この方法は、がん遺伝子依存に関与する主要な遺伝子の同定など、がん治療薬分野の主要な質問に答えるのに役立ちます。この技術の主な利点は、多くのキナーゼ駆動癌に共通する腫瘍遺伝子依存性を利用するため、複数の腫瘍タイプに適用可能である。この方法は白血病に関する洞察を提供することができますが、肺癌、脳腫瘍、膵臓腫瘍などの固形腫瘍のような他のシステムにも適用できます。一般的に、この方法に新しい個人は、分子生物学や細胞生理学の経験の欠如のために苦労します。まず、安楽死させたマウスから脚の骨を収穫します。メスで骨から組織をきれいにします。その後、氷の上に冷たいPBSに骨を置き、害虫で骨を粉砕します。この後、70マイクロメートルのセルストレーナーを通して、10ミリリットルのピペットを用いた50ミリリットルのスクリューキャップチューブにセルサスペンションをフィルターします。乳鉢と害虫を冷たいPBSで複数回洗い、50ミリリットルチューブに細胞懸濁液を加えます。次に骨髄を遠心分離し、真空とガラスピペットで上清を取り除きます。細胞ペレットを5ミリリットルのPBSで再懸濁する。ピペットを使用して、中断した骨髄を室温ポリショ糖の上部にゆっくりと加えます。この後、ポリショ糖を400 Gsで20分間回転させてブレーキをオフにします。遠心分離後、ピペットで曇った全単核細胞界面を回収し、新しい15ミリリットルチューブに移します。PBSで10ミリリットルにボリュームを持参し、カウントのために20マイクロリットルを取り除く。遠心後、上清を捨てて、ペレットを氷の上に置きます。次に、EDTAおよびBSAでPBS中の細胞ペレットを再懸濁する。CD117マイクロビーズを細胞懸濁液に加え、渦を混ぜ合わせます。この後、4°Cで10分間、懸濁液をインキュベートします。その後、EDTAとBSAと遠心分離機で1200 Gで5分間、10ミリリットルまでのボリュームを持って来ます。真空を使用して上清を除去し、EDTAおよびBSAでPBS中の細胞ペレットを一時停止する。次に、EDTAとBSAを持つPBSを磁気カラム付きのマグネットスタンドに3ミリリットル加え、回収管に流れ込みます。バッファーが列を通過し終わったら、セルの懸濁液を追加し、列を通ってコレクションチューブに流れるようにします。次に、EDTAとBSAを持つPBSをさらに5ミリリットル追加し、プランジャーですぐにフラッシュします。プランジャーを取り外し、セルを数える前にフラッシュを繰り返します。細胞を遠心分離した後、上清を取り除き、培養のために完全なIMDMで細胞ペレットを一時停止する。細胞を摂氏37度で一晩培養し、5%の二酸化炭素を使用します。まず、レトロウイルスプラスミドDNAを1.5ミリリットルチューブに包装プラスミド、塩化カルシウム、水と組み合わせます。その後、HBSの500マイクロリットルを別の1.5ミリリットルのチューブに加えます。最も低い設定に設定されたボルテクサーと同時に混合しながら、HBSを含むチューブにトランスフェクション混合物をドロップワイズ追加します。トランスフェクション混合物を室温で20~30分間インキュベートする。インキュベーション期間中、HEK細胞にクロロキンを加え、インキュベーターに保管します。トランスフェクションミックスを加えた後、新鮮なDMEM10培地を14ミリリットル加えて細胞培地を変化させる摂氏3%CO2インキュベーターで8時間培養します。皿の壁に対してゆっくりとピペットを入れ、HEK細胞の剥がしを防ぐのに役立ちます。その後、摂氏37度5%CO2インキュベーターで細胞を一晩インキュベートします。ウイルス上清を収集するために10ミリリットルの注射器を使用してください。その後、0.45マイクロメートルのシリンジフィルターをシリンジに取り付けます。ウイルス上清を新しいコレクションチューブに突っ込みます。約16時間後にウイルス上清を収集します。この後、PBSに2ミリリットルの組換えヒトフィブロネクチン断片を未治療6ウェル培養プレートに添加する。翌日、ピペットを使用してプレートからフィブロネクチンを取り除きます。PBSで2%BSAでプレートをブロックし、室温で30分間インキュベートします。真空を使用してBSAを取り除き、PBSでプレートを洗ってから真空で取り除きます。この後、すぐに濾過されたウイルス上清を加え、プレートを2時間遠心分離する。真空を使用して上清を取り除き、プレートを再び回転させます。次に真空で上清を取り除き、PBSでウイルスコーティングされた井戸を洗浄します。真空でPBSを取り出し、以前培養したc-Kipとマウス細胞をウイルスコーティングされたウェルに加える。遠心分離後、セルを摂氏37度と5%の二酸化炭素でインキュベートします。導入から48時間後、遠心分離を介して細胞をペレット化し、FACS緩衝液1で再懸濁した。まず、マウスのメチルセルロースをサイトカインで室温で解凍します。次いでFACSチューブ中のメチルセルロースの3ミリリットルをアリコートした。メチルセルロースの3ミリリットルに適切な阻害剤を用いて細胞懸濁液の100マイクロリットルを加える。その後、サスペンションを10〜30秒間高く渦液します。気泡のほとんどが逃げた後、1ミリリットルのシリンジを使用して、3つの複製プレートに1ミリリットルの培地をプレートします。皿を大きなペトリ皿に入れ、滅菌水を入れた1つのオープンディッシュと一緒に入れます。その後、大きなペトリ皿をカバーし、37°Cで細胞をインキュベートします。10ミリリットルの水に10ミリグラムの塩化ヨドニトロテトラゾリウムを溶解して、ヨドニトロテトラゾリウムクロリドステインを調製します。フィルターは0.2マイクロメートルフィルターが装備されているLuerロックシリンジと染色液を殺菌する。組織培養フードに、CFUプレートの周りに滴下して100マイクロリットルの染色液を加える。その後、細胞培養インキュベーターで摂氏37度で一晩プレートをインキュベートします。最後に、コロニーが濃い赤色の茶色に変わった後、染色されたCFUプレートの写真を撮るために、無菌組織培養フードに白い背景を使用する。このプロトコルでは、複数の公平な遺伝子発現分析を使用して、遺伝子中毒を調整する遺伝的構成要素を同定した。白血病における治療標的としてのc-FosおよびDusp1の役割を検証するために、BCR-ABL1オンコジーンを発現する細胞でそれらの過剰発現が確認された。RT-qPCRおよびウェスタンブロッティングによる分析は、両方ともBCR-ABL1によって誘導され、その発現が成長因子IL-3によって増強されることを確認した。YFPプラス発現細胞は、さらにインビトロおよびインビボアッセイのためにFACSを介して選別した。結果は、c-FosおよびDusp1の削除のみによってCFU数が阻害されることを示した。c-FosとDusp1の両方を欠いている細胞は、コロニーを形成する能力が著しく損なわれ、イマチニブ治療はC-FosおよびDusp1の喪失がBCR-ABL1発現に対して合成致死的であることを示すCFUsのすべてを一掃した。この開発の後、この技術は、がんの分野の研究者が、遺伝子の依存を促進する遺伝的ネットワークと遺伝子を探求し、これらの遺伝子がマウスモデルおよびヒト患者における特定の応答にどのような影響を与えるかを探求する道を開いた。レトロウイルスや患者のサンプルを使用すると、この手順を実行している間に非常に危険で安全な実験室の練習を観察することができることを忘れないでください。
