的集料在 5 d 轨道晃动后具有不同的直径。相反, 在5d 型容器中生长的集料直径要均匀得多。传统的菜肴培养体显示有小的集料 (50-200μm), 而 o 形容器中的培养物不包含任何小的集料 (图 2a)。根据基于图像的尺寸测量, 传统的菜文化显示了两个不同的峰值 (图 2b1), 表明了骨料尺寸的大偏差。这一结果表明, 在同一培养条件下, 常规菜中的聚集体生长在两种不同的条件下, 可能导致异质细胞质量。另一方面, o 型容器中的集料比传统的盘状容器显示出单一的峰值, 直径偏差较小 (图 2b2和2b3), 这表明此类集料的质量可能更为均匀。

血氧西林和 eosin 染色的集料横截面表明, 在常规菜中生长的集料有一些脱核细胞和坏死核 (图 3a)。然而, 在 o 型容器中生长的集料没有显示任何坏死核心 (图 3b和3b)。特别是, 在 o 形袋中生长的集料与传统菜品中的集料一样大, 但没有任何坏死 (图 3c)。这些结果表明, 透气袋提供了足够的氧气培养。

细胞计数显示, 85% 以上的细胞在每个血管中存活了5d 悬浮培养 (图 4a)。最终细胞密度约为 1.5-2.0 x10 6 细胞。虽然没有显著差异, 但 o 形袋的生长率高于其他容器 (图 4b)。常规盘中、o 形盘和 o 形袋中细胞的特异性生长速率分别为0.018、0.018 和 0.018h-1。
