De protokoller, der er beskrevet ovenfor kræver praksis men bør opnås med minimal fejlfinding. På en gennemsnitlig dag ville vi få 6-8 brugbar arterioler fra isolation og opnå 3-4 vellykkede eksperimenter. Hvis der opstår problemer, men er der nogle skridt, der kan træffes for at forbedre succesrate. Lejlighedsvis har vi fundet, især når du bruger yngre rotter (< 8 uger), at udbyttet af arterioler kan være lav. For at omgå dette problem, vil vi foreslå, centrifugering den retinale væv (10-30 s 500 x g) mellem hver af ændring skridt i trin 1.12-1.14. Dette ofte bidrager til at forbedre udbyttet af fartøjer, men vil også øge mængden af celle debris inden for udarbejdelsen.
Når cannulating fartøjer til pres blodkar undersøgelser er det vigtigt at kontrollere arterioler nøje for eventuelle sidegrene, der kan have været kløvet tæt på webstedet tvedeling. Arterioler kan isoleres fra nethinder af eksperimentelle gnavere modeller af diabetes ved hjælp af de metoder, der er beskrevet i afsnit 1. Når eksperimentere på isolerede retinal arterioler fra diabetisk dyr, er det vigtigt at forsøge at nøje efterligne hyperglykæmisk betingelser opleves af fartøjer i vivo. Derfor ville vi normalt hæve D-glucose niveauer i vores isolation og eksperimenterende løsninger til 25 mM. Fortykkelse af vaskulære kælderen membraner er et velkendt fænomen i retinal fartøjer under diabetes34,35. Når du bruger dyr med langvarig diabetes (> 1 måneder sygdom varighed), er øget enzym koncentrationer eller fordøjelsen gange ofte nødvendig hen til muliggøre anvendelse af patch-clamp optagelse metoder.
En vigtig begrænsning af brug af ex vivo isoleret retinal arterioler at studere retinal vaskulær fysiologi og patofysiologi er tabet af de omkringliggende retinale neuropile. Selv om fjernelse af de retinale neuronal og glial celler giver nem adgang til de retinale vaskulære glatte muskelceller for celle fysiologi undersøgelser, kan svar af fartøjer til vasoaktive mediatorer ændre sig dramatisk i fravær og tilstedeværelsen af retinal væv. Handlinger af adenosin-tri-phosphat (ATP), for eksempel, giver en god illustration af dette punkt. I isolerede rotte retinal arterioler, tilsætning af ATP udløser en robust konstriktion af fartøjer36, mens i tilstedeværelsen af et intakt neuropile, fartøjer spile37. Derfor, når det er muligt, vi normalt forsøger at validere hovedresultaterne fra vores isolerede arteriole præparater ved hjælp af ex vivo retinal hele-mounts og i vivo målinger af fartøjet diameter og blood flow16,37 . Af note, er nye metoder for nylig dukket op for at studere små arterioler og kapillærer i hele perfunderet svin nethinder ex vivo38,39. Sådanne præparater er tilbøjelige til at forbedre vores forståelse af hvordan den retinale neuropile regulerer retinal arteriolære og kapillær tone og hvordan ændringer i retinal Hæmodynamik modulere neuronal aktivitet i nethinden.
Selv om de procedurer, der er beskrevet i denne hvidbog er fokuseret på brugen af isolerede rotte retinal arterioler til forståelse arteriolære glat muskel celle fysiologi, vi er i øjeblikket at udvikle protokoller for at også aktiverer undersøgelse i endothelial cellefunktion i disse fartøjer. I indledende arbejde, har vi haft succes med modificerer den enzymatiske fordøjelsen af retinale arteriolære segmenter til udbytte levedygtige endotel celle rør, der er indstillet til Ca2 + imaging og patchclamp optagelse undersøgelser. Cannulation af de isolerede arterioler i begge ender, aktivering intraluminal leveringen af narkotika, kan i fremtiden også aktiver endotel-afhængig vasodepressivt svar skal undersøges i disse fartøjer.

Forstå hvordan blodgennemstrømningen er kontrolleret i nethinden er et vigtigt mål, da unormal blodgennemstrømning har været involveret i patogenesen af en række sight-truende retinal sygdomme1,2,3, 4. Den retinale omsætning, der forsyner den indre nethinde neuroner og gliaceller, har en ende arterie arrangement, med alle blod fra retinal arterier og arterioler passerer gennem kapillærerne retinale vener og endelig vener5. Blodgennemstrømningen i nethinden er reguleret af tonen i de retinale arterier og arterioler samt de kontraktile aktivitet af pericytes beliggende på væggene i retinal kapillærer og post kapillær venules6,7, 8. kontrol af retinal vaskulær tone er komplekse og moduleres af en vifte af input fra kredsløbssygdomme og omkringliggende retinale væv, herunder blod gasser, cirkulerende molekyler og hormoner, og vasoaktive stoffer frigives fra de retinal vaskulær endotel og macroglia9,10,11. De retinale arterioler er den lille arterielle filialer af nethinden og er sammensat af et enkelt lag af vaskulære glatte muskelceller og en indre foring af langs arrangeret endotelceller12,13, 14. disse fartøjer udgør de vigtigste site af vaskulære modstand inden for den retinale omsætning og derfor spiller en vigtig rolle i den lokale kontrol af retinale blodgennemstrømning. Retinal arterioler regulere kapillær blod flyde i nethinden ved at dilatere eller snærende deres luminale diameter, medieret af ændringer i vaskulære glatte muskulatur kontraktilitet10,15,16. Forstå de molekylære mekanismer gennem hvilke retinal arterioler regulere retinal perfusion derfor kræver præparater hvor arteriolære glatte muskelceller kan tilgås og studerede under forhold så tæt på fysiologiske som muligt.
Ex vivo præparater af isolerede retinale blodkar giver adgang til de vaskulære glatte muskelceller, mens de stadig bevare deres funktionalitet og sammenkobling med det underliggende endotelet. De fleste undersøgelser til dato ved hjælp af isolerede fartøjer har fokuseret på store kvæg eller svin arteriel fartøjer (60-150 µm). Disse kan monteres i kommercielt tilgængelige wire eller pres myograph systemer til at sætte farmakologiske forhør af vaskulær glat muskel celle kontraktile mekanismer17,18. Sådanne præparater har i høj grad bidraget til vores viden om retinal vaskulær fysiologi under normale forhold. Få studier har brugt retinal arterioler isoleret fra små forsøgsdyr som deres mindre diameter (~ 8-45 µm) forhindrer deres brug i konventionelle blodkar systemer19,20,21,22. En vigtig fordel, dog er af ved hjælp af fartøjer fra små forsøgsdyr genetisk modificerede, transgene og retinal sygdomsmodeller bred tilgængelighed. Lille laboratoriedyr er også mere modtagelig for i vivo intervention undersøgelser.
Her beskriver vi ligetil protokoller til isolering og cannulating rotte retinal arterioler for pres blodkar eksperimenter. Ca2 + imaging og Elektrofysiologi protokoller ved hjælp af disse fartøjer er også detaljeret. Nethinden er en yderst metabolisk aktive væv, der kræver en betydelig blodforsyning. Den retinale omsætning understøtter den indre Nethinde, mens de choroidal fartøjer levere fotoreceptorer. Ændringer i retinal perfusion bidrage til talrige sight-truende lidelser, herunder diabetisk retinopati, glaukom og retinal gren vene tillukning.
