De uitdagingen van de succesvolle bemonstering (bulk en incrementele) van gebit is afhankelijk van de toegang tot kennis met betrekking tot boren technieken en proef van de voorbereiding, naast de investering in relatief goedkope apparatuur. Stabiele koolstof en zuurstof isotoop analyses van tand email carbonaat is gebruikt om te studeren voorbij menselijke inname, spenen, mobiliteit, alsmede faunistische afhankelijkheid van de vegetatie, het verkeer van dieren en vee foddering. Deze toepassingen zijn uitvoerig besproken en geëvalueerd voor allerlei milieuomstandigheden, met vermelding van de gevolgen van lokale dorheid, temperatuur, waterbronnen en vegetatie composities1,2, 3,4,5,6. De verscheidenheid van mogelijke toepassingen in Archeologie en paleontologie, alsook het goede behoud van tand email carbonaat, heeft het een aantrekkelijk materiaal voor stabiele isotoop werk3gemaakt. Methoden van bemonstering, voorbehandeling, en diagenese screening worden kort beschreven in een aantal eerdere publicaties1,7. Grondige verbale en visueel demonstraties blijven echter grotendeels niet beschikbaar is, vooral voor mensen buiten Archeometrie laboratoria en tussen laboratorium groepen met beperkte financiële middelen waar de interesse in het gebruik van deze techniek groeit 5.
Tandglazuur is voornamelijk samengesteld uit hydroxyapatiet (bioapatite) kristalaggregaten8 groter dan die in het bot, waardoor het beter bestand tegen post mortem diagenetic Ionische vervangingen en besmetting3. Moderne studies hebben aangetoond dat stabiele koolstof-isotoop (δ13C) metingen van faunistische tand betrouwbaar record dierlijke voeding en gedrag9,10 geëmailleerde. De stabiele zuurstof isotoop (δ18van O) waarde van tandglazuur wordt bepaald door de zuurstof isotopische samenstelling van geconsumeerde water, waarin water in plant en dierlijk voedsel, drinkwater, ademhaling, evenals verschillende milieueffecten op het water die kunnen leiden tot verdere isotopische fractionering (bv., droogte, temperatuur, hoogte, regenval bedrag, continentale locatie)11. Dit is een populaire methode voor dieet- en milieu wederopbouw gemaakt in paleontologische, archeologisch en paleoecological onderzoek.
De periode van de vorming van de glazuur van de tand is relatief kort (jaar) en verschilt afhankelijk van de tand worden bemonsterd. Voor de mens, eerste molaire emaille mineralizes tussen geboorte en 3 jaar, premolars mineralize tussen de 1,5 en 7 jaar oud, tweede kiezen mineralize tussen 2,5 en 8 jaar oud en derde kiezen mineralize tijdens de adolescentie, tussen 7 en 16 jaar12 . Gegeven dat tandglazuur vormen stapsgewijs over haar periode van vorming, kan worden bemonsterd in bulk langs de gehele groei as of bemonsterd opeenvolgend te onderzoeken van de veranderingen in dieet en omgeving die hebben plaatsgevonden tijdens de periode van de formatie13 . Chronologisch bestelde dieet veranderingen binnen een bepaalde tand is waarneembaar voor de mens en andere dieren1,14, voorlichting over Inter jaarlijkse variatie van de seizoensgebonden en dieet.
Terwijl glazuur meestal resistent tegen diagenese is, isotopische wijzigingen als gevolg van de begrafenis milieu zijn mogelijk en15,16, waardoor experimentele controles en voorbehandeling keuzes nuttig zijn waargenomen. Hoewel het is niet de enige beschikbare methode, heeft Fourier transform infrarood spectroscopie (FTIR), met name in verzendmodus verzwakt, ontpopt als een snel, goedkoop, en relatief toegankelijk methode voor de beoordeling van de taphonomic wijziging van tandglazuur, met name in paleontologische contexten17,18,19,20. Gedetailleerde protocollen en opname normen blijven echter relatief ontoegankelijk voor veel mensen buiten het gebied van de geochemie of materiaal wetenschap.
Reactietijden en de chemische stoffen in de voorbehandeling van tandglazuur door onderzoekers in dienst ook verschillen in de literatuur, vaak met beperkte aandacht over de vraag wat deze variabiliteit om stabiele koolstof en zuurstof isotoop waarden van het monster21 doen kan ,22. Wij rapporteren hier, een aanpak dat gebruik verdund azijnzuur (0.1 M) voor de voorbehandeling van glazuur poeder monsters. Echter, gezien het feit dat de verschillen in isotopische metingen, als gevolg van de voorbehandeling relatief kleine voor tandglazuur zijn, is het best voor de onderzoekers te volgen van de protocollen voor datasets waarmee ze willen hun gegevens vergelijken met11. Bovendien, waar kleine opeenvolgende monsters zijn genomen, met name op de monsters van het Holoceen, geen voorbehandeling kan worden gekozen (na diagenetic proeftoetsen) monster verspilling.
Hoewel de methoden we hier rapporteren niet nieuw, om onze kennis absoluut, is dit de eerste keer dat een grondige geschreven en visuele documentatie van bulk en sequentiële bemonstering, voorbehandeling keuzes en diagenetic selectievakje methoden (in de vorm van FTIR) om tand geëmailleerde zijn wijd beschikbaar gesteld aan een gevarieerd publiek van academische. Terwijl wij hopen dat onze inspanningen zal deze aanpak gemakkelijker toegankelijk maken voor een groter aantal personen en laboratoria, onderzoekers die wilt toepassen en publiceren deze techniek moet rekening houden met minimale rapporteringstandaarden, diagenetic overwegingen, en presentatie eisen overviewed elders20, alsmede potentiële interpretatieve complexiteiten die uniek zijn voor de regio van hun studie, taxa geanalyseerd, en tijd periode5.

Stabiele koolstof en zuurstof isotoop analyse van mens en dier tandglazuur carbonaat is toegepast, in paleodietary, paleoecological, en paleomilieu onderzoek uit recente historische perioden terug naar meer dan 10 miljoen jaar geleden. Bulk benaderingen bieden een representatieve steekproef voor de periode van glazuur mineralisatie, terwijl opeenvolgende monsters binnen een tand dieet- en milieu wijzigingen tijdens deze periode bijhouden kunnen. Hoewel deze methodologieën zijn algemeen toegepast en beschreven in Archeologie, ecologie en paleontologie, zijn er geen expliciete richtlijnen om te helpen bij de keuze van apparatuur van het laboratorium nodig en grondig beschrijven gedetailleerde laboratorium bemonstering en protocollen. In dit artikel documenteren we tekstueel en visueel, het gehele proces van monsterneming door voorbehandeling en diagenetic screening de methodologie breder ter beschikking te stellen onderzoekers overwegen de toepassing in een verscheidenheid van laboratorium-instellingen.

Met behulp van de hierboven weergegeven monstertrekkingsprocedure, werden incrementele emaille bioapatite monsters voorbereid.
