Vi beskriver en simpel kirurgisk induktion af CHL med verifikation ved hjælp af Otoskop visualisering og adfærdsmæssige Clap chok respons i rotte. Her viser vi metoden på rotte og tidligere denne metode er blevet anvendt til gerbils og mus. Metoden kan nemt vedtages til andre gnavere. Induktion af CHL gør det muligt at studere en diskret form for høretab, som manifesterer sig i auditive kortikale forandringer og psykofysiske adfærdsmæssige fund4,5,</...

Forekomsten af høretab hos børn og voksne er henholdsvis ca. 19,5%1 og 15,2%2 . Imidlertid modtager ca. 39,3% af alle nyfødte med en unormal Hørescreening ikke afhjælpende behandling som rapporteret af centre for Disease Control3. Høretab er en almindeligt undersøgt tilstand, og gnaver er en robust model til at studere normal hørelse og høre relaterede lidelser4,5,6,7,8,9 ,10,11,12,13,14,15. Hørelidelser såsom konduktivt høretab (CHL) fører til en stigning i den kortsigtede synaptiske depression i auditiv cortex4, hvilket resulterer i lavvandede Psykometriske skråninger forbundet med frekvens modulerings detekterings tærskler 5. konduktivt høretab modeller ved kirurgisk fjernelse/forskydning af Malleus, trommehinden membran (TM) punktering eller øreprop er let anvendes og tillade hurtig induktion af høretab model5,14 ,15,16,17,18. Formålet med denne protokol og metode er at demonstrere en simpel og reproducerbar CHL-model i gnavere.
Denne protokol er billig (≈ USD 300 med alle værktøjer) og kan let ændres til forskellige forskningsaktiviteter. Rotten har haft detaljerede vurderinger af mellemøret anatomi19,20,21,22,23, kirurgiske tilgange24, modeller i otitis media25, 26,27 og TM punktering regenerering16,17,18,28,29,30, hvilket gør det til en ideel model til at studere Høretab. Her er en simpel CHL-induktions procedure beskrevet med verifikation af Otoskop og adfærdsmæssig vurdering med Clap-chok-respons i rotte, som derefter kan bruges til at udforske yderligere sequelae af høretab. CHL-proceduren induceres ved kirurgisk punktering af TM. Verifikation af CHL-proceduren udføres af Otoskop visualisering for at fastslå fravær af TM. Adfærdsmæssige vurderinger udføres af et højt decibel (dB) lydtryksniveau (SPL) hånd-Clap. Denne metode er blevet anvendt tidligere i en række gnavere. Det er let at replikere, producerer robuste Psykometriske forskelle og ændringer i neurale fysiologiske responser4,5,16,17,18.

Konduktivt høretab (CHL) er en udbredt nedsat hørelse hos mennesker. Målet med protokollen er at beskrive en simpel kirurgisk procedure for inducerende CHL i gnavere. Protokollen demonstrerer CHL ved tympaniske membran punktering. Verifikation af CHL kirurgi var ved Otoskop undersøgelse og adfærdsmæssige vurdering af Clap chok Response, både replikkable og pålidelige, og er enkle metoder til at demonstrere høretab har fundet sted. Den enkle CHL-procedure er fordelagtig på grund af dens reproducerbarhed og fleksibilitet til forskellige gøreformål i høretab forskning. Begrænsningerne af inducerende CHL ved en kirurgisk tilgang er forbundet med indlæringskurven til at udføre den kirurgiske procedure og tillid til audiologisk undersøgelse. Inducerende en hørenedsættelse af CHL giver en til let at studere neurale manifestationer og adfærdsmæssige udfald af høretab.

Den enkle CHL-procedure blev udført på 90 rotter og ud af denne gruppe 2 havde signifikant blødning og 2 havde ikke høretab den næste dag som vurderet af adfærdsmæssige Clap startle. Disse fire rotter blev kasseret. Rotter bør kasseres som beskrevet af årsagerne i diskussionen på grund af komplikationer. Det overordnede mål med denne undersøgelse er at etablere en pålidelig høretab induktion via kirurgisk tympanisk membran punktering og verifikation ved otoskop visualisering og adfærdsmæssige vurdering af klappe startle. Formålet med vores praktiske er at udvikle en model for menneskelige hørelse konduktive lidelser. Vi demonstrerer i den praktiske, konduktiv høretab kirurgisk procedure, som er en forenkling i malleus fjernelse, metoden til at kontrollere konduktivt høretab ved otoskop visualisering, og en simpel adfærdsmæssige klap startle, som validerer, at konduktivt høretab har fundet sted.Den største fordel ved den procedure, vi beskrev, er ikke-invasive procedurer, verifikation af konduktivt høretab ved otoskop og validering ved klaptab.
