1. 肌肉 Microbiopsy

</ol>2. 细胞的治疗

<ol><li>从准备特定的培养基开始 DF20, 通过加入20% 的热灭活胎牛血清, 5 毫升青霉素 (1000)-链霉素 (1万µg/毫升), 和2.5 毫升的两性霉素 B (250 µg/毫升) 到一瓶500毫升的商业可用培养基与谷氨酰胺和苯酚红色。</li>

	<li>把瓶子里的细胞放在干冰上最后使用。</li>
</ol>3. 注射细胞
注: 两个人需要执行注射干细胞。
<ol><li>将细胞悬浮温度逐渐升高至室温。将悬浮液吸入2毫升注射器。</li>
	<li>用19克皮下注射针将 detomidine 10 µg/千克注入颈静脉, 使马镇静。等待5分钟, 以观察镇静的充分的作用, 由马的特征的顶头向下位置变得可看见。</li>
	<li>剪辑一个区域 20 x 10 厘米2在马的左喉区与一把剪刀, 如图 2所示。然后, 将 polyiodide 肥皂和酒精的外科擦洗用在修剪过的区域。</li>
	<li>放置一个灵活的标准视频内窥镜通过左鼻孔的马和推进它向鼻咽。调整内窥镜的位置, 直到得到杓软骨和会厌的完整视图。在手术过程中将内窥镜保持在这个位置, 并将视频内窥镜的屏幕转向操作者。</li>
	<li>将刺激/注射针连接到神经刺激器的负极。保持针的不育条件。将神经刺激器的正电极连接到电极贴片上, 粘在被修剪过的皮肤区域。</li>
	<li>将含有细胞悬浮液的注射器连接到注射线上, 预先填充管以追赶系统的空气。记住系统的体积, 并准备一个注射器与无菌盐水溶液的数量相同。</li>
	<li>如果不熟悉这一技术, 使用消毒5至7兆赫线性超声传感器, 以可视化的解剖结构在该地区的兴趣。使用无菌超声耦合凝胶提高图像质量。可视化喉部和在喉背区运行的血管。</li>
	<li>一旦熟悉该地区的解剖学, 介绍刺激/注射针向侧方面的喉。当接近喉的侧方面时, 在1赫兹刺激模式下启动神经刺激器, 电流为2毫安。</li>
	<li>在观察屏幕上的内窥镜视图时, 轻轻地将针头移向喉返神经的位置。一旦左杓软骨的绑架运动在视频屏幕上可见, 减少电流, 直到运动消失, 同时保持针到位。</li>
	<li>确定针的理想位置。考虑到 0.5 mA 的电机响应损失, 作为理想的距离神经。当杓软骨运动持续在低于 0.4 mA 的电流, 不要注入细胞, 因为这可能导致 intraneuronal 注射。</li>
	<li>当观察到 0.5 mA 的马达反应损失时, 注射含有 107自体干细胞的细胞悬浮液, 并用步骤3.6 中制备的盐水溶液冲洗注射线。这将注入其余的细胞悬浮液留在注射管。</li>
	<li>注射后的细胞, 取出针和内窥镜。</li>

nerve stimulator-guided injection of autologous stem cells near the equine left recurrent laryngeal nerve

在本研究中使用的细胞是根据以前发表的7协议编写的。所有分数的细胞都能 trilineage 分化, 表达 CD90 和 CD44, 没有表达 CD45 和 MHC II, 并具有克隆能力。15–25% 分数的细胞已被选择进行进一步的处理, 因为它们显示了最大的表达 CD90 和最大的增殖能力。
