Protokollene beskrevet ovenfor krever praksis, men bør være oppnåelig med minimal feilsøking. På en gjennomsnittlig dag ville vi få 6-8 brukbare arterioler fra isolasjon og oppnå 3-4 vellykket eksperimenter. Hvis det oppstår problemer, men er det noen tiltak som kan iverksettes for å forbedre suksessrate. Noen ganger har vi funnet, spesielt når du bruker yngre rotter (< 8 uker), at avkastningen av arterioler kan være lav. For å omgå dette problemet, vil vi foreslå sentrifugering netthinnen vev (10-30 s på 500 x g) mellom hver av føden trinnene i fremgangsmåten 1.12-1.14. Dette ofte bidrar til å forbedre avkastningen av fartøy, men vil også øke mengden av celle rusk i utarbeidelse.
Når cannulating fartøy for press myography studier er det viktig å sjekke arterioler nøye for side-grenene som kan ha vært kløyvde nær bifurkasjonen området. Dette er en vanlig årsak til lekkasje og tap av press under eksperimentering. Hovedproblemet som kan oppstå med [Ca2 +]jeg protokollene er nivået av fargestoff lasting. Dårlig fargestoff lasting fører til lav signal-til-støy-forhold, mens overbelastning fører til avbrudd i normal Ca2 + homeostase. For å redusere sannsynligheten for noen av disse problemene, en liten aliquot av netthinnen homogenate kan fjernes, og isolert fartøy kontrolleres regelmessig under lasting protokollen. Som fremhevet tidligere var en viktig motivasjon bak utviklingen av protokollene i dette manuskriptet å bedre forstå hvorfor blodstrøm avbrytes under retinal vascular sykdommer. Når du eksperimenterer på isolerte retinal arterioler fra diabetiker dyr, er det viktig å prøve å kopiere tett hyperglycemic forholdene oppleves av fartøy i vivo. Derfor vil vi normalt øke D-glukose nivåene i isolasjon og eksperimentelle løsninger til 25 mM. Jevning av vaskulær kjelleren membraner er et velkjent fenomen i netthinnen fartøy i diabetes34,35. Når du bruker dyr med langvarig diabetes (> 1 måneder sykdom varighet), er økt enzym konsentrasjoner eller fordøyelsen ganger ofte nødvendig å aktivere programmet patch-klemme opptak metoder.
En viktig begrensning av benytter ex vivo isolert retinal arterioler å studere retinal vaskulær fysiologi og patofysiologi er tapet av det omkringliggende retinal neuropile. Selv om fjerning av netthinnen glial og neuronal cellene gjør enkel tilgang til den retinal vaskulære glatte muskelcellene for cellen fysiologi studier, kan responsen av fartøyene vasoactive meglere endre seg dramatisk i fravær og tilstedeværelsen av retinal vev. Handlingene til adenosin tri-fosfat (ATP), for eksempel gi en god illustrasjon på dette punktet. I isolerte rotte retinal arterioler, tillegg av ATP utløser en robust innsnevring av fartøy36, mens i nærvær av en intakt neuropile, fartøyene dilate37. Selv om prosedyrene som er beskrevet i denne artikkelen fokuserer på bruk av isolerte rotte retinal arterioler for forståelse arterioler glatt muskel celle fysiologi, utvikler vi for tiden for å også aktivere studiet av endothelial celle funksjon i disse fartøyene. I forarbeidet, har vi vært vellykket i å endre enzymatisk fordøyelsen av netthinnen arterioler segmenter å gi levedyktige endothelial celle rør som er mottakelig for Ca2 + bildebehandling og patchclamp innspillingen studier. Cannulation av de isolerte arterioler i begge ender, slik at intraluminal levering av narkotika, kan i fremtiden også aktivere endotelet avhengige av vasodilatory svar undersøkes i disse fartøyene.

