この方法は、長期培養中のエンジニア血管成長およびリモデリングのダイナミクスのモニタリングなど、血管組織工学分野における重要な質問に答える手助けとなります。光コヘレンス断層撮影(OCT)を使用する主な利点は、設計された容器の構造特徴と改造プロセスを特徴付けるための、容易に入手可能でリアルタイムで非破壊的なイメージング戦略であるということです。この手順のデモンストレーションは、私の研究室の技術者であるシャンミン・リュウとウェンタオ・マでもあります。この手順を開始するには、テキストプロトコルDip PGAスキャフォールドに記載されているようにPGA足場を1分間1モル/リットル水酸化ナトリウムで作製し、メッシュの容量構造を調整する。その後、スキャフォールドを組織培養グレードの水にそれぞれ2分間3回浸します。毎回、スキャフォールドをティッシュペーパーで軽く軽く軽く乾かします。その後、1時間ブロワーでフードに足場を乾かします。OCTイメージング用のバイオリアクターとy-Junctionを組み立てるために、まず、自己開発したガラス円筒形バイオリアクター、PGAスキャフォールド、シリコーンチューブ、生体適合性チューブ、コネクタ、攪拌バー、および95%エタノールタンクに組み立てるための機器を2時間浸します。コネクタで接続されたバイオリアクターのサイドアームを通してPGA足場を引っ張るだけでなく、OCTガイドワイヤーを提供するために使用されるyジャンクションを持つ別の側に引っ張ります。同じようにバイオリアクターに別のPGA足場を組み立てます。ポリテトラフルオロエチレンを4-0縫合で締めてバイオリアクターの唇にフィットさせる。バイオリアクターをエタノールタンクに再び入れてから、ブロワーをつけてフードで一晩乾燥させます。さて、ヒト臍帯平滑筋細胞をヒト臍帯動脈から標準的な外植技術で単離する PGA足場に細胞を保存する原因として、バイオリアクターの底部に細胞懸濁液が滴り落ちるのを避ける。また、バイオリアクターに、汚染を防ぐため、細胞の座席後できるだけ早くシリコーンストッパーの蓋を覆います。平滑筋成長培地で細胞を拡大し、維持します。上記培養培地中の500万細胞/ミリリットルの濃度でヒト臍動脈平滑筋細胞をPGA足場に播種する。バイオリアクターにサーバーを配置した後、1つの給餌管と3つの短いチューブセグメントをガス交換用に挿入したシリコーンストッパーリッドをカバーします。各空気交換管にPTFE 0.22ミクロンフィルターを取り付け、1つのヘパリンキャップを給餌管に取り付けます。1分間に13ラウンドの攪拌速度で攪拌バーを調整します。次いで、ガラスバイオリアクター、シリコーンストッパー蓋、及びPGA足場を培養システムに組み立てる。細胞を左と右にスタンドで15分ごとに傾けて45分間接着させます。今度は、LOO-OH-YEEポンプ、リン酸緩衝生理食塩水袋、およびドライバーを生体適合性チューブに接続して、拡散システムを作成します。ドライバを開いて、チューブをPBSで満たします。全体のバイオリアクターを加湿インキュベーターに入れ、5%のCO2を摂氏37度で置きます。培養液を450ミリリットルで培養チャンバーに充填する。播種された足場を静的培養下で1週間増殖させ、古い培地の半分を給餌管を通して吸引し、反応器に同等量の新鮮な培養培地を補充することによって、3〜4日ごとに培養培地を変化させる。OCTイメージング用の拡散システムを準備するために、PBSバッグ内の流体をポンプで送り込み、生体適合性チューブを介してそれらを循環させ、バッグに戻します。ドライバーのパワーを開き、毎分60拍の周波数と水銀の120ミリメートルの出力収縮期圧力でポンプ設定を調整します。組織工学血管培養のニーズに応じて機械的パラメータを調整します。実行ボタンをクリックして、拡散システムを動作させます。