人工多能性幹細胞またはIPS細胞から生成された心筋細胞は、心臓電気生理学の分野における様々な生物学的問題に答えるためにますます使用されています。この技術の主な利点は、心室特異的プロモーター要素を使用して、筋細胞のような心室におけるスループットの劇的な増加とサブタイプ特異的なアクションポテンシャル記録を可能にすることです。レンチウイルス伝達を通じて遺伝的にコード化された電圧センサーの導入により、ヒト由来の多能性幹細胞由来心筋細胞の作用電位の光学イメージングが可能になる。レンチウイルス含有細胞培養上清と心筋細胞維持培地を1対1の割合で混合することから始める。ヘキサジメテリン臭化物を1ミリリットル当たり8マイクログラムの最終濃度で添加し、感染効率を高める。IPS細胞由来心筋細胞培養皿中の心筋細胞維持培地を、感染培地と交換する。感染した心筋細胞を摂氏37度で12時間培養し、感染培地を新鮮な心筋細胞維持培地に置き換える。イメージングの前に、細胞培養培地をチローデの溶液と交換し、カルシウムを補う。次に、画像スプリッターを搭載した反転性蛍光顕微鏡のステージ上の画像チャンバーに心筋細胞を配置します。電気ペーシングが必要な場合は、撮像チャンバーにペーシング電極を取り付け、電極を刺激発電機に接続します。細胞に焦点を合わせ、視野の中央に遺伝的にコードされた電圧インジケータを表すセルを見つけます。光が放射された光がイメージスプリッターにルーティングされるように光路を設定し、顕微鏡内のフィルターキューブを画像スプリッタ内のフィルターと組み合わせるものに切り替えます。カメラを制御するソフトウェアでは、高速イメージングが可能になるように取得設定を行い、メーカーの指示に従って画像スプリッタを設定します。2 つの異なる発光波長バンドを表す 2 つの画像は、それぞれ画像の半分を占める互いに隣接している必要があります。カメラで生成された画像の焦点を確認し、必要に応じて調整します。また、ピクセルの彩度が避けないように、照明ライトの明るさを調整します。時系列の取得を設定し、測定に影響を与える迷い光を避けるために室内の光を暗くします。次に励起光シャッターを開き、画像シリーズの取得を開始し、適当な時点で電気ペーシングを開始し、実験的に適宜行う。取得が終了したら、必要に応じて励起ライトシャッターを閉じ、記録をハードドライブに保存します。次に、記録設定を変更せずに、セルを含まないカバースリップの領域の背景を記録します。光学膜電位記録の解析のために、ImageJ内の細胞を含むティフスタックを開き、自由手選択ツールを使用して蛍光心筋細胞の上に関心領域を描き、赤色チャネルで、収縮中に細胞が領域外に移動しないように領域が十分に大きいことに注意する。次に、[分析]、[ツール]、[関心のある地域マネージャー] を選択し、[T の追加] を選択して、リージョンを [対象地域 1] として追加します。緑色のチャネル内の同じセル上に対象領域をドラッグし、この領域を [対象地域 2] として追加します。[分析]メニューと[計測値を設定]メニューで、[平均]グレーの値を除くすべてのオプションの選択を解除します。[対象のリージョン マネージャー] で、[その他のメジャーとマルチ メジャー] をクリックして[すべてのスライスを測定] オプションと [スライスごとに 1 行] オプションを選択し、[OK] をクリックします。3 行のスプレッドシートを含むウィンドウが開きます。[ファイル] と [名前を付けて保存] を使用して、スプレッドシートをカンマ区切り値ファイルに保存します。[対象の領域] マネージャ ウィンドウを閉じずに、イメージ スタックとスプレッドシート ウィンドウのあるウィンドウを閉じ、背景の計測値を含む tiff スタックを開きます。[対象のリージョン マネージャー] で、[その他のメジャーとマルチ メジャー] をクリックして[すべてのスライスを測定] オプションと [スライスごとに 1 行] オプションを選択し、[OK] をクリックします。次に、バックグラウンド データを別の csv ファイルに保存します。ここで、代表的な単一IPS細胞由来心筋細胞細胞は、対象領域を示す白い点線で描かれ、RFPおよびGFPチャネルにおける画像解析中に描かれた図が示されている。RFPチャネルからの信号は、膜電位の変化によって生じる強制または共鳴エネルギー伝達の増加による各作用電位の間の蛍光強度の周期的増加を示す。一致して、GFPシグナルは、蛍光強度の周期的な減少を示す。RFP/GFP比は、アクション電位期間50、90測定など、下流解析で使用される生物学的信号です。例えば、この方法を用いて、0.5ヘルツに配置されたIPS細胞由来心筋細胞のような30日齢の心室は、平均作用電位持続時間50の439ミリ秒、および平均作用電位持続時間520ミリ秒の平均作用電位持続時間90を示した。IPS細胞由来心筋細胞の電気刺激は、5拍ごとに刺激速度を増加させると、拍動速度が増加するにつれて作用電位の進行的な短縮を示した。さらに、イソプロテルノールを用いた自発的に打ち負かすIPS細胞由来心筋細胞の治療は、非選択的βアドレナリン受容体アゴニストであり、鼓動速度の迅速な増加につながり、アゴニストの用量の増加に伴ってさらに増加することができる。この方法は、古典的な電気生理学的測定と比較して優れたスループットを提供するが、蛍光電圧センサの固有の運動特性によって制限される時間分解能を含む、その限界を認識する必要があります。
