Lungekræft er stadig langt den største cancer-relaterede dræber i både mænd og kvinder1. Faktisk, ifølge American Cancer Society, hvert år flere mennesker dør af lungekræft end af bryst-, prostata- og tyktarmskræft kræft sammen1. Indtil for nylig, blev størstedelen af patienter med ikke-småcellet lungekræft (NSCLC), som er den mest rigelige undertype af lungekræft, behandlet med platin-baseret kemoterapi i en første-line indstilling, for det meste med tilsætning af angiogenese hæmmere2. Kun en delmængde af patienter havne muterede mutationer i epidermal vækstfaktor receptor (EGFR), i Anaplastisk lymfom kinase (ALK) eller i ROS1, og kan behandles med tilgængelige målretning narkotika3,4. For at studere lungekræft, en bred vifte af prækliniske modeller er tilgængelige, herunder spontan modeller udløses af kemikalier og kræftfremkaldende stoffer og genetisk manipulerede mus modeller (CLAUSS) hvor autokton tumorer opstår efter en betinget aktivering af onkogener og/eller inaktivering af tumor suppressor gener6,7,8. Brug af musen modeller er uundværlig for at studere Patofysiologi af forskellige sygdomme. Med hensyn til lungekræft, flere modeller er til rådighed, herunder genetisk manipuleret modeller samt transplantation modeller. Dog er gensplejsede musemodeller tidskrævende og dyrt, der henviser til, at nogle orthotopic transplantation modeller er svært at reproducere. Her, er en non-invasiv intratrakeal leveringsmetode til lunge tumorceller som en alternativ orthotopic transplantation modellen beskrevet. Brug af musen lunge adenocarcinom celler og syngeneic graft modtagere giver mulighed for at studere tumordannelse under tilstedeværelse af en fuldt aktive immunsystem. Derudover genetiske manipulationer af tumor celler før transplantation gør denne model en attraktiv tidsbesparende tilgang at undersøge effekten af genetiske faktorer på tumorvækst og tumor celle genekspression profiler fysiologiske betingelser. Ved hjælp af denne model, vi vise at lunge adenocarcinom celler programmeret udtrykkelige forhøjede niveauer af T-celle suppressor død-ligand 1 (PD-L1) når der dyrkes i deres naturlige miljø i forhold til dyrkning i vitro.

Den ortopiske transplantation af muselunge adenocarcinom celler i syngeneic graft modtagere tillader undersøgelse af tumorigenesis i overværelse af et fuldt aktivt immunsystem under fysiologiske forhold. Selvfølgelig udvikler tumorerne sig til en immunkompetence mus, og det er meget mere fysiologisk, at hvis du udfører en transplantation i immundefekte mus. Efter at have bekræftet en manglende reaktion på tå knivspids, anvende salve til øjnene af en bedøvet, synergetiske otte til 12 uger gamle mus og krog de øverste fortænder over en sutur streng på tværs af en intubation platform med brystet vinkelret på suturen.Placer en fiberoptisk kabel mellem de forreste lemmer til at belyse brystet og forsigtigt åbne munden ved hjælp af desinficeret flade kraftbefæstet til at udtrække tungen. Kig efter emission af hvidt lys til at finde strubehovedet, epiglottis, og arytenoid brusk. Fjern hurtigt nålen fra kateteret og kontroller, om det hvide lys kan observeres, der skinner gennem kateteret.For at bekræfte en korrekt placering af kateteret i luftrøret fastgøres en 1 milliliter sprøjte vand til kateteret. Vandet i sprøjten skal bevæge sig hurtigt op og ned i tide med vejrtrækningen. Før cellesuspensionen leveres, opvarmes cellerne i hånden, og der indlæsser 50 mikroliter celler i kateteret.Så snart suspensionen er indsugning, fastgør en tom en milliliter sprøjte til kateteret og dispensere 300 mikroliter luft ind i kateteret for at sikre en fuldstændig fordeling af tumorcellerne i lungerne. Fjern derefter forsigtigt kateteret og læg musen på en varmepude med overvågning, indtil den er fuld recumbency. Efter det relevante eksperimentelle endepunkt lægges slagtekroppen i blød i 70 %ethanol, og dyret fastgøres til et dissektionsbræt.Lav en ventrale midterlinjen snit og forsigtigt vende huden for at udsætte thorax væg muskler og abdominale organer. Brug en saks til at punktere mellemgulvet og til at skære ribbenene, udsætter brysthulen. Skær en lille åbning i venstre ventrikel og bruge en 27 gauge nål til at gennemgå lungen tre gange gennem højre hjertekammer med seks til otte milliliter iskold PVS per infusion.Efter den sidste perfusion, lungerne skal være helt fri for blod og synes hvid. Efter høst, bruge en saks til at hakke lungevævet i små stykker. Lungefragmenterne overføres til et mikrocentrifugerør med to milliliter, der indeholder 1,5 milliliter lungefordøjebuffet, i en 30 til 60 minutters inkubation ved 37 grader Celsius med omrystning.Ved afslutningen af fordøjelsen overføres cellesuspensionen gennem en 70 mikrocellesting i et 50 milliliterrør, og enden af et sterilt sprøjtestempl på 10 milliliter anvendes til at trykke eventuelle resterende vævsfragmenter gennem filteret. Skyl si med 15 milliliter PVS, suppleret med 2% FCS. Og indsamle cellerne ved centrifugering.Pellets re-suspension i en milliliter ammoniumchlorid kalium lysis buffer i en fem minutters inkubation ved stuetemperatur og centrifuge cellerne igen. Derefter re-suspendere de hvide blodlegemer pellet i en milliliter PVS suppleret med 2% FCS og plette cellerne for flow cytometrisk analyse i henhold til den eksperimentelle protokol. Som forventet er modtagerens mus's overlevelse korreleret med antallet af indgraferede celler.Og lungerne høstet på dødstidspunktet viser en jævn fordeling af tumor knuder i alle lapperne. Når man sammenligner sektioner fra lungerne huser autokthonous tumorer, med tumorer efter ortopisk transplanterede lungetumorceller, ingen morfologiske forskelle er observeret. Flow cytometrisk analyse af PD-L1 udtryk for lungeceller isoleret tre uger efter ortotopisk tumorcelletransplantation i immunkompetente mus, som netop påvist, afslører opregulering i PD-L1 celle overflade markør udtryk, sammenlignet med in-vitro dyrkede lungetumorceller.Efter ortotopisk transplantation kan der anvendes andre analysemetoder, herunder immunhisemokisk analyse, og mRNA-profilering for at løse yderligere eksperimentelle spørgsmål.
