Polyhistidine タグ付きタンパク質の固定化した卓上型金属アフィニ ティー ・ クロマトグラフィー (IMAC) は大学生によって習得されて簡単と現代文献の最も広く利用された蛋白質の浄化方法となっています。しかし、金属結合蛋白質、鉄などの酸化還元敏感な金属との特にそれらへのアフィニ ティー ・ クロマトグラフィーの適用が多い所が学部で定期的に利用できない機器 - グローブ ボックスへのアクセスを実験室。この記事で示す分離、IMAC ポリ ヒスチジンのタグ付きの精製と金属イオンの溶解、レドックス活性、非ヘム鉄バインド extradiol ジオキシゲナーゼおよび様々 な基板を用いたジオキシゲナーゼのアッセイのベンチトップ メソッド濃度と酸素を飽和します。これらのメソッドは学生によって実行される、アクセス可能な主に学部機関で手頃な価格は、インストルメンテーションで学部教育と研究室で実装します。
