De hier gepresenteerde fixatie procedure voor knaagdieren longen is niet nieuw; Dit systeem heeft echter verschillende voordelen. COPD is een van de leidende wereldwijde oorzaken van overlijden1. Sigarettenrook is de belangrijkste oorzaak van COPD, maar de mechanismen van pathogenese blijven niet volledig gedefinieerd. COPD toont twee hoofdkenmerken, waaronder progressieve beperking van de luchtstroom en een abnormale ontstekingsreactie van de longen. Emfysemateuze stoornis treedt vaak op in de longen van COPD-patiënten2. De pathologische bevindingen van emfyseem worden gekenmerkt door alveolaire wand vernietiging3. Er zijn verschillende diersoorten gebruikt voor het genereren van COPD-modellen in vivo (d.w.z. honden, cavia's, apen en knaagdieren)4. Echter, de muis is uitgegroeid tot de meest gebruikte in de bouw van COPD modellen. Dit heeft vele voordelen, met inbegrip van de lage kosten, het vermogen om genetisch gemodificeerd, uitgebreide genoominformatie beschikbaarheid, beschikbaarheid van antilichamen, en het vermogen om een verscheidenheid van muis stammen gebruiken5. Momenteel is er geen muismodel dat de volledige kenmerken van menselijke COPD kan nabootsen; individuele onderzoekers moeten dus kiezen welk model het meest geschikt is voor het specifieke COPD-onderzoek6. De emfysemateuze muismodel is een van de vele COPD Muismodellen die momenteel beschikbaar zijn. Aanvullende modellen omvatten de exacerbatie muismodel, systemische co-morbidities model, en COPD susceptibiliteit model7.
Het emfysemateuze muismodel kan worden gegenereerd door verschillende soorten exogene agentia, waaronder chemische agentia en blootstelling aan sigarettenrook4. Chemische blootstelling (bv. aan elastase) produceert een ernstig type emfyseem, terwijl sigarettenrook resulteert in milde emfyseem8,9. Sigarettenrook wordt verondersteld de belangrijkste oorzaak te zijn voor de pathogenese van COPD; Daarom is de keuze van sigarettenrook als middel om een COPD-muismodel te creëren redelijk10. Veel studies hebben sigarettenrook gebruikt om emfyseem in de muis te maken. Zo heeft Nikula et al. met succes een emysemateuze muismodel gemaakt van B6C3F1 vrouwelijke muizen door ze bloot te stellen aan sigarettenrook gedurende 7 of 13 maanden11. We hebben ook een emysemateuze muismodel ingesteld via senescentie marker proteïne/SMP-30 KO muizen12. Het is cruciaal om een Long fixatie methode uit te voeren die dit milde emfyseem-model door blootstelling aan sigarettenrook goed kan visualiseren.
Er zijn verschillende methoden voor Long fixatie vastgesteld13. Er is echter geen gouden standaardmethode voor longweefsel fixatie voor het evalueren van emfyseem14. Verschillende studies van dit Lab hebben aangetoond dat het hier gepresenteerde fixatiesysteem nuttig is door een stabiele conditie te creëren voor het evalueren van emfyseem12,15,16,17,18. Het belangrijkste voordeel van het huidige systeem is dat het veel longen met dezelfde aandoening in één keer kan repareren zonder longcollaps of deflatie. Het huidige Long fixatiesysteem gebruikt een aantal speciale apparatuur waarmee Long specimens gedurende een bepaalde periode kunnen worden opgeblazen bij een geschikte constante druk. Deze speciale uitrusting bestaat uit drie delen, waaronder een onderste container, bovenste container en pomp. Long specimens worden in de onderste container geplaatst die is verbonden met bevestigingsmiddelen onder druk, wat resulteert in een 25 cmH2O drukverschil in het niveau van de agentia tussen de bovenste en onderste containers19.

Emfyseem is een belangrijk kenmerk van chronische obstructieve longziekte (COPD). Studies met een emfysemateuze muismodel vereisen een optimale Long fixatie om betrouwbare histologische specimens van de longen te produceren. Vanwege de aard van de structurele samenstelling van de longen, die grotendeels uit lucht en weefsel bestaat, bestaat het risico dat het tijdens het fixatie proces instuilt of leegmaakt. Er bestaan verschillende Long fixatie methodes, die elk zijn eigen voor-en nadelen hebben. De Long fixatie methode die hier wordt gepresenteerd, maakt gebruik van constante druk om optimale weefsel evaluatie mogelijk te maken voor studies met een emfysemateuze muis Long-model. Het belangrijkste voordeel is dat het in één keer veel longen met dezelfde aandoening kan oplossen. Long specimens worden verkregen uit chronische sigarettenrook blootgestelde muizen. Long fixatie wordt uitgevoerd met behulp van gespecialiseerde apparatuur die de productie van constante druk mogelijk maakt. Deze constante druk handhaaft de longen in een redelijk opgeblazen toestand. Zo genereert deze methode een histologisch monster van de longen dat geschikt is om door sigarettenrook veroorzaakte milde emfyseem te evalueren.

