Deze bepaling (of soortgelijke testen waar andere ziekteverwekkers worden vervangen door P. aeruginosa of E. faecalis) is handig voor allerlei doeleinden, met inbegrip van Geneesmiddelenontwikkeling. De identificatie en ontwikkeling van een effectief, breedspectrum antibiotica, inmiddels bijna een eeuw geleden, geleid tot een moment van de waterscheiding in de volksgezondheid waar er een wijdverbreide geloof dat besmettelijke ziekte zou een gesel van het verleden. Binnen een paar korte decennia begon dit optimisme te afnemen, als pathogeen nadat pathogen resistentie mechanismen die deze keer miraculeuze behandelingen beperkt ontwikkeld. Voor enige tijd leek de wapenwedloop tussen drug discovery inspanningen en de ziekteverwekkers evenwichtig. Echter, het misbruik van antimicrobiële middelen onlangs heeft geresulteerd in het ontstaan van pan-resistente stammen van Klebsiella pneumoniae, immunodeficientie baumanii, Serratia marcescensen P. aeruginosa1, 2,3,4.
P. aeruginosa is een opportunistische, gram negatieve, meerdere host pathogenen die een ernstige bedreiging vormt voor patiënten met ernstige brandwonden, degenen die zijn immuungecompromitteerde, of cystic fibrosis. Het wordt ook in toenemende mate aangeduid als een ziekteverwekker in ernstige ziekenhuisinfecties, met name als gevolg van de voortdurende overname van antimicrobiële resistentie. Om te beginnen met het inspelen op deze dreiging, hebben we de goed gedocumenteerde C. elegans-P. aeruginosa infectie systeem5gebruikt. Onze lab heeft dit systeem voor de ontwikkeling van een vloeistof gebaseerde, high-throughput, hoge-content screening platform ter identificatie van nieuwe verbindingen die de mogelijkheid van het pathogene agens te doden de host6beperken leveraged. Deze verbindingen lijken intrigerend, tot ten minste drie algemene categorieën, met inbegrip van antimicrobiële stoffen7 en virulentie remmers8te behoren. Andere hoge-inhoud drug discovery testen in C. elegans voor Mycobacterium tuberculosum, Chlamydia trachomatis, Yersinia pestis, Listeria monocytogenes, Francisella tularensis, Staphylococcus aureus, Candida albicans, zijn gerapporteerd en Enterococcus faecalis, o.a.9,10,11,12,13,14,15,16. Deze types van tests hebben verschillende goed erkende voordelen, zoals het beperken van vals positieve hits die mogelijk toxisch voor zowel de host en de pathogenen, verhoogde kans op biologische beschikbaarheid ten opzichte van een chemische scherm, en het vermogen om te identificeren hits voorbij microbiële groei, zoals anti-virulents, immuun stimulerend moleculen of verbindingen die anders het kantelen van het saldo van de host-pathogen interactions ten gunste van de voormalige gewoon te beperken. Bovendien zijn de verbindingen ontdekt in deze schermen vaak effectief in zoogdieren hosts.
Het is vermeldenswaard dat ten minste twee andere testen17,18 zijn beschikbaar voor het verrichten van high-throughput schermen in C. elegans in vloeistof. Elk van deze tests is echter een wijziging waarmee de prototypische intestinale-kolonisatie assay, bekend als traag-doden, moet worden uitgevoerd in vloeibare, verhoging van de doorvoer en waardoor verbindingen gemakkelijker worden vertoond. Zorgvuldige karakterisering overtuigend heeft aangetoond dat de mechanismen van bacteriële virulentie tussen deze testen en onze vloeistof gebaseerde scherm7 verschillen. Aangezien beide soorten virulentie bij zoogdieren systemen worden waargenomen, is het belangrijk om na te gaan welke virulentie determinant meest relevant voor de de experimentator belangen vóór assay selectie.
Hier tonen we een geoptimaliseerde versie van de vloeistof gebaseerde C. elegans-P. aeruginosa assay. Ook melden wij de aanpassing van onze assay vloeistof gebaseerde methode om de gram-positieve bacteriële pathogenen Enterococcus faecalis. E. faecalis wordt met een toenemende bewapening van antimicrobiële resistentie trajecten1steeds meer als een bedreiging van ernstige nosocomiale geïdentificeerd zoals P. aeruginosa. Hoewel een vorige methode voor high-throughput screening van E. faecalis 14 bestaat, vereist het preinfection met het pathogene agens oplevert, die compliceert de procedure en verhoogt de kans op besmetting van apparatuur zoals de COPAS-FlowSort. Ons protocol elimineert de noodzaak voor pre-infectie, verbetering van de veiligheidsprofiel. Tot slot melden wij een middel waarmee ofwel van deze tests kunnen worden gecombineerd met het voeden van RNAi, waardoor de gebruiker kan zoeken voor gastheer factoren die een rol in de oprichting van spelen, of weerstand tegen infectie.

a high-throughput, high-content, liquid-based c. elegans pathosystem

Het aantal nieuwe drugs geïdentificeerd door traditionele, in vitro schermen is afgenomen, vermindering van het succes van deze aanpak in de zoektocht naar nieuwe wapens ter bestrijding van meerdere resistentie. Dit heeft geleid tot de conclusie dat onderzoekers niet alleen maar hoeft te vinden van nieuwe geneesmiddelen, maar ook ontwikkelen van nieuwe manieren moeten van het vinden van hen. Onder de meest veelbelovende kandidaat methoden zijn geheel-organisme, in vivo onderzoek dat gebruik high-throughput, fenotypische uitlezingen en gastheren variërend van Caenorhabditis elegans tot Danio rerio. Deze hosts hebben verschillende krachtige voordelen, met inbegrip van dramatische vermindering valse positieve hits, zoals verbindingen die giftig voor de host en/of de biounavailable zijn vallen meestal in het eerste scherm, voorafgaand aan dure follow up.
Hier laten we zien hoe onze test is gebruikt om het ondervragen van host variatie in de goed gedocumenteerde C. elegans—Pseudomonas aeruginosa vloeibare doden pathosystem. We tonen ook diverse uitbreidingen van deze techniek goed uitgewerkt. Wij zijn bijvoorbeeld kunnen uitvoeren van high-throughput genetische schermen met behulp van RNAi in 24 - of 96-wells plaat formaten naar query gastheer factoren in deze host-pathogen interactions. Met behulp van deze test, kunnen hele genoom schermen worden uitgevoerd in slechts een paar maanden, die de taak van de identificatie van de doelstellingen van de drug, mogelijk zonder de noodzaak voor een aanpak van de moeizame biochemische zuivering dramatisch kunnen vereenvoudigen.
Ook rapporteren we hier een variatie op onze methode die de gram-positieve bacterie Enterococcus faecalis voor de gram-negatieve pathogen P. aeruginosa vervangt. Veel zoals het geval voor P. aeruginosa, is doden door E. faecalis afhankelijk van de tijd. In tegenstelling tot eerdere C. elegans—E. faecalis testen, onze assay voor E. faecalis geen preinfection vereist, verbetering van haar veiligheidsprofiel en vermindering van de kans op besmetting van de vloeistof-behandelingsapparatuur. De bepaling is zeer robuust, ~ 95% sterftecijfers 96 h post infectie tonen.

Belangrijke parameters voor assay prestaties
Een goed begrip van de biologie die ten grondslag liggen aan deze bepaling is noodzakelijk voor het oplossen van problemen en het optimaliseren van de bepaling.
