fPhycobiliproteins zijn wateroplosbaar pigment-eiwitcomplexen die belangrijke componenten van de antennes licht-oogst in prokaryotische cyanobacteriën (Cyanophyceen) en diverse eukaryotic taxa (Glaucophyta, Rhodophyta vertegenwoordigen , en Cryptophyta)1. Ze ontstaan voornamelijk supramoleculaire complexen genaamd phycobilisomes en ze zijn meestal gekoppeld aan het oppervlak van de fotosynthetische membranen op de stromale kant, met uitzondering van Cryptophyta, waar de phycobiliproteïnen zijn gelokaliseerd in de thylakoïde lumen2. Vier soorten phycobiliproteïnen up-to-date zijn geïdentificeerd: de kern-allophycocyanin, en de perifere phycocyanin, phycoerythrin en phycoerythrocyanin1. Als de belangrijkste licht-oogst complexen vormen phycobilisomes een van de cruciale factoren van algen en cyanobacteriën massa culturen productiviteit. Dit protocol is gericht op de kwantitatieve bepaling van phycobiliproteïnen in het model cyanobacterium Synechocystis. Blauwalgen zijn de vroegste Oxygene fotosynthetische autotrophs; ze hebben de vorming van de aardse biosfeer voor meer dan 2,4 miljard jaar6. Ze spelen een cruciale rol in wereldwijde biogeochemische cycli van koolstof, stikstof, zuurstof en andere elementen. Dus, de specifieke golflengten en absorptie coëfficiënten zoals gebruikt voor de bepaling van phycocyanin en allophycocyanin in Synechocystis gelden niet in het algemeen aan andere stammen. Bovendien bevat geen Synechocystis phycoerythrin en phycoerythrocyanin die worden in sommige andere algen en cyanobacteriën gevonden kan. Met het oog op de bepaling van phycobiliproteïnen in stammen dan Synechocystis, is het aanbevolen om de spectrofotometrische vergelijkingen voor elke soort afzonderlijk evalueren.
Hoewel het protocol bevat twee langere stappen ('s nachts vriesdrogen van de cellulaire pellets en 1 uur eiwit extractie), is de tijd van de totale arbeid voor de kwantificering van phycobiliproteïnen niet dan 2 uur.

spectrophotometric determination of phycobiliprotein content in cyanobacterium synechocystis

Dit is een simpel protocol voor de kwantitatieve bepaling van phycobiliprotein in het model cyanobacterium Synechocystis. Phycobiliproteïnen zijn de belangrijkste onderdelen van de phycobilisomes, het grote licht-oogst antennes in cyanobacteriën en verschillende taxa van de algen. De phycobilisomes van de Synechocystis bevatten twee phycobiliproteïnen: phycocyanin en allophycocyanin. Dit protocol beschrijft een eenvoudige, efficiënte en betrouwbare methode voor de kwantitatieve bepaling van zowel phycocyanin en allophycocyanin in dit model cyanobacterium. We vergeleken de verschillende methoden van phycobiliprotein extractie en spectrofotometrische kwantificering. De extractie procedure zoals beschreven in dit protocol was ook met succes toegepast voor andere cyanobacteriën stammen zoals Cyanothece sp., Synechococcuselongatus, Spirulina sp., Arthrospira sp., en Nostoc sp., alsmede over de rode algen Porphyridium cruentum. Echter de coëfficiënten uitsterven van specifieke phycobiliproteïnen van verschillende taxa kunnen verschillen en het is daarom aangeraden voor het valideren van de methode spectrofotometrische kwantificering voor elke één stam individueel. Het protocol vereist weinig tijd en kan worden uitgevoerd in een laboratorium van de wetenschap van het leven van de standaard aangezien het vereist alleen standaard uitrusting.

Voor de eerste methode tests, was Synechocystis als batch culturen in conische kolven op een shaker gekweekt in BG11 teelt middellange20 (aangevuld met 17 mM HEPES) bij 25 ° C, onder een warm wit licht met een intensiteit van 50 µmol (fotonen) / (m 2·s) en met 1% CO2 in de kweken atmosfeer.
