De verbindingen van de BSA-Au(III) bereid met een pH van 12 vertonen rode fluorescentie bij een golflengte van de emissie van λem= 640 nm wanneer opgewonden met ultraviolet (UV) licht λex= 365 nm (figuur 1A). De opkomst van rode fluorescentie is een langzaam proces en duurt een paar dagen bij kamertemperatuur te verhogen tot een maximale intensiteit. Deze verbinding is uitgebreid bestudeerd door vele onderzoekers op het gebied van nano-wetenschap in de afgelopen jaren14,15,16. De BSA-Au-compound is geïnterpreteerd als Au25 nanoclusters. Het doel van de onderhavige methode is deze compound in detail onderzoeken en begrijpen van de oorsprong van de rode fluorescentie. Door het volgen van de gepresenteerde aanpak, kunnen de aanwezigheid van meerdere Au bandplaatsen, en de oorsprong van de fluorescentie, alternatief voor de nucleatie van één site van Au25 nanoclusters, worden geïllustreerd. Dezelfde aanpak kan worden gebruikt om te bestuderen hoe andere eiwitten17,18,19 complexvorm met Au(III) hun intrinsieke fluorescerende eigenschappen kunt wijzigen.
De synthese van het rood-fluorescerende BSA-Au samengestelde vereist een smalle controle van de molaire verhoudingen van BSA aan Au (BSA:Au) om de intensiteit van de fluorescentie en de locatie van de pieken in de excitatie-emissie kaart (EEM)20te maximaliseren. Kan worden aangetoond dat er meerdere bandplaatsen bestaan voor Au(III) te binden, met inbegrip van de Asparagine fragment (of ASP-fragment, de eerste vier aminozuurresidu's op de N-terminus van BSA)21,22. De 34th aminozuur van BSA (Cys-34) blijkt ook te coördineren van Au(III) en te worden betrokken in het mechanisme van de rode fluorescence([Cys34-capped-BSA]-Au(III))20. Op het splijten van alle Cys-Cys disulfide bindingen en aftopping alle thiolen, rode fluorescentie is niet geproduceerd ([all-thiol-capped-BSA]-Au(III)). Dit wijst op de noodzaak van Cys-Cys bisulfide obligaties als de Au(III) binding site voor de productie van de rode fluorescentie.
Eiwit chemie technieken hebben niet wijd gebruikt om te studeren het BSA-Au(III) complexen in de nano-wetenschappelijke gemeenschap. Het doel van de voorgestelde protocollen is te bestuderen van het proces van Au(III) binding aan BSA, opbrengst conformatie verandering-geïnduceerde rode fluorescentie (λem = 640 nm) van BSA-Au(III) complexen. De methode past de pH om te laten zien dat de opkomst van de rode fluorescentie is gecorreleerd met de pH-geïnduceerde evenwicht overgangen van de BSA conformaties.