静的培養の1週間後に生体適合性チューブを繰り本気で加圧することで、3週間の間、固定拍動シミュレーションを血管に提供します。光源を使用して、10~20ミクロンの軸方向解像度と1〜2ミリメートルの画像深さを確保し、周波数領域OCT血管内イメージングシステムに基づいて組織操作された血管(TEVB)の構造を特定します。電源スイッチをオンにしてイメージキャプチャソフトウェアを開きます 光ファイバーイメージングカテーテルを、カテーテルの自動リトリート機能でドライブモーターと光コントローラに接続します。画像取得レートのパラメータを 10 フレーム/秒に設定し、自動プルバック速度は 10 ミリ/秒に設定します。次に、18ゲージの針でヘパリンキャップを介してy-Junctionにイメージングカテーテルを取り付けます。カテーテルをシリコンチューブに入れ、PGAスキャフォールドをバイオリアクターにロードする前に、PGAメッシュの構造密閉性を特定します。次に、カテーテル先端を対象領域の上に置きます。プルバックデバイスを調整し、画質を確認します。個々の TEBV の 1、4、7、10、14、17、21、28 日のカルチャで画像を取得します。表面形態、内部構造、組成を含むTEBV微細構造のリアルタイム観察で、これらの画像を順次保存します。測定を3回繰り返して、毎回操作した容器の信頼性の高い測定を行います。イメージ キャプチャ ソフトウェアを使用して、テスト全体を通して一連のイメージをキャプチャします。画像解析ソフトウェアを使用して組織設計された血管壁の厚さを測定するには、まず解析する画像を選択します。次に、追跡ツールをクリックして、組織で設計された血管の内側をソフトウェアで自動的に識別し、手動で外側をスケッチします。画面に厚さの図が表示されます。5回測定を繰り返して、構築物の信頼性の高い測定を行います。取得した情報に義務付けられている 2 人の独立した調査員を使用することを検討してください。培養が終了したらバイオリアクターの上に置かれたシリコーンストッパー蓋を開け、培養液を廃棄する。バイオリアクターの唇からポリテトラフルオロエチレンを緩め、ポリテトラフルオロエチレンの外側からシリコーンチューブをはさみで切ります。バイオリアクターからTEBVを収穫し、走査電子顕微鏡検査のために切り分けに切ります。支持するシリコーンチューブを引き出し、4%のパワーホルムアルデヒドでセクションを固定します。コラーゲンとPGAの形態を調べるために、マッソンのトリクロームとサーシャスレッドで日常的な組織学的染色を行います。PGAの含有量とコラーゲン成分を評価するには、偏光顕微鏡を通してシリの赤い染色で組織学的サンプルを観察します。OCT画像は、4週間培養後のTEBの病理組織学的発見と比較される。OCT画像は、組織学的評価と比較して、コンパクトな微細構造および特定の成分を示しています。培養培地には、シリコーンチューブ、TEBV、およびPGA断片が見える。マッソンのトリクローム染色は、一定の方向に分布するコラーゲン繊維と、設計された血管のメディア層のPGA残骸を示しています。シリウスの赤染色は、偏光顕微鏡を用いてPGA残骸とコラーゲン成分を明らかにする。工学的血管の走査電子顕微鏡写真は、コンパクトな微細構造を示しています。一方、このイントラミナルイメージングモダリティは、長期にわたる文化におけるリモデリングプロセスや視覚形態を含むTEBVの非破壊かつ容易なモニタリングを採用しています。ここに示すように、改変は早く起こり、形態学的変化はTEBV厚さの比較によって動的群でより明らかに現れる。この技術は、血管組織工学の分野の研究者が構造特徴と設計された血管の長期的な改造プロセスを探求する道を開きます。偏光顕微鏡とOCTイメージングを組み合わせたポリマー残骸の明確な表示は、足場の劣化を評価するのに有用である可能性があります。
