1. syntese av 1-(Iodoethynyl) -4-Methylbenzene (2, 1-Iodoalkynes)

Iodoalkynes og iodoalkenes er mye brukt viktig prekursorer og byggesteinene i Organisk syntese1,2,3,4, biologisk aktive stoffer, og nyttig i syntesen av materialer og komplekse molekyler gitt enkel konvertering C-jeg bånd5,6,7,8. De siste årene, har den oksidativt iodination av terminal alkynes fått mer oppmerksomhet til syntese av iodoalkyne og iodoalkene derivater. Så langt, effektive metoder som bruker metall katalysatorer9,10,11,12, hypervalent-iodonium katalysatorer13,14, en anodic oksidasjon system 15, ioniske flytende systemer16, KI (eller2)-oksiderende kombinasjoner17,18,19,20, ultralyd21, fase-transfer katalysatorer 22, N- iodosuccinimide9,22,23,24,25, n- BuLi26,27, 28 , 29 , 30 , 31og Grignard reagenser32morpholine katalysatorer17,33,24,35 har blitt utviklet for iodination av alkynes. Vi har nylig rapportert en praktisk og chemoselective protokoll for syntese av 1-iodoalkynes, 1,2-diiodoalkenes og 1,1,2-triiodoalkenes36. Funksjonene i denne metoden er grønne og praktiske: (1) toksisitet av hypervalent-jod katalysatorer som oksidativt functionalization reagenser er lav sammenlignet med andre konvensjonelle heavy-metal-baserte oksidanter37,38, 39,40,41,42, og (2) TBAI og/eller KI brukes som jod kilder. I tillegg gir systemet utmerket selektivitet under mild forhold. Chemoselective syntese av 1-iodoalkynes, 1,2-diiodoalkenes og 1,1,2-triiodoalkenes krever nøyaktig kontroll over forskjellige faktorer, inkludert sammensetningen, oksiderende, jod kilden og løsemiddelet. Blant disse er jod kilden den viktigste faktoren for chemoselectivity av reaksjonen. Etter screening av flere typer og belastninger av jod kilden samt løsemidlene, ble tre metoder identifisert og etablert. For det første er TBAI som en jod i kombinasjon med PIDA (TBAI-PIDA) selektiv for syntese av 1-iodoalkynes. Alternativt oppnås 1,2-diiodoalkenes effektivt ved å bruke en KI-PIDA. Begge metodene råd til de tilsvarende produktene i høy kapasitet og høy chemoselectivity. Den tilsvarende tri-iodinationproducts, dvs., 1,1,2-triiodoalkenes, ble oppnådd i god avkastning fra en-potten syntese som kombinerer de TBAI-PIDA og KI-PIDA systemer36.
Her, vil vi demonstrere hvordan chemoselectivity for iodination av terminal alkynes kan styres fra 1-iodoalkynes 1,2-diiodoalkenes og 1,1,2-triiodoalkenes under lignende reaksjon forhold, utheving nøyaktig kontroll som kan utøves av judiciously velge oksiderende, jod kilde og løsemidler. For utvikling av denne nye syntetiske teknikken, ble p- tolylethyne brukt som en modell substrat. Selv om følgende protokoller fokusere på syntesen av 1-(iodoethynyl) -4-methylbenzene, (E) -1-(1,2-diiodovinyl)-4-methylbenzene, og 1-metyl - 4-(1,2,2-triiodovinyl) benzen, disse forbindelsene er representative for 1-iodoalkynes, 1,2 - diiodoalkenes, og 1,1,2-triiodoalkenes, henholdsvis, dvs, protokollene er bredt i omfang, og de samme teknikkene kan brukes på chemoselective iodination av aromatiske og alifatisk terminal alkynes36.
Reagenser ansatt i chemoselective iodination av terminal alkynes og små avvik fra teknikker beskrevet resultere i dramatiske forskjeller når det gjelder målet produktene. For eksempel har endring av jod kilde fra TBAI KI og endre løsemiddel CH3CN til CH3CN-H2O en dramatisk innvirkning på chemoselectivity av iodination. Detaljert protokollen tar sikte på å hjelpe nye utøvere i feltet med chemoselective iodination av terminal alkynes å unngå mange vanlige fallgruvene under syntesen av 1-iodoalkynes, 1,2-diiodoalkenes og 1,1,2-triiodoalkenes.

Vi presenterer chemoselective syntese av 1-(iodoethynyl) -4-methylbenzene, 1-(1,2-diiodovinyl)-4-methylbenzene og 1-metyl - 4-(1,2,2-triiodovinyl) benzen som eksemplene for praktisk chemoselective utarbeidelse av 1-iodoalkynes , 1,2-diiodoalkenes og 1,1,2-triiodoalkenes fra chemoselective-iodination av terminal alkynes formidlet av hypervalent-jod reagenser. Chemoselectivity ble bekreftet ved hjelp av p- tolylethyne som en modell substrat for å skjermen en rekke jod kilder og/eller hypervalent-jod reagenser. En kombinasjon av tetrabutylammonium iodide (TBAI) og (diacetoxyiodo) benzen (PIDA) genererer selektivt 1-iodoalkynes, mens en kombinasjon av KI og PIDA genererer 1,2-diiodoalkenes.
