Depuis Salmonella spp /Shigella spp. sont souvent associés aux intoxications alimentaires et transmission fécale-orale de la gastro-entérite aiguë28,29 , et la méthode de routine est beaucoup de travail ou de longue durée 7, nous décrivons une plate-forme haut débit pour la Salmonella spp / le dépistageShigella spp., à l’aide de la PCR en temps réel combinée avec la culture guidée.

La diarrhée est toujours un problème de santé courant à forte incidence taux globalement1,2. Bien que la mortalité est relativement faible, certains patients présentent des symptômes divers pendant des semaines (par exemple, molles et aqueuses tabourets, une urgence à aller aux toilettes), qui rendent l’impact socioéconomique très élevé3,4. Plus sérieusement, certains patients peuvent même développer le syndrome du côlon irritable si laissé non traité5. Il existe différents types de bactéries, les virus et les parasites qui peuvent causer la diarrhée6. Salmonella spp /Shigella spp sont parmi les bactéries les plus courantes pour la transmission de la gastro-entérite aiguë7,8,9,10,11. Par conséquent, beaucoup de comtés ont émis des lois ou aux règlements réguliers Salmonella spp /Shigella spp. dépistage parmi les gens qui traiteront de nourriture et eau. Par exemple, le gouvernement chinois ont publié des lois pour obligatoire Salmonella spp /Shigella spp. dépistage une fois par an.
La méthode de l’étalon-or pour Salmonella spp / détection deShigella spp est la culture de bactéries. Par le biais de la culture de bactéries et identification biochimique successive et caractérisation sérologique, nous pouvons identifier les espèces de bactéries, ce qui pourraient faciliter la gestion des épidémies maladie et profilage antimicrobienne pour faciliter le traitement des patients 12. il pourrait aussi aider à remonter à la source de l’infection au cours de la Salmonella spp /Shigella spp éclosion13. Toutefois, cette méthode est fastidieuse (nécessitant une opération manuelle) et beaucoup de temps (prendre plusieurs jours), en particulier pour les essais d’un grand nombre d’échantillons7. En outre, viable mais non cultivable (VBNC) Salmonella spp /Shigella spppeut exister dans certains échantillons de selles14. Compte tenu de ces inconvénients, plusieurs laboratoires ont tenté de développer de nouvelles techniques pour la détection de Salmonella spp /Shigella spp.15,16,17,18 , 19 , 20 , 21 , 22 , 23 , 24 , 25. tous ces méthodes utilisent le test nucléaire acid amplification (TAAN), parmi lesquels la réaction en chaîne par polymérase (PCR) est le plus fréquent. Une limitation majeure de ces méthodes TAAN basé est que les bactéries mortes, ADN génomique incomplet contenant des débris même bactérienne, pourraient montrer des résultats positifs26, qui peuvent largement influencer le diagnostic précis de maladie. Blanco et coll. ont montré que test moléculaire est très sensible, non seulement viable Salmonella dans les cultures, mais aussi aux génomes partielles et bactéries infections passées ou contamination26morts ou non viables. Donc, les nouvelles technologies devraient être développés.
Ici, nous avons décrit une nouvelle méthode que combine le TAAN base de méthode et culture. Tel qu’illustré à la Figure 1, cette nouvelle méthode s’applique de PCR en temps réel contrôle premier et ensuite les échantillons positifs sont envoyés pour l’identification et de la culture de bactéries.

La transmission fécale-orale de gastro-entérite aiguë se produit de temps en temps, surtout quand les personnes qui manipulent les aliments et l’eau sont infectées par Salmonella spp/Shigella spp. La méthode de l’étalon-or pour la détection de Salmonella spp/Shigella spp est culture directe mais c’est long et fastidieux. Nous décrivons ici une plate-forme haut débit pour Salmonella spp/Shigella spp de dépistage, à l’aide de la PCR en temps réel (PCR) combiné avec la culture guidée. Il y a deux grandes étapes : PCR en temps réel et la culture guidée. Pour la première étape (PCR temps réel), nous expliquer chaque étape de la méthode : échantillons de collection, pré-enrichissement, extraction d’ADN et la PCR en temps réel. Si le résultat de la PCR en temps réel est positif, la deuxième étape (culture guidée) est réalisée : la culture sélective, identification biochimique et caractérisation sérologique. Nous illustrons aussi résultats représentatifs générés à partir d’elle. Le protocole décrit ici serait une plate-forme utile pour le dépistage rapide, précise, sensible et haut débit de Salmonella spp /Shigella spp.

Salmonella spp /Shigella spp sont pathogènes courants attribués à la diarrhée. Ici, nous décrivons une plate-forme haut débit pour la détection de Salmonella spp /Shigella spp., à l’aide de la PCR en temps réel combinée avec la culture guidée.

