We beschrijven een eenvoudige chirurgische inductie van CHL met verificatie met behulp van Otoscoop visualisatie en Behavioral Clap schrikreactie respons bij ratten. Hier demonstreren we de methode op rat en eerder is deze methode toegepast op gerbils en muizen. De methode kan gemakkelijk worden overgenomen door andere knaagdieren. De prevalentie van het gehoorverlies bij kinderen en volwassenen is respectievelijk ongeveer 19,5%1 en 15,2%2 . Echter, ongeveer 39,3% van alle pasgeborenen met een abnormale Gehoorscreening krijgen geen corrigerende behandeling zoals gerapporteerd door de centra voor Ziektecontrole3. Gehoorverlies is een algemeen bestudeerde aandoening en het knaagdier is een robuust model voor het bestuderen van normale gehoor-en gehoor aandoeningen4,5,6,7,8,9 ,10,11,12,13,14,15. Gehoor aandoeningen zoals Conductief gehoorverlies (CHL) leiden tot een toename van de korte termijn synaptische depressie in de auditieve cortex4, wat resulteert in onlagere psychometrische hellingen geassocieerd met frequentiemodulatie detectiedrempels 5. geleidende gehoorverlies modellen door chirurgische verwijdering/verplaatsing van de malleus, trommelvlies membraan (TM) punctie of oordopje worden gemakkelijk gebruikt en maken de snelle inductie van het gehoorverlies model5,14 ,15,16,17,18. Het doel van dit protocol en de huidige methode is om een eenvoudig en reproduceerbaar CHL-model bij knaagdieren aan te tonen.
Het huidige protocol is goedkoop (≈ USD $300 met alle tools), en gemakkelijk te gebruiken voor verschillende onderzoeksactiviteiten. De rat heeft gedetailleerde beoordelingen van de anatomie van het middenoor19,20,21,22,23, chirurgische benaderingen24, modellen in otitis media25, 26,27 en TM punctie regeneratie16,17,18,28,29,30, waardoor het een ideaal model om te studeren Gehoorverlies. Hier wordt een eenvoudige CHL-inductie procedure beschreven met verificatie door Otoscoop en gedrags beoordeling met Clap schrikreactie respons bij ratten, die vervolgens kunnen worden gebruikt om extra gevolgen van gehoorverlies te onderzoeken. De CHL-procedure wordt geïnduceerd door een chirurgische punctie van de TM. Verificatie van de CHL-procedure wordt uitgevoerd door Otoscoop-visualisatie om de afwezigheid van de TM te bepalen. Gedrags beoordeling wordt uitgevoerd door een hoge decibel (dB) geluidsdrukniveau (SPL) hand-clap. Deze methode is eerder toegepast in een verscheidenheid van knaagdieren. Het is gemakkelijk te repliceren, produceert robuuste psychometrische verschillen en veranderingen in neurale fysiologische reacties4,5,16,17,18.

Conductief gehoorverlies (CHL) is een overwegend gehoorstoornis bij de mens. Het doel van het protocol is om een eenvoudige chirurgische procedure te beschrijven voor het induceren van CHL bij knaagdieren. Het protocol demonstreert CHL door trommelvlies membraan punctie. Verificatie van de CHL-chirurgie was door Otoscoop-onderzoek en gedrags beoordeling door Clap schrikreactie-respons, zowel repliceerbaar als betrouwbaar, en zijn eenvoudige methoden om gehoorverlies aan te tonen. De eenvoudige CHL-procedure is voordelig vanwege de reproduceerbaarheid en flexibiliteit van verschillende activiteiten in onderzoek naar gehoorverlies. De beperkingen van inducerende CHL door een chirurgische benadering worden geassocieerd met de leercurve om de chirurgische ingreep en het vertrouwen in Audiologisch onderzoek uit te voeren. De eenvoudige CHL-procedure werd uitgevoerd op 90 ratten en uit deze groep 2 had een significante bloeding en 2 had de volgende dag geen gehoorverlies als beoordeeld door Behavioral Clap Startle. Het doel van onze praktijk is het ontwikkelen van een model voor menselijke gehoorgeleidende stoornissen. Introduceer de microscissors in het midden van het gehoorkanaal, met veel aandacht om het weefsel niet te stelen. Evalueer elk rattenoor voor en na de operatie met een otoscoop met een speculum met een kleine diameter. Bevestig bindbaar gehoorverlies met gedragsklappenschrrijf. De CHL rat wordt naast een normale rat geplaatst in twee aangrenzende kooien.Deze worden geplaatst in een stille kamer.
