1-iodoalcynes, 1, 2-diiodoalkenes et 1,1,2-trichlorotrifluoroéthane-triiodoalkenes peuvent être chimiosélective synthétisé utilisant des réactifs d’iode hypervalent comme un médiateur efficace pour iodination(s) oxydatif. Les facteurs les plus critiques de ces protocoles d’iodation chimiosélective sont la nature et le chargement de la source d’iode, ainsi que le solvant. Iodoalcynes et iodoalkenes sont des précurseurs importants largement utilisés et blocs de construction en synthèse organique1,2,3,4, substances biologiquement actives et utiles dans la synthèse de matériaux et molécules complexes, étant donnés la facilité de conversion de la C-j’ai coller5,6,7,8. Ces dernières années, l’iodation oxydative des alcynes terminaux a attiré plus d’attention à la synthèse de dérivés iodoalkyne et iodoalkene. Jusqu'à présent, efficaces des méthodes qui utilisent des catalyseurs métalliques9,10,11,12, hypervalents-iodonium catalyseurs13,14, un système d’oxydation anodique 15, systèmes liquides ioniques16, KI (ou j’ai2)-oxydant combinaisons17,18,19,20, échographie21, catalyseurs de transfert de phase 22, N- iodosuccinimide9,22,23,24,25, n- BuLi26,27, 28 , 29 , 30 , 31et de réactifs de Grignard32morpholine catalyseurs17,33,24,35 ont été développés pour l’iodation des alcynes. Récemment, nous avons rapporté un protocole chimiosélective et pratique pour la synthèse de 1-iodoalcynes, 1, 2-diiodoalkenes et 1,1,2-triiodoalkenes36. Les caractéristiques de cette méthode sont vertes et pratique : (1) la toxicité des catalyseurs de l’iode hypervalent comme réactifs de fonctionnalisation oxydatif est faible par rapport aux autres oxydants classiques de base heavy-metal37,38, 39,40,41,42et (2) TBAI et/ou KI est utilisé comme sources d’iode. En outre, notre système offre excellente sélectivité dans des conditions douces. La synthèse de chimiosélective du 1-iodoalcynes, 1, 2-diiodoalkenes et 1,1,2-trichlorotrifluoroéthane-triiodoalkenes nécessite un contrôle précis sur les divers facteurs, dont la composition, l’oxydant, la source de l’iode et le solvant. Parmi ceux-ci, la source de l’iode est le facteur le plus important pour la chimiosélectivité de la réaction. Après la projection de plusieurs types et charges de la source d’iode ainsi que les solvants, les trois méthodes ont été identifiés et mis en place. Tout d’abord, TBAI comme source d’iode en combinaison avec PIDA (TBAI-PIDA) est sélectif pour la synthèse du 1-iodoalcynes. Par ailleurs, 1, 2-diiodoalkenes sont obtenus efficacement en utilisant un système de KI-PIDA. Les deux méthodes s’offrir les produits correspondants à haut rendement et haute chimiosélectivité. Le tri-iodinationproducts correspondant, c'est-à-dire., 1,1,2-triiodoalkenes, ont été obtenus avec un bon rendement de la synthèse monotope qui combinent la TBAI-PIDA et KI-PIDA systèmes36.
Ici, nous allons démontrer comment la chimiosélectivité pour l’iodation des alcynes terminaux peut être orientée de 1-iodoalcynes de 1, 2-diiodoalkenes et de 1,1,2-triiodoalkenes dans des conditions de réaction similaires, mettant en évidence le contrôle précis qui peut être exercée en choisissant judicieusement oxydant, source d’iode et solvant. Pour le développement de cette nouvelle technique synthétique, p- tolylethyne a été utilisé comme substrat modèle. Bien que les protocoles suivants se concentrent sur la synthèse de 1-(iodoethynyl) -4-méthylbenzène, (E) -1-(1,2-diiodovinyl)-4-méthylbenzène et 1-méthyl - 4-(1,2,2-triiodovinyl) benzène, ces composés sont représentatifs pour 1-iodoalcynes, 1, 2 - diiodoalkenes et 1,1,2-trichlorotrifluoroéthane-triiodoalkenes, respectivement, par exemple, les protocoles sont larges dans le champ d’application, et la même technique peut être appliquée à l’iodation chimiosélective des alcynes terminaux aromatiques et aliphatiques,36.
Réactifs utilisés dans l’iodation chimiosélective des alcynes terminaux et petites déviations par les techniques décrites entraînent des différences spectaculaires en ce qui concerne les produits de la cible. Par exemple, changer de source d’iode de TBAI à KI et changeant de solvant de CH3CN à un CH3CN-H2O a un impact dramatique sur la chimiosélectivité de l’iodation. Le protocole détaillé vise à aider les nouveaux praticiens dans le domaine avec l’iodation chimiosélective des alcynes terminaux afin d’éviter les nombreux pièges communs au cours de la synthèse du 1-iodoalcynes, 1, 2-diiodoalkenes et 1,1,2-trichlorotrifluoroéthane-triiodoalkenes.

chemoselective preparation of 1-iodoalkynes, 1,2-diiodoalkenes, and 1,1,2-triiodoalkenes based on the oxidative iodination of terminal alkynes

Nous présentons la synthèse chimiosélectif de 1-(iodoethynyl) -4-méthylbenzène, 1-(1,2-diiodovinyl)-4-méthylbenzène et 1-méthyl - 4-(1,2,2-triiodovinyl) benzène comme exemples représentatifs pour la préparation pratique chimiosélective de 1-iodoalcynes , 1, 2-diiodoalkenes et 1,1,2-trichlorotrifluoroéthane-triiodoalkenes de l’iodation de chimiosélective des alcynes terminaux médiée par les réactifs hypervalents-iode. La chimiosélectivité a été confirmée à l’aide de p- tolylethyne comme substrat modèle pour dépister une variété de sources d’iode et/ou les réactifs hypervalents-iode. Une combinaison de tetrabutylammonium iodure (TBAI) et (diacétoxyiodo) benzène (PIDA) génère sélectivement 1-iodoalcynes, alors qu’une combinaison de KI et PIDA génère 1,2-diiodoalkenes. Une synthèse one-pot basée sur deux TBAI-PIDA et KI-PIDA donne le triiodoalkenes-1,1,2 correspondant. Ces protocoles ont été ensuite appliqués à la synthèse de synthétiquement important aromatiques et aliphatiques 1-iodoalcynes, 1, 2-diiodoalkenes et 1,1,2-triiodoalkenes, qui ont été obtenus avec un bon rendement avec excellent chimiosélectivité.