Zoals eerder beschreven, kunnen de gespecialiseerde apparatuur, die een uitgebreide constante druk genereert, worden onderverdeeld in drie delen (Figuur 3A). De constante druk in het fixatieapparaat behoudt de longen in een redelijk opgeblazen toestand, waardoor een histologisch longexemplaar ontstaat dat geschikt is voor de evaluatie van milde sigarettenrook geïnduceerde emfyseem. Het belangrijkste voordeel van dit model is dat het veel longen tegelijkertijd kan repareren met dezelfde constante druk zonder instorting of deflatie van de longen. Het aantonen van de procedure zal Naoko Arano een afgestudeerde student van mijn laboratorium.Na bevestiging van een gebrek aan reactie op reflexbeweging maak een incisie in de huid en spierweefsel langs de mediale lijn van de muis, gericht op de kopvormige regio, en lateraal gesneden om een bredere werkruimte te creëren. Na het blootstellen van de middenriflaag doorboren het weefsel met tangen en open de thoracale ruimte. Snijd het achterstevengebied waardoor de longen en het hart kunnen worden gevisualiseerd.En snijd het linker atrium en de rechter hartkamer. Steek een 24-meter canule aangesloten op een perfusiepomp in het rechter ventrikelgebied en richt de canule naar het cephalic gebied totdat het de longslagader bereikt. Zet vervolgens de pomp aan en doorsnijd het weefsel met ongeveer 200 milliliter PBS gedurende een uur totdat al het longweefsel is veranderd in een witte kleur.Aan het einde van de perfusie snijd het bindweefsel rond de luchtpijp, longen en hart, en gebruik een hechting af te binden van de juiste belangrijkste bronchus. Oogst vervolgens alle individuele lobben van de rechterlong en plaats het hart en de kwabben van de linkerlong in individuele 10 milliliter spuiten met een geschikte fixatief. Trek de zuiger in waardoor een vacuüm in de spuit ontstaat om de longen op te blazen en de long uit de spuit te verwijderen.Steek een canule van 20 meter in de luchtpijp en gebruik een hechting om de canule op zijn plaats te zetten. Sluit een een milliliter spuit gevuld met fixatief op de canule en vul de long met extra fixatief om te controleren op lekken. Zorg ervoor dat de invoegplaats is beveiligd om te voorkomen dat het longmonster tijdens de fixatie loskomt van het longbord.Breng het weefsel vervolgens over naar de drukapparatuur voor longfixatie, verwijder het monster en bindt de luchtpijp aan het einde van de fixatieperiode met een knoop. De onderste component van de longfixatiedrukapparatuur waarin het longmonster wordt ingebracht, wordt via 20 meter cannula verbonden met het puntje van de formaline stroom met een driewegstopcock. Druk wordt gegenereerd door de verschillende oppervlakteniveaus van het fixatief tussen de onderste en bovenste compartimenten.Het drukverschil is 25 centimeter water, maar de hoogteverstellingsknop kan worden gebruikt om de druk aan te passen aan 25 tot 30 centimeter water. Een pomp verbindt de onderste en bovenste containers, het behoud van een 25 centimeter verschil in de fixatieve oppervlakte hoogte, en stuurt de stroom van het middel. Na 48 uur fixatie vertonen de longen van SMP30 knock-out muizen die zijn blootgesteld aan omgevingslucht geen duidelijke luchtruimuitbreiding.Significante luchtruim uitbreiding en alveololerende muur vernietiging, echter, worden waargenomen in SMP30 knock-out muizen blootgesteld aan chronische sigarettenrook. Bovendien zijn de gemiddelde lineaire onderschepping en destructieve index van de longmonsters aanzienlijk groter in de rook blootgestelde SMP30 knock-out muizen dan in de lucht blootgestelde dieren. Ons lab maakt gebruik van 10% formaline, maar onderzoekers kunnen gebruik maken van andere fixatie middelen op basis van hun experimentele behoeften.Zoals we hebben gemeld met behulp van de aangetoonde fixatie systeem longmonsters kunnen worden geoogst uit verschillende muis emfyseem modellen voor hun morfometrische evaluatie. Aangezien bevestigingsmiddelen gevaarlijk kunnen zijn, moet u altijd de juiste beschermingsmiddelen voor het personeel dragen en in een goed geventileerde ruimte werken.