Cette méthode fournit une plate-forme à haut débit pour le dépistage de Salmonella et Shigella. Le principal avantage de cette technique est la répétition, spécifique, sensible, et le débit élevé. Cette technique combine la méthode et la culture rapides naat dans lesquelles le criblage en temps réel de PCR a été appliqué d’abord, et puis positifs ont été envoyés pour la culture et l’identification de bactéries.Bien que cette méthode se concentre sur le dépistage de Salmonella et de Shigella, elle peut également être appliquée à d’autres agents pathogènes tels que Vibrios, Staphylococcus, E.Coli, et cetera. La démonstration visuelle de cette méthode est critique car le choix de la colonie correcte est difficile en raison de la flore de fond dans les échantillons. Pour commencer, ajouter trois millilitres de bouillon nutritif à chaque échantillon.Ensuite, incubez les échantillons à 36 degrés Celsius pendant six heures. Après cela, centrifuger la culture de pré-enrichissement à 800 gs pendant deux minutes. Ensuite, transférez le supernatant dans un nouveau tube, et centrifugez-le à nouveau pendant cinq minutes à 12 000 gs.Après avoir jeté le supernatant, ajouter 100 microlitres de solution d’extraction d’ADN à la pastille. Ensuite, vortex le tube pendant une minute. Ensuite, chauffez l’échantillon à 1000 degrés Celsius dans le bain sec.Après avoir répété la centrifugation, recueillir le supernatant dans un nouveau tube. Tout d’abord, créer le mélange de réaction et définir le programme de cyclisme selon le protocole de texte. Ensuite, effectuez un PCR en temps réel sur une machine PCR fluorescente en temps réel.Tout d’abord, ajouter 100 microlitres de la culture de pré-enrichissement à cinq millilitres de cristal de sélénite moyenne. Puis, incuber la culture à 36 degrés Celsius pendant 18 à 24 heures. Ensuite, recueillir une boucle de la culture et l’étendre sur une plaque.Ensuite, incuber la plaque à 36 degrés Celsius pendant 18 à 24 heures. Après l’incubation, sélectionnez les colonies suspectes et transférez-les dans une assiette. Incuber la plaque à 36 degrés Celsius pendant 18 à 24 heures.Ensuite, identifiez la colonie suspecte à l’aide d’un système automatisé d’identification microbienne. Pour commencer la caractérisation des antigènes, ajoutez une goutte de l’O-antigène polyvalent sera à une glissade propre. Transférez une boucle de la colonie à la glissière et broyez-la dans le sera.Si les bactéries ressemblent à du sable qui coule, répétez le traitement précédent avec du sérail monovalent o-antigène jusqu’à ce que l’antigène soit caractérisé. Pour commencer la caractérisation de l’antigène H, ajoutez une goutte d’antigène H polyvalent sera à une glissade propre. Ensuite, recueillir une boucle de la colonie et la griller dans le sera.Utilisez h-antigène monovalent sera pour répéter le traitement jusqu’à ce que l’antigène H spécifique est caractérisé. Pour séparer la culture, recueillir une boucle de la culture pré-enrichissement et la répandre sur une plaque. Puis, incuber l’endroit à 36 degrés Celsius pendant 18 à 24 heures.Après l’incubation, recueillir une colonie suspecte à la recherche sur une plaque. Incuber la plaque à 36 degrés Celsius pendant 17 à 24 heures. Ensuite, soumettez la colonie à l’identification biochimique à l’aide d’un système automatisé d’identification microbienne.Ensuite, appliquez une goutte d’espèce Shigella polyvalent sera sur une glissade propre. Ensuite, utilisez une micro-boucle pour recueillir certaines des bactéries et les moudre dans le sera. Enfin, si le sera ressemble à du sable qui coule, utilisez shigella espèces monovalent sera pour caractériser davantage les bactéries.À l’aide de ce protocole, des échantillons de selles prélevés sur des patients ont fait l’objet d’un dépistage des espèces de Salmonella et de Shigella. À l’aide du PCR en temps réel, il y a eu amplification réussie dans le canal HEX, ce qui indique que l’échantillon était positif pour les espèces de Salmonella. D’autres PCR en temps réel ont indiqué que l’échantillon deux était positif pour Salmonella et qu’il avait été choisi pour la culture guidée des espèces de Salmonella.Dans la culture guidée des espèces de Salmonella, les colonies roses et violettes ont été plaquées séparément et soumises à l’identification biochimique. Les résultats ont indiqué que les colonies de l’échantillon deux étaient Salmonella paratyphi B.En utilisant le PCR en temps réel, il y a eu amplification réussie dans le canal FAM, ce qui indique que l’échantillon était positif pour les espèces de Shigella. D’autres PCR en temps réel ont indiqué que l’échantillon 10 était positif pour Shigella et a été choisi pour la culture guidée.Dans la culture guidée des espèces de Shigella, les colonies rose-rouge et incolore ont été plaquées séparément et soumises à l’identification biochimique. Les résultats ont indiqué que l’échantillon 10 contenait des bactéries Shigella sonnei phase II. Tout en essayant cette procédure, il est important de se rappeler la limitation de la méthode due à la variation de séquence.Suite à cette procédure, d’autres méthodes, comme la culture directe, peuvent être effectuées afin d’éviter de faux résultats négatifs en temps réel pcr. Cette technique ouvre la voie aux chercheurs dans le domaine de la détection de Salmonella et shigella, car le nouveau protocole pourrait augmenter le taux positif de deux fois et réduire considérablement la charge de travail et le TAT. N’oubliez pas que travailler avec Salmonella et Shigella peut être extrêmement dangereux, et l’équipement de protection individuelle, PPE, doit toujours être pris lors de l’exécution de cette procédure.