Forstå hvordan blodstrøm er kontrollert i netthinnen er et viktig mål siden unormal blodstrøm har vært innblandet i patogenesen av en rekke sight-truende retinal sykdommer1,2,3, 4. Netthinnen sirkulasjon, som leverer den indre retinal nevroner og gliacellene, har en slutt arterien ordning, med alle blod fra netthinnen arterier og arterioler passerer gjennom kapillærene retinal venules og til slutt årer5. Blodstrøm i netthinnen er regulert av tonen i netthinnen arterier og arterioler samt kontraktile aktiviteten til pericytes på veggene i netthinnen kapillærene og post kapillært venules6,7, 8. kontroll av netthinnen vaskulær tone er kompleks og modulert av en rekke innganger fra sirkulasjonssystemet og omkringliggende netthinnens vev, inkludert blod gasser, sirkulerende molekyler og hormoner, og vasoactive stoffer ut fra den netthinnen vaskulære endotelet og macroglia9,10,11. Netthinnen arterioler er små arteriell grenene av netthinnen og består av en enkelt lag av vaskulær glatte muskelcellene og en indre foring av langs ordnet endotelceller12,13, 14. disse fartøyene form hovednettstedet vaskulær motstand i netthinnen sirkulasjon og derfor spille en viktig rolle i kontroll av netthinnen blodstrøm. Netthinnen arterioler regulere kapillær blodstrøm i netthinnen av utvidelsen eller constricting deres luminal diameter, formidlet av endringer i vaskulær glatt muskel contractility10,15,16. Forstå molekylære mekanismer gjennom hvilke retinal arterioler regulere retinal perfusjon derfor krever forberedelser der den arterioler glatte muskelcellene kan nås og studerte under forhold så nær fysiologiske som mulig.
Ex vivo forberedelser av isolerte retinal blodårer gir tilgang til vaskulær glatt muskelceller, mens du fremdeles beholder sin funksjonalitet og tilkoblinger med underliggende endotelet. De fleste studier ennå med isolert skip har fokusert på store storfe eller svin arteriell fartøy (60-150 µm). Dette kan monteres i kommersielt tilgjengelig wire eller press myograph systemer å aktivere farmakologiske avhør av vaskulær glatt muskel celle kontraktile mekanismer17,18. Slike forberedelser har sterkt bidratt til vår kunnskap om retinal vaskulær fysiologi under normale forhold. Noen studier har brukt retinal arterioler isolert fra små forsøksdyr som deres mindre diameter (~ 8-45 µm) forhindrer bruken i konvensjonelle myography systemer19,20,21,22. En viktig fordel, men er ved å bruke fartøy fra små forsøksdyr bredt tilgjengeligheten av genetisk modifisert, transgene og retinal sykdom modeller. Liten forsøksdyr er også mer mottagelig for i vivo intervensjon studier.
Her beskriver vi enkel protokoller for å isolere og cannulating rotten retinal arterioler for press myography eksperimenter. Ca2 + bildebehandling og elektrofysiologi protokoller bruker disse fartøyene er også detaljert. Dette kan gi ytterligere innsikt i regulering av vaskulær glatt muskel contractility og blodstrøm i netthinnen.

Netthinnen er en svært metabolsk aktive vev som krever en betydelig blodtilførsel. Netthinnen sirkulasjon støtter indre netthinnen, mens choroidal fartøyene forsyning av fotoreseptorer. Endringer i netthinnen perfusjon bidra til mange sight-truende lidelser, inkludert diabetisk retinopati, grønn stær og retinal gren vene occlusions. Forstå molekylære mekanismer involvert i kontrollen av blodstrøm gjennom netthinnen og hvordan dette er endret under okulær sykdom kunne føre til identifisering av nye mål for behandling av disse forholdene. Netthinnen arterioler er viktigste motstand fartøyene av netthinnen, og derfor spiller en viktig rolle i å regulere retinal hemodynamics gjennom endringer i luminal diameter. De siste årene, har vi utviklet metoder for å isolere arterioler fra rotte netthinnen som er egnet for en rekke applikasjoner, inkludert celle fysiologi studier. Dette preparatet har allerede begynt å gi ny innsikt i hvordan blodstrøm styres i netthinnen og har tillatt oss å identifisere noen av de viktige endringene som oppstår under okulær sykdom. I denne artikkelen vi beskriver metoder for isolering av rotte retinal arterioler og inkludere protokoller for bruk i patch-klemme elektrofysiologi, kalsium bildebehandling og trykk myography studier. Disse fartøyene er også mottagelig for bruk i PCR-, vestlige blotting og immunohistochemistry-baserte studier.
