At identificere genetiske komponenter af necroptosis har fremmet brugen af dyremodeller for at afprøve implikationen af necroptosis i fysiologi og sygdom1,2,3,4,5. Knockout af RIPK3 eller MLKL i mus havde minimal implicit i udvikling og voksen homøostase tyder på, at necroptosis ikke er afgørende for liv3,6. characterization of mlkl-mediated plasma membrane rupture in necroptosis

Necroptosis er en programmeret celle død pathway udløst ved aktivering af receptor interaktion protein kinase 3 (RIPK3), som phosphorylates og aktiverer blandet lineage kinase-lignende domæne pseudokinase, MLKL, at briste eller permeabilize plasmamembran . Necroptosis er en inflammatorisk pathway forbundet med flere patologier herunder autoimmunitet, smitsomme og hjerte-kar-sygdomme, slagtilfælde, neurodegeneration og kræft. Her beskriver vi protokoller, der kan bruges til at karakterisere MLKL som bøddel af plasma membran brud i necroptosis. Vi visualisere processen med necroptosis i celler ved hjælp af live-celle imaging med konventionelle og konfokal Fluorescens mikroskopi og faste celler ved hjælp af elektronmikroskopi, som sammen afslørede en omfordeling af MLKL fra cytosol til plasma membran før induktion af store huller i plasma membranen. Vi præsenterer i vitro Kernemagnetisk resonans (NMR) analyse ved hjælp af lipider for at identificere formodede modulatorer af MLKL-medieret necroptosis. Baseret på denne metode, identificeret vi kvantitative lipid-bindende præferencer og phosphatidylphosphatid-inositol fosfater (kerner) som kritiske bindemidler af MLKL, der er nødvendige for plasma membran målretning og permeabilization i necroptosis.

Visualisere reguleret necroptosis udførelse i levende celler har været muligt via inducerbar udtryk for en minimal afkortet MLKL konstruktion, NBB140-2xFV-Venus. Denne konstruktion bevarer evnen til at fremkalde plasma membran permeabilization og aktiveres via Dim-induceret oligomerisering af FKBP kassette (2xFV).
