Aqui, apresentamos um método rápido e sensível para a utilização de citometria de fluxo para medir a atividade celular mitophagy em células expressando mt-Keima. Passando por um elevado nível de mitophagy de células apresentam uma maior proporção de PE-CF594 (561 nm) / BV605 (405 nm) excitação. Assim, a atividade mitophagy pode ser expressa como a porcentagem de células exibindo um elevado rácio de 561/405. As mitocôndrias são organelas que são essenciais para a fisiologia e a proliferação celular. As mitocôndrias são responsáveis pela geração de mais de 80% da fonte de ATP via fosforilação oxidativa, e eles também fornecem vários intermediários metabólicos para biossíntese e metabolismo de1,2. Além de seus papéis no abastecimento de energia e metabolismo, as mitocôndrias desempenham um papel central na muitos outros processos importantes, incluindo a geração de (ROS) de espécies reactivas de oxigénio, a regulação da morte celular e Ca2 + dinâmica intracelular3 . Alterações na função mitocondrial têm sido associadas com doenças humanas, incluindo o câncer, diabetes mellitus e neurodegenerativas várias doenças4,5. Aumento de disfunção mitocondrial e mutações de DNA mitocondriais também são pensadas para contribuir para processos de envelhecimento normal a6,7,8. Portanto, o controle de qualidade é uma questão crucial nas mitocôndrias. As mitocôndrias são organelas altamente dinâmicas que continuamente podem mudar a sua forma entre redes alongadas e formulário curto, fragmentado. Dinâmica de mitocôndrias desempenha um papel importante em manter a função das mitocôndrias, bem como a sua degradação a mitophagy9.
Mitophagy é um mecanismo que envolve a degradação seletiva de mitocôndrias inteira usando a maquinaria de autofagia. Mitophagy é o princípio mecanismo subjacente mitocondrial volume de negócios e a remoção de mitocôndrias danificadas ou disfuncionais. Neste processo, as mitocôndrias são primeiro rodeadas por uma membrana, resultando na formação de autophagosomes, que depois se fundem com lisossomos para degradação hidrolítica9. Estudos genéticos anteriores em Drosophila sugeriram que dois genes relacionados com a doença de Parkinson, induzida por PTEN putativa quinase 1 (PINK1) (PARK6) e Parkin (PARK2), funcionam na mesma via10,11. Subsequence estudos têm demonstrado que a via PINK1-Parkin é responsável por eliminar as mitocôndrias danificadas e defeitos deste resultado de percurso em mitophagy disfuncional e pode contribuir para doenças humanas12,13. Recentemente foram encontrados em várias doenças humanas, incluindo câncer, doenças cardíacas, doenças do fígado e de doenças neurodegenerativas 14defeitos em processos de mitophagy. Além disso, estudos recentes têm mostrado também que mitophagy é fundamental para muitos processos fisiológicos, tais como a diferenciação, desenvolvimento e a resposta imune15,16,17,18 ,19, sugerindo que mitophagy pode desempenhar um papel mais ativo no controle de funções de células.
Apesar da recente confirmação que mitophagy desempenha um papel importante no controle de qualidade tanto em mitocôndrias e doenças humanas, os mecanismos moleculares subjacentes mitophagy permanecem mal compreendido. Embora mecanismos relacionados com mitophagy têm sido sistematicamente estudados em levedura, estudos vistos explorar mecanismos relacionados com mitophagy em células de mamíferos têm focado principalmente via PINK1-Parkin. Estudos anteriores estabeleceram que o pathway PINK1-Parkin é principalmente responsável para a remoção seletiva de mitocôndrias danificadas através de mitophagy12,20,21. No entanto, estudos recentes têm relatado que, em alguns modelos, mitophagy pode ser ativado mesmo na ausência de funcional PINK122,23,24. Estes resultados sugerem que além da via PINK1-Parkin, lá uma adicional via não identificada que pode ativar o mitophagy.
A ausência de um método conveniente para avaliar a atividade de mitophagy é um grande obstáculo para explorar os caminhos que regulam a mitophagy e o seu papel em eventos fisiopatológicos. Microscopia eletrônica é uma poderosa ferramenta para visualizar diretamente as mitocôndrias autophagosome-tragado. No entanto, a microscopia eletrônica tem limitações para quantificar a atividade de mitophagy. Embora as estratégias que usam microtubule-associada da proteína-1 luz corrente 3 (LC3)-conjugadas sondas fluorescentes, tais como a GFP-LC3, são atualmente as abordagens mais amplamente utilizado,25, a natureza transitória do sinal LC3-baseado e sua alta taxa de sinais de falso-positivos limitam sua sensibilidade para a detecção de mitophagy em células26. A combinação da mancha ocidental para medir o nível de proteína mitocondrial, a quantificação do DNA mitocondrial e análise de microscopia de fluorescência para colocalize mitocôndrias com autophagosomes ou lisossomos pode ser uma boa abordagem para a avaliação mitophagy. No entanto, limitações de quantificação e falta de conveniência das metodologias existentes exigem novas abordagens. A introdução de uma nova proteína fluorescente dependente do pH, o alvo mitocondrial Keima (mt-Keima), melhorou muito a capacidade de detectar mitophagy27. Fundindo a sequência mitocondrial-direcionamento de subunidade de citocromo c oxidase VIII (VIII COX) em Keima, mt-Keima é direcionado para a matriz mitocondrial. A grande mudança no pico da excitação de Keima de 440 nm a 586 nm (correspondente a pH 7 e pH 4, respectivamente) pode ser utilizada para avaliar a mitophagy com boa sensibilidade em ambos in vitro e in vivo experimentos28,29 . Mais importante, a resistência de mt-Keima a degradação lisossomal faz com que a integração do sinal do mt-Keima em lisossomos, permitindo a fácil medição quantitativa da atividade mitophagy. A mudança de fluorescência que ocorre no mt-Keima pode ser analisada usando qualquer microscopia confocal ou fluxo cytometry28,29. No entanto, um método baseado em citometria de fluxo para medir a atividade de mitophagy usando mt-Keima não foi apresentado em detalhe a data.
Aqui, descrevemos um protocolo detalhado para uma técnica baseada em citometria de fluxo para medir a atividade de mitophagy nas células usando mt-Keima. Embora nós mostramos aqui que análise de citometria de fluxo detectado com sucesso mitophagy CCCP-induzida em células HeLa, expressando Parkin, esta técnica pode ser aplicada a uma ampla variedade de tipos de células.

sensitive measurement of mitophagy by flow cytometry using the ph-dependent fluorescent reporter mt-keima

Mitophagy é um processo de remoção seletiva de mitocôndrias danificadas ou desnecessárias usando máquinas de autofagia. Foram encontradas relações estreitas entre mitophagy defeituoso e várias doenças humanas, incluindo doenças neurodegenerativas, câncer e doenças metabólicas. Além disso, estudos recentes têm mostrado que mitophagy está envolvido em processos celulares normais, tais como a diferenciação e desenvolvimento. No entanto, o papel preciso da e mecanismos moleculares subjacentes mitophagy exigem um estudo mais aprofundado.
