この手順の全体的な目標は、小動物の外部線放射療法の生体内有効性の評価のためにマウスの幹細胞由来奇形を選択的に標的化することです。外部放射線ビーム療法の主な利点は、全身的に提供される治療法のオフターゲットの悪影響を回避する幹細胞関連テロマの標的療法である点です。外部ビーム放射線療法の進歩により、通常の組織への損傷を避けながら小さな奇形腫を特異的に標的化することが可能となり、放射線感受性奇形腫の治療に最適です。この方法は、細胞療法の最適化戦略に関する洞察を提供するだけでなく、ヒト療法において臨床的に適用可能になる可能性を秘めています。まず、iPSCを含む6ウェルプレートを取ることから始めます。免疫不全動物で四トマ形成を誘導するために、ヒト誘発多能性幹細胞を含む6ウェルプレートのウェル当たり1ミリリットルの組換え細胞解離酵素ミックスを室温で5分間のインキュベーション用の二重融合レポーター遺伝子で導入した。インキュベーションの終わりにピペット処理により細胞を分散させ、各ウェルに等量の細胞培養培地を添加して酵素反応を停止する。15ミリリットルの円錐チューブに単一細胞懸濁液を引き込みます。遠心分離が完了した後、上清を吸引し、マトリゲルマトリックス溶液の30マイクロリットルでペレットを再懸濁し、チューブを氷の上に置きます。二重融合報告細胞を利用する場合、これらの二重陽性影響細胞も30マイクロリットルのマトリックスに懸濁されるべきである。次に、各細胞集団を28.5ゲージ針を備えた1ミリリットルの注射器にロードし、8〜10週齢のアミチンミクスヌードレシピエントマウスの各麻酔でつまみつまみに対する応答の欠如を確認する。次いで、各レシピエント動物の側側腹に1つの細胞マトリックス混合物を皮下に注入する。接種後の適切な実験時点で、各レシピエントに腹腔内に1キログラム当たり375ミリグラムを注入する。標準的な生物発光画像化プロトコルに従って、1分間の取得窓を用いて30分間、各麻酔付きレポータープローブに生物発光シグナルを注入した。テラトーマ照射の場合、適切な実験時点で、まず麻酔付きレシピエント動物を画像誘導前臨床照射器のベッドに置き、40ピークキロ電圧、2ミリアンペアX線ビームを使用して360度以上の400投影微小断層撮影画像を取得し、画像を0.2ミリメートルの異方性ピクセルの体積画像に再構成する。奇形腫の容積画像を再構築することが重要です。これらの画像は、周囲の組織のオフターゲット放射線を最小限に抑えながら、テラトマへの治療の特定の配信のための放射線治療を計画するのに役立ちます。次に、マイクロCT画像とRT画像ソフトウェアパッケージを使用して、放射線治療プロトコルを計画します。3日間連続して治療を行い、合計18個の灰色を標的腫瘍に送達する。注射されたマウスは、典型的には、生物発光イメージングによって確認された腫瘍細胞注射の4週間後に奇形腫増殖形成を実証する。細胞送達の1ヶ月後に18グレイの累積行いで照射すると、腫瘍は劇的に縮小し、ルシファーゼシグナルの有意な一致減少をもたらす。重要なことに、これらの画像が示すように、照射された部位から5ミリリットルを収穫した正常組織生検は、重大な損傷を持続するようには見えない。このビデオを見た後、あなたは生体内で幹細胞由来の奇形を作成し、効果的かつ安全に外部ビーム放射でそれらを扱う方法を明確に理解する必要があります。この簡単なアプローチでは、隣接する組織を避けながら、一連の放射線ビーム療法を処方して標的腫瘍を照射することができる被験者の高解像度CT画像を取得する必要があります。放射線を扱うことは非常に危険であり、適切なシールドを使用するなどの予防措置は、この手順を実行する際に必ず取られるべきであることを忘れないでください。
