De hier geschetste protocol kan worden gebruikt om krachtig en reproducibly kwantificeren cel biologische processen gevisualiseerd door fluorescentie gebaseerde imaging. Biologische context en technische beperkingen moeten zorgvuldig worden overwogen bij de proefopzet. Neurodegeneratieve ziekten miljoenen mensen elk jaar en de incidentie neemt toe met een verouderende bevolking1. Terwijl elke neurodegeneratieve ziekte heeft een unieke etiologie, zijn aggregatie van eiwitten van de misfolded en de verdeling van het proteostasis-netwerk gemeenschappelijk pathologische kenmerken van veel van deze ziekten. Ophelderen hoe verstoring van deze fundamentele en onderling verbonden processen misgaat is om bij te dragen tot de neuronale cel en dysfunctie dood essentieel voor het begrip van neurodegeneratieve ziekten, alsmede het begeleiden van therapeutische interventies. Fluorescentie gebaseerde beeldvorming voorziet in onderzoek van deze complexe en dynamische processen in neuronen en heeft bijgedragen aan ons begrip van de neuronale celbiologie. Analyse van fluorescently tagged eiwitten is uitdagend, met name wanneer de experimenten zijn uitgevoerd in vivo, als gevolg van de zeer compacte weefsels, diverse celtypes en morfologische heterogeniteit. Handmatig geassisteerde kwantificering is betaalbaar en eenvoudig, maar is vaak tijdrovend en onder voorbehoud van menselijke bias. Daarom is er behoefte aan een onpartijdige, reproduceerbaar zijn, en toegankelijk benaderingen voor het extraheren van meetbare gegevens van imaging studies.
We hebben een eenvoudige en flexibele workflow met behulp van Fiji/ImageJ, een krachtige en vrij toegankelijke beeldverwerking software2,3, kwantitatieve om gegevens te extraheren uit imaging fluorescentiestudies in experimentele modellen van geschetst neurodegeneratie met behulp van Drosophila. Door het volgen van dit protocol om te kwantificeren aggregatie van eiwitten en autophagic flux — twee biologische functies die zeer relevant voor neurodegeneratieve ziekte pathologie zijn cel — we de gevoeligheid en de reproduceerbaarheid van deze aanpak aangetoond. Onderzoek van fluorescently tagged mutant huntingtin (Htt) eiwitten in de Drosophila optic kwab bleek het aantal, de grootte en de intensiteit van de eiwit-aggregaten. Wij een tandem fluorescerende verslaggever van autophagic flux binnen het visuele systeem van Drosophila , waarin verschillende emissie signalen afhankelijk van de compartimentaire omgeving4gevisualiseerd. Ratiometric gebaseerde analyse van de tandem verslaggever toegestaan voor een kwantitatieve en compleet beeld van autophagy-lysosoom flux van autophagosome formatie, rijping en vervoer tot degradatie in het lysosoom en daarnaast gemarkeerd kwetsbaar compartimenten verstoord in neurodegeneratieve omstandigheden. Nog belangrijker is, in beide analyses uitgevoerd we semi-geautomatiseerde drempelmethode en segmentatie stappen in ons protocol aan onbewuste bias te minimaliseren, vergroten van de macht van de bemonstering en bieden een standaard om vergelijkingen tussen soortgelijke studies. De eenvoudige werkstroom is bedoeld om krachtige Fiji/ImageJ plugins (ontwikkeld door informatici die zijn gebaseerd op wiskundige algoritmen) toegankelijker voor neurobiologists en de biowetenschappen Gemeenschap in het algemeen.

Met de stijgende prevalentie van neurodegeneratieve ziekten is het steeds belangrijker om te begrijpen van de onderliggende pathofysiologie, dat tot neuronale disfunctie en verlies leidt.
