fPhycobiliproteins är vattenlösliga pigment-proteinkomplex som representerar stora delar av ljus-skörd antenner i prokaryota cyanobakterier (Cyanophyta) och flera eukaryota taxa (Glaucophyta, Rhodophyta , och Cryptophyta)1. De förekommer främst som Supramolekylära komplex kallas phycobilisomes och de är oftast knutna till ytan av fotosyntetiska membran på stromaceller sida, med undantag för Cryptophyta, där phycobiliproteins är lokaliserade i de tylakoida lumen2. Fyra typer av phycobiliproteins har identifierats upp till datum: den core allophycocyanin och perifera phycocyanin, phycoerythrin och phycoerythrocyanin1. De har en avgörande roll i globala biogeokemiska cykler av kväve, kol, syre och andra element. Flera metoder för phycobiliprotein utvinning och kvantifiering jämförs i detta protokoll. Därför, den specifika våglängder och absorption koefficienter som används för bestämning av phycocyanin och allophycocyanin i Synechocystis är inte allmänt tillämpliga för andra stammar. Dessutom innehåller inte Synechocystis fykoerytrin och phycoerythrocyanin som kan hittas i vissa andra alger och cyanobakterier. För bestämning av phycobiliproteins i stammar än Synechocystisrekommenderas det att utvärdera spektrofotometrisk ekvationerna för varje stam individuellt.

spectrophotometric determination of phycobiliprotein content in cyanobacterium synechocystis

Detta är ett enkelt protokoll för kvantitativ bestämning av phycobiliprotein innehåll i den modell Cyanobakterien Synechocystis. Phycobiliproteins är de viktigaste komponenterna i phycobilisomes, stora ljus-skörd antenner i cyanobakterier och flera alger taxa. Phycobilisomes av Synechocystis innehåller två phycobiliproteins: phycocyanin och allophycocyanin. Det här protokollet beskriver en enkel, effektiv och tillförlitlig metod för kvantitativ bestämning av både phycocyanin och allophycocyanin i denna modell Cyanobakterien. Vi jämförde flera metoder för phycobiliprotein utvinning och spektrofotometrisk kvantifiering. Extraktionsmetod som beskrivs i detta protokoll tillämpades också till andra cyanobakterier stammar såsom Cyanothece sp., Synechococcuselongatus, Spirulina sp., Arthrospira sp., och Nostoc sp., samt att röda alger Porphyridium cruentum. Dock utrotning koefficienter av specifika phycobiliproteins från olika taxa kan variera och därför rekommenderas att validera metoden spektrofotometrisk kvantifiering för varje enskild stam individuellt. Protokollet kräver lite tid och kan utföras i ett laboratorium som standard life science eftersom det kräver endast standardutrustning.

För den ursprungliga metod för proven Synechocystis odlades som batch kulturer i Erlenmeyer-kolvar på en shaker i BG11 odling medium20 (kompletteras med 17 mM HEPES) vid 25 ° C, under ett varmvitt ljus av en stödnivå på 50 µmol (fotoner) / (m 2·s) och med 1% CO2 i culturing atmosfären.
