重组病毒的传递

</ol>6. 重组 hvt 的核查

间接免疫荧光法
	<ol>
<li>在感染前一天用 2.5 x 10 cef 播种的24井板中, 用步骤5.2 中获得的第二代重组 hvt 感染 cef。在感染后48小时内取出细胞培养培养基, 在 pbs 中加入500μl 的4% 聚甲醛来固定细胞。在室温下将板材加氢 30分钟, 然后取出固定物, 用 pbs 清洗细胞层3x。 注: 在此步骤中, 电池可能会在4°c 下存储数周。</li>
		<li>取出 pbs, 加入 500μl 0.1% triton x-100, 使细胞渗透 15分钟. 用 pbs 清洗细胞层3x。</li>

</ol>7. 重组病毒的稳定性
<ol><li>种子 2.6 x 106 6 cef细胞进入每个 t25 瓶的前一天, 病毒扩张。</li>
	<li>从步骤5.2 中解冻至少三个阳性克隆;在每个 t25 瓶中加入一小瓶细胞病毒。在38.5°c 下, 用5% 的 co2 进行培养 , 直到观察到50% 的细胞病理效应。</li>
	<li>在培养基2毫升中采集细胞;感染50μl 到一个新的 t25 烧瓶预置 cef 细胞的下一代。持续传递重组病毒至少15代。通过 pcr 分析每一代病毒是否存在 vp2 序列, 并通过 vp2 表达的间接免疫荧光分析 (ifa) 来评估重组病毒的稳定性。</li>

generating recombinant avian herpesvirus vectors with crispr/cas9 gene editing
